Spontaneous subminature end-plate potentials in mouse diaphragm muscle: evidence for synchronous release.

1. Miniature end-plate potentials (min.e.p.p.s) were recorded from small muscle cells of mouse diaphragms. Min.e.p.p. amplitude histograms showed successive peaks which were integral multiples of the smallest peak. The smallest potentials (submin.e.p.p.s) averaged 0-3-0-6mV and the mean of the larger min.e.p.p.s averaged 3-7 mV, depending on the muscle cell diameter. There was a positive correlation between time-to-peak and min.e.p.p. amplitude. Time-to-peak of the submin.e.p.p.s fell slightly below the regression line through the larger min.e.p.p.s. 2. Sometimes min.e.p.p. amplitude distributions changed spontaneously such that the mean of the major mode min.e.p.p.s decreased twofold during which time the mean of the submin.e.p.p.s did not change. Spontaneous decreases were most pronounced during low frequencies of release (10/min) achieved at 32 degrees C. 3. Small changes in temperature (2 degrees C steps in the range 32-40 degrees C) greatly altered the number of peaks of min.e.p.p. amplitude histograms without noticeably changing the position of the submin.e.p.p. peak. At 32 degrees C submin.e.p.p.s composed 5-20% of the histograms and the amplitude of the major mode peak was twelve to fifteen-times that of the submin.e.p.p.s. Over-all bell-shaped distributions were obtained at 37 degrees C which showed up to eight peaks with the major peak at the fourth to sixth peak. Temperatures slightly above 37 degrees C gave a flat distribution with the mean amplitude at the third peak. Min.e.p.p. amplitude histograms were initially skewed (mostly small min.e.p.p.s) after a 40 degrees C heat challenge. 4. Two to eight-times the normal concentration of Ca2+ in the saline reversibly increased the min.e.p.p. frequency and also decreased the mean of the major mode min.e.p.p.s (two to nine-times) without noticeably changing the mean of the submin.e.p.p.s. 5. Botulinum toxin A, 10(5) X intraperitoneal median lethal dose (10(5)I.P.LD50)/ml., almost abolished min.e.p.p.s in 30-90 min. The relative proportion of submin.e.p.p.s increased and the mean of the major mode min.e.p.p.s usually did not change during the initial decrease in frequency. Major mode min.e.p.p.s essentially ceased after 200-1000 were generated and remaining min.e.p.p.s of some cells showed skewed distributions with three small peaks that were integral multiples of the submin.e.p.p. peak. Smaller min.e.p.p.s were more resistant to block than the larger min.e.p.p.s and, although frequencies were low, small min.e.p.p.s were recorded after 4 hr of botulinum toxin incubation. 6. Colchicine (5 X 10(-4)M) within minutes reduced the major mode min.e.p.p.s by half (mean of major peak reduced to sixth or seventh peak). Additional colchicine (10(-3)M reduced the major mode min.e.p.p. amplitude to a fifth of that of control (mean of major mode min.e.p.p.s at the third peak) with no change in position of the submin.e.p.p. peak. Min.e.p.p. amplitudes slowly recovered to half control values after washing. 7...

Non-Poisson distribution in time of small- and large-mode miniature end-plate potentials.

Minature end-plate potentials (MEPP's) were recorded from frog (Rana pipiens) sartorius muscle cells of 60--80 mum diam; MEPP frequency remained constant up to 3 h and was 100 times greater in 80-mum cells than in 6-mum cells. Rapid cooling or heating of preparations initiated or accentuated bursts of MEPP's. Records from cells under control conditions and from cells where the MEPP population had been reduced to the small mode showed many instances of small clusters of MEPP's occurring within 100-ms intervals, indicating quantal release interdependencies. Some small cells initially generated many large MEPP's 2 or more times the average amplitude and fewer small clusters. Analysis of the occurrence of both large- and small-mode MEPP's for both 1.0-s and 100-ms nonoverlapping bins demonstrated a highly significant deviation from a Poisson distribution with significance increasing with decreasing cell diameter and/or MEPP frequency. It is suggested that doublet MEPP's are generated from one release site and that large bursts results from the activity of several release sites, since the size of bursts decreases with cell diameter (junctional area). It is concluded that release characteristics are the same for large-mode (classical) and small-mode MEPP's and that the occurrence of MEPP's is not Poisson.

Multimodal distribution of frog miniature endplate potentials in adult denervated and tadpole leg muscle.

Amplitude histograms of spontaneous miniature endplate potentials (MEPPs) from adult sartorius muscle cells show a definite bimodality with the mean amplitude of the larger mode five to seven times that of the smaller mode which accounted for 2-5 % of the total MEPPs. Histograms were plotted after high frequency MEPP generation induced by increasing temperature, increasing external calcium or nerve stimulation. These plots showed a reversible left-shift of the major mode as well as a reversible increase in the proportion of small mode MEPPs. Repeated challenges shifted almost all MEPPs into the small mode. An increase in the percentage of small mode MEPPs also occurred spontaneously during the course of denervation before the quiescent period and some of the histogram profiles showed multiple modes whose means were integer multiples of the small mode mean. In the early stages of hind leg development the greatest proportion of MEPPs were of the small mode size; as metamorphosis progressed, the histograms showed a definite multimodality with the mean of each mode being an integer multiple of the small mode mean and with the proportion of MEPPs in each mode about the same. During tail resorption the percentage of larger MEPPs increased until the adult histogram profile was reached. Thus, the changes in MEPP amplitude histograms over the course of metamorphosis are the reverse of those found with denervation.

Oculomotor neurons in fish: electrotonic coupling and multiple sites of impulse initiation.

Oculomotor neurons are electrotonically coupled in three teleosts. Electron microscopy revealed axosomatic synapses with close appositions of pre- and postynaptic membranes. Similar junctions are associated with electrotonic coupling in many other cases. Stimulation of the ipsilateral eighth nerve usually initiated impulses at sites distant from the cell bodies; stimulation of the ipsilateral ophthalmic nerve initiated impulses close to the cell bodies. Electrotonic coupling may synchronize impulses arising near the, cell bodies to generate synchronous muscle contractions. Impulses arising distant from the cell bodies may lead to contractions of graded strength.

Electrical characteristics of tunicate heart cell membranes and nexuses.

The tubular ascidian heart is composed of a single layer of cells joined together by apical (zonulae occludentes) and spot (maculae occludentes) nexuses. Intercalated discs or desmosomes were not observed in this tissue. Rectangular pulses of current were applied across the opened and flattened myocardium. Assuming that all the transepithelial current flowed through a uniform gap between cells, the resistivity in the gap must be very high compared to that in bulk solution. It is likely, therefore, that the gap width is of the order of an ionic radius or smaller. Assuming that all the transepithelial current flowed through the cells and that the inner and outer membranes had the same resistivity, the membrane resistivity was about 210 ohms cm(2) and the membrane capacitance was about 1.6 microF per cm(2). The myocardial cells were found to be in electrical continuity with each other through the nexuses since current could be passed through a strip of myocardium in a sucrose gap. Assuming that the longitudinal resistance of the cytoplasm was negligible, the cell-to-cell resistivity of the nexuses was 0.2 ohm cm(2). It is concluded that the nexuses provide a low resistance pathway between cells and a transepithelial barrier.

Conduction velocity and intracellular action potentials of the tunicate heart.

The tubular heart of tunicates is composed of a single layer of myoendothelial cells. The direction of contraction reverses every few minutes. The conduction times in both directions are the same. Conduction velocity was greatest in the middle of the arms of the V-shaped heart and slowest in the apex. The greater the heart length, the greater was the conduction velocity. The Q(10) of conduction velocity was 2-2.3. Removal of the raphe which attaches the heart to the pericardium and removal of a line of undifferentiated cells opposite the raphe did not change the conduction velocity or prevent the heart from reversing the direction of conduction. The median resting potential of 42 cells was -71 mv and the median action potential was 75 mv. At 20 degrees C the duration of the action potential was 1.2 sec and the maximal rate of depolarization was 3-10 v/sec. An increase in the beat frequency produced by electrically stimulating the heart decreased the resting potential, rate of rise, the duration, and the overshoot of the action potential. The shape of the action potential was sometimes different in the two directions of conduction. The electrophysiological evidence indicates only one cell type and suggests that the mode of the spread of excitation is by local current flow from cell to cell.

Transmitter release: prepackaging and random mechanism or dynamic and deterministic process.

Stepwise variations in end-plate potential amplitudes that are also multiples of spontaneous miniature end-plate potentials (MEPPs) demonstrate a quantal nature of evoked transmitter release at the vertebrate neuromuscular junction. Both the number of quanta which form relatively small end-plate potentials (EPPs) and the time intervals between MEPPs were found to fit Poisson statistics. These observations suggested that the release process randomly liberates uniform quantities of transmitter. Initial studies showed that quantal size remained stable after seemingly high rates of release which was interpreted to indicate that a large store of equally sized, equally available, and independently releasable quanta are present in the nerve terminals. The observation of numerous presynaptic vesicles that contain transmitter provided a morphological basis for prepacked transmitter (i.e., quanta). However, physiological studies over the last 15 years have yielded data that are difficult to incorporate into the quantum-vesicle hypothesis. With normal conditions and during most treatments which increase the rate of release, two classes of MEPPs have been found and both show a substructure. The bell-MEPP class was characterized by Fatt and Katz and the smaller skew-MEPP class has been studied by Kriebel. The ratio of the two classes and substructure compositions of both classes are variable. Short series of MEPPs and unitary EPPs (U-EPPs) show preferred amplitudes and longer series of MEPPs and U-EPPs show stepwise variations in amplitude. Slow-MEPPs and giant MEPPs belong to the skew class and represent nearly synchronous bursts of smaller MEPPs. Transmitter packet formation, preferred amplitudes, stepwise variations in amplitudes, random-like distributions and organized bursts can be simulated by a simple deterministic system, the drop formation process, that is known for its periodic and chaotic behaviors which are determined by the single parameter of flow rate. MEPP intervals, sizes and classes, are also dependent on rates of release which demonstrate that the release process(es) is highly organized and sensitive to different conditions. We demonstrate that the processes of drop formation and release of a packet of transmitter have similar properties and that deterministic characteristics describe MEPP and U-EPP time dependencies and amplitude substructures. The data and model presented here suggest that packet size of acetylcholine may be determined at the moment of release.
