Single-strand breaks and repair in nuclear DNA in the presence of hydralazine assayed by the nucleoid technique.

The nucleoid sedimentation assay was used to study hydralazine-induced DNA structural changes and repair in the fibroblasts cultured in vitro. The drug induced a dose dependent loss in negative DNA supercoiling due to the physical breakage of the DNA. Relaxation of supercoiled DNA resulted in the nucleoids sedimenting with lower velocities than those of undamaged control cultures. Repair incubation of the cells did not cause the restoration of DNA supercoiling to control level. Unsuccessful repair of DNA damaged by hydralazine may result in maintaining the damaged DNA in the cell which could have immunologic consequences.

[Causes of peri-operative mortality in patients with esophageal cancer treated surgically (personal material)].

99 surgical procedures of thoracic part of esophagus resections were performed between 1985 and 1995. All thoracotomies were right-sided and alimentary canal continuity were substituted with a stomach (or with a small intestine or a colon in only a few cases) transposed inside a thorax cavity. Almost all cases there were planoepitheliale cancers. The sex index (men to women) was 6:1. The main reason of perioperative mortality were pulmonary complications (almost 90%): a respiratory insufficiency, an atelectasias, infections, and a broncho-pleural leakage. The nutrition state, stapler-using, anastomosis leak, and wound-healing way had no influence on mortality rate. Other mortality reasons there were a pleural hemorrhage, a mediastinitis as a result of esophago-gastric anastomosis disruption, a heart infarct, and a thrombo-embolic complications.

[The role of Shouldice's operation in treating inguinal hernia].

Clinical estimation of Shouldice procedure in the surgical treatment of inguinal hernia was performed in the group of 95 patients (102 operations) treated in the Second Department of General and Vascular Surgery Clinic at Zabrze between 1994 and 1996. Their ages ranged from 18 to 78 years with an average 53.6. The sex index (men to women) was 11:1. The bilateral hernia was found in seven cases and all of them were treated surgically at the same time. The most treated hernias was considered as type II and V by Gilbert classification. Among treated patients there were six cases of recurrence hernia earlier operated according to other procedures (mainly Bassini, Girard, Halsted, and artificial knitted-graft placed up intraperitoneally). In 28 cases of fascia transversalis large deficiency (the third and fourth type of Gilbert classification) authors used the two-layer modification method of reconstruction instead four layer of hernial canal posterior wall reconstruction the typical for Shouldice procedure. It was sufficient in all cases without respect to hernia size and fascia transversalis deficiency. There were no complications in postoperative periods. The recurrence of hernia, appeared only in one patient (0.98%) nine months after operation. In authors opinion, the Shouldice procedure is the most effective among all classical ways of inquinal hernia repairs.

Plasma selenium concentration in patients with stomach and colon cancer in the Upper Silesia.

Plasma selenium concentration was assessed in 44 patients with cancer of the gastrointestinal tract (19 subjects with stomach cancer and 25 with colon cancer) and 25 age-matched healthy control subjects. Selenium concentration was determined by the fluorometric method. The observed plasma selenium concentrations in gastrointestinal cancer patients (37.0 +/- 11.05 ng Se/ml or 38.4 +/- 12.6 ng Se/ml in stomach or colon cancer patients, respectively) were significantly lower as compared to the healthy age-matched control group (51.4 +/- 14.4 ng Se/ml). The diagnosed low selenium status may be considered as a high risk for cancer development.

Comparison of the action of transforming growth factor-beta 1 and interleukin-1 beta on matrix metabolism in the culture of porcine endothelial cells.

The purpose of the experiment was to evaluate the metabolism of the connective tissue (collagen and proteoglycans) in the endothelial cell culture (enzymatically isolated from porcine aortas). The endothelial cells were activated by 1-10 ng/ml TGF-beta 1 and 250 pg/ml IL-1 beta together and separately. Intensity of proteoglycan synthesis has been evaluated by using radioactive [35S]-sodium sulfate whereas the intensity of collagen was evaluated by incorporation of [3H]-proline. The following conclusions have been drawn as the result of the experiment: 1) TGF-beta 1 stimulates the synthesis of proteoglycans and collagen in endothelial cell culture, whereas IL-1 beta has the opposite action; and 2) TGF-beta 1 reduces IL-1 beta action which hampers the synthesis of proteoglycans and collagen, provided that the culture's preincubation with TGF-beta 1 precedes the incubation with IL-1 beta.

Increased lipid peroxidation as a mechanism of methionine-induced atherosclerosis in rabbits.

Methionine is converted by the transmethylation/transsulfuration pathway to homocysteine which may exert atherogenic effects by several mechanisms, including lipid peroxidation. Therefore, the excessive dietary methionine may induce the development of atherosclerosis. To test this hypothesis, plasma and aortic thiobarbituric acid reactive substances (TBARS), as well as activities of aortic and erythrocyte superoxide dismutase (SOD), catalase and selenium-dependent glutathione peroxidase (GPX) were measured in rabbits fed a diet enriched with 0.3% methionine for 6 or 9 months. Histological examinations of aortas also were performed. Feeding rabbits a methionine-enriched diet for 6 or 9 months resulted in significant increases in plasma and aortic TBARS levels and aortic antioxidant enzyme activities. However, a decrease in plasma antioxidant activity (AOA) was observed. In erythrocytes, SOD activity increased, catalase remained normal and GPX decreased in the treated animals. Histological examination of aortas showed typical atherosclerotic changes, such as intimal thickening, deposition of cholesterol, and calcification in methionine-fed rabbits. These results confirm that high-methionine diet may induce atherosclerosis in rabbits and indicate disturbances in lipid peroxidation and antioxidant processes as possible mechanisms of its atherogenic influence.

[A study of erythrocyte oxidative enzyme activity in patients with laryngeal cancer before and after treatment with a selenium preparation CEROSEL].

The preliminary results of activity changes of antioxidant enzymes (glutathione peroxidase, catalase and superoxide dismutase) in erythrocyte in 19 patients with laryngeal cancer were presented. Enzymes activity in patients was signified three times: before surgery, before and after radiotherapy. During Co-60-therapy patients received leaven preparation of selenium CE-ROSEL in dose 100 micrograms Se/day. Obtained results showed increase of enzymes activity after CE-ROSEL supplementation. This changes are probably the result of selenium influence on protected mechanism before the free radicals.

[About the matter of injury to the superior laryngeal nerve during strumectomy].

The attention is paid to the frequently overlooked in clinical practice problem of diagnosis and strumectomy, in the light of literature data on peripheral nerve regeneration. The authors point to important causes of frequent failures in the treatment of such complications. They conclude that at the present level of knowledge, the only practical way to avoid the aforementioned complication is the change of surgical technique in routine strumectomy.

Modulation of procainamide toxicity by selenium-enriched yeast in rats.

Free radical processes are proposed to play a crucial role in the development of procainamide adverse effects. Therefore, selenium, as a potent antioxidant, may modified procainamide toxicity. To test this hypothesis plasma and liver thiobarbituric acid-reacting substances (TBARS), plasma antioxidant activity (AOA), erythrocyte and liver superoxide dismutase (SOD), catalase, as well as selenium-dependent glutathione peroxidase (Se-GPX) were determined in the following four groups of rats: selenium-treated (Se), procainamide-treated (P), procainamide and selenium-treated (P + Se), and control (C). Morphological studies of leukocytes [tested for lupus erythematosus (LE) cells] and liver were also made. Atypical, i.e. enlarged and swollen, leukocytes resulting from procainamide and selenium treatment were observed. These changes were found in four out of five rats in the Se group, eight out of ten in the P group, and in seven out of ten in the P + Se group. LE-like cells were observed in two rats in the P + Se group. A statistically significant decrease in plasma and liver TBARS by 20% and 36%, respectively, increased activity of SOD by 20%, catalase by 48% and Se-GPX by 15% in erythrocytes, and decreased activity of liver SOD by 17% and catalase by 22% were found in the P + Se group as compared to the P group. These results indicated that selenium exerted antioxidant effects on the procainamide-treated rats. However, selenium did not prevent the development of disturbances in leukocyte morphology, on the contrary, it possibly promoted the conversion of leukocytes to LE cells.

Increase in antioxidant activity in procainamide-treated rats.

Recent studies suggest that in vivo procainamide oxidation underlies induction of autoimmunity by this drug. Since drug metabolism may be accompanied by generation of reactive oxygen species, plasma and liver thiobarbituric acid reacting substances (TBARS), activity of erythrocyte and liver superoxide dismutase, catalase, selenium-dependent glutathione peroxidase (Se-GPX), and plasma antioxidant activity in procainamide treated rats were evaluated. Procainamide administration increased liver lipid peroxide levels, intensified the activity of liver catalase and erythrocyte superoxide dismutase, as well as plasma antioxidant activity. The remaining biochemical parameters in the treated rats were within control values, except for the decreased erythrocyte catalase activity. We conclude, that the increased activity of free radicals observed in the treated rats could contribute to the development of procainamide induced side effects.

Elastin metabolism parameters in sera of patients with lung cancer.

Elastin metabolism parameters (elastin-derived peptides and elastase-like activity) were determined in sera of patients with lung cancer and in healthy controls. The concentration of elastin-derived peptides was statistically significantly elevated in the lung cancer group. There was no statistically significant difference in the serum elastase-like activity between the groups studied. These data seem to indicate an enhanced metabolism of elastin in patients with lung cancer.

Silicon metabolism. The interrelations of inorganic silicon (Si) with systemic iron (Fe), Zinc (Zn), and copper (Cu) pools in the rat.

The influence of silicon treatment on the levels of trace elements zinc (Zn), copper (Cu), and iron (Fe) in serum and tissues was studied in rats. The concentrations of silicon, iron, and zinc were estimated in samples of sera and tissues of rats receiving per os a soluble, inorganic silicon compound--sodium metasilicate nonahydrate (Na2SiO3.9H2O), dissolved in the drinking water. An increase of copper concentrations in liver and aortic walls in the experimental group was observed, with simultaneous reduction of zinc amounts in serum and all the tissue samples in the course of the experiment. The iron concentrations in the analyzed samples did not show any significant changes between both groups. The silicon levels in serum and in all the examined tissues were significantly higher in the tested group. The results provide evidence for the silicon interaction with copper and zinc, which could result in a number of metabolic process modifications, antiatheromatous activity among them.
