Cartilage-derived anti-tumor factor (CATF) inhibits the proliferation of endothelial cells in culture.

Previous studies showed that a cartilage-derived anti-tumor factor (CATF), which was extracted from bovine cartilage with 1 M guanidine hydrochloride, inhibited the growth of solid sarcoma 180, but not the growth of ascites tumors of sarcoma 180 [Suzuki, F. et al. Jpn. J. Bone Metab., 2, 231-235 (1984)]. This study showed that CATF inhibited the proliferation and DNA synthesis of endothelial cells isolated from bovine pulmonary artery, but not the proliferations of sarcoma 180, L1210 and L cells. These results suggest that CATF has anti-angiogenic properties, thereby inhibiting the growth of solid tumors.

Papillary cystadenoma.

A rare case of a papillary cystadenoma, mucous-cell type, in the lower lip of a 73-year-old male is presented. The tumor consisted of many small cysts with multiple papillary projections into their cystic spaces. The projections and cyst walls were lined with pseudostratified mucous-cell epithelium. Clinically and histologically, the present papillary cystadenoma was considered as a true tumor originated from the ducts of minor salivary glands. There exists only one case report of a papillary cystadenoma with adequate description and photomicrographs in the Japanese dental literature.

Induction of spermidine/spermine N1-acetyltransferase by parathyroid hormone in rabbit costal chondrocytes in culture.

Parathyroid hormone (PTH) increased the activity of spermidine/spermine N1-acetyltransferase, a rate-limiting enzyme of polyamine biodegradation, in rabbit costal chondrocytes in culture. The enzyme activity increased in a dose-dependent manner after addition of PTH to the culture, reaching a maximum at 8 h. The increase in the enzyme activity was abolished by cycloheximide or actinomycin D. Dibutyryl cyclic AMP also induced the acetyltransferase to some extent. These results suggest that the induction of spermidine/spermine N1-acetyltransferase by PTH may play some significant role in the expression of the differentiated phenotype of chondrocytes.

How does Japan differentiate hyperplasia from neoplasia?

A Japanese pathologist's contribution to the discussion of the problem on differentiation of hyperplasia from neoplasia is to introduce his experience and knowledge in human and experimental pathology of gastric, hepatic, and uterine cervical cancers, all of which are prevalent in Japan. Canine and rodent gastric cancers induced experimentally by N-ethyl-N'-nitro-N-nitrosoguanidine or N-methyl-N'-nitro-N-nitrosoguanidine, respectively, show different histologic types which are similar to human gastric cancer when examined routinely by endoscopic method. Dogs show more similarities to human gastric cancer than rats in the morphologic features and responses to chemotherapy. Serial liver biopsies performed on patients with liver diseases revealed the final stages of liver cell carcinoma in some of them. They all progressed to liver cirrhosis before terminating in carcinoma. However, this does not mean that the hyperplastic nodule is an obligatory precursor of carcinoma in human. Among experimental models of liver cancer produced by a large number of agents, only carbon tetrachloride and luteoskyrin seem to induce liver cell carcinoma combined with cirrhotic lesions in rodents. The mode of manifestation of atypical changes in the proliferating cells as preneoplastic or neoplastic lesions seems to differ according to tissue. The cellular pathology of cervicovaginal smears is a reliable index for detection of carcinoma in the cervix, where the appearance of atypical cells represents a landmark between benign and malignant tumors.

Parathyroid morphology in rats after administration of active vitamin D3.

The morphology of the parathyroid gland was examined in rats treated for one month with an active vitamin D3, 1 alpha-hydroxyvitamin D3. On continuous administration of 12.5 micrograms/kg/day of 1 alpha-hydroxyvitamin D3, the first histological change of the parathyroid gland, seen on day 10, was atrophy of the chief cells with marked accumulation of prosecretory granules. Replacement of the parenchyma by small or large cysts was evident on days 20 and 30. The remaining portion of the parathyroid parenchyma showed various histological changes: widened intercellular spaces intermingled with many cytoplasmic processes, shrinkage of the cytoplasm of the chief cells, and the presence of a few ghost cells in cysts. The appearance of cysts may be caused by suppression of parathyroid hormone secretion and is a characteristic lesion in hypervitaminosis in rats induced by treatment with active vitamin D3.

Bacteriophage chi sensitivity and motility of Escherichia coli K-12 and Salmonella typhimurium Fla- mutants possessing the hook structure.

The production of hook protein and flagellin in 29 Fla- mutants of Escherichia coli K-12 was determined by the complement fixation assay. Six mutants produced hook protein, and four of them also produced flagellin. A flaE mutation was introduced into these fla mutants carrying the hook structure. All of these mutants made polyhooks and were used as hosts for a newly isolated host-range mutant of chi phage that has a high affinity for the hook structure. All except one mutant produced significant amounts of progeny phages. A flaD flaE double mutant was that exception which did not yield significant amounts of progeny by the phage propagation method. All of the flaE double mutants produced comparable amounts of polyhooks, and no qualitative difference was detected between chi-sensitive and chi-insensitive mutants by the complement fixation assay. Accordingly, it was thought that the polyhook of the flaD flaE mutant had a mechanical defect for chi phage infection. This assumption was confirmed by tethered-cell experiments; the flaD flaE mutant did not rotate. These results are well explained by a proposed regulation pathway of flagellar genes. flaE mutants can express other genes which govern the final step of the flagellar morphogenesis, whereas flaD mutants cannot rotate, possibly because the mocha operon is not expressed. The results obtained in E. coli were also found to be applicable to Salmonella typhimurium.

[Report of a family with cancer family syndrome].

A report of multiple primary early duodenal cancer and ileocecal cancer with cancer family syndrome is presented. Analysis of 65 individuals of this family showed 7 large bowel cancers, 1 lung cancer and cancer of the uterine cervix. The characteristics of this family including increased occurrence of adenocarcinoma, especially large bowel cancer, 3 multiple primary malignant neoplasms, early age of onset of cancer and suspicion of autosomal dominant inheritance, satisfy the criteria for the cancer family syndrome. We recommend screening the individuals in this family for early signs of cancer.

Mapping of the pin locus coding for a site-specific recombinase that causes flagellar-phase variation in Escherichia coli K-12.

Although the vh2 mutation almost entirely prevents phase variation in Salmonella spp., an Escherichia coli strain that carried the Salmonella H1 and H2 region, including the vh2 mutation, showed phase variation. From this strain, EJ1076, a number of mutants defective in phase variation were isolated, and the symbol pin was assigned to their mutant gene. The pin locus was mapped between purB and trp near purB by interrupted matings using Tn10 sites inserted near pin. The locus was cotransduced with purB by P1 vir at a frequency of around 0.33. All the mutations tested were clustered at this locus. Three E. coli K-12 strains probably derived via different lines from the wild type have been tested for the presence of pin+ by introducing the two Salmonella H regions; two were pin+, and one was a pin mutant.

Time course of biochemical and histological alterations following a single feeding of carbon tetrachloride to mice.

Carbon tetrachloride (0.02, 0.5 or 1.5 ml/kg) was given through a stomach tube to female C57BL/6J mice. Liver weights, the quantities of nine liver (protein, free glucose, glycogen, triglyceride, phospholipid, free and esterified cholesterol, DNA and RNA) and three blood components (glucose and free and esterified cholesterol), and SGOT activities were measured at 15, 24, 39, 48, 63, 87, 159 or 327 hr after carbon tetrachloride feeding. Livers were examined histologically. Chemical components of the liver and blood changed generally in a dose-related manner, but more slowly in the larger-dose groups. The biochemical alterations were reflected well in the histological findings. Degenerative and regenerative findings occurred later in the larger-dose animals. Histological changes appeared first in the midzones in the 0.5 and 1.5 ml/kg groups.

Enteric coated polymyxin B in the treatment of hyperammonemia and endotoxemia in liver cirrhosis.

Effects of enteric coated polymyxin B capsules on hyperammonemia and endotoxemia in liver cirrhosis were investigated. Six million units of polymyxin B were orally administered daily to 21 patients with liver cirrhosis and 3 patients with hepatoma cum liver cirrhosis, whose plasma ammonia was higher than normal limit and/or whose plasma endotoxin was positive, for 5-32 days, and serum polymyxin B concentration (in 5 cases), changes of plasma ammonia level (in 19 cases) and plasma endotoxin (in all cases) were observed. Serum polymyxin B concentration was below the detectable limit (0.5 unit/ml) in all cases observed. In the patients with liver cirrhosis, plasma endotoxin and ammonia levels decreased rapidly after polymyxin B treatment, and the decreases in endotoxin levels were kept throughout the treatment. Twelve patients with liver cirrhosis (10 among them were treated with lactulose) were served as controls. All patients who were treated with lactulose alone showed rapid decrease in plasma ammonia, but the decrease in endotoxin in these patients was slower than that in those treated with polymyxin B. From these results, oral administration of polymyxin B is concluded to be useful in the treatment of hyperammonemia and endotoxemia in liver cirrhosis, as a poorly absorbed antibiotic and as an antiendotoxin agent.

High incidence of angiosarcomas in brown-fat tissue and livers of mice fed sterigmatocystin.

BDF1 female mice fed 30 or 120 ppm sterigmatocystin in their diet for 55 or 51 wk, respectively, developed angiosarcomas of the liver and of the dorsal brown-fat tissue. No tumours were observed in five mice from each group, including the controls, killed after 43 wk, but angiosarcomas were observed in mice from the test groups that died or were killed after more than 43 wk of treatment with sterigmatocystin. Thirty-four out of 53 mice (64.2%) fed 30 ppm sterigmatocystin developed hepatic angiosarcomas. Six (11.3%) of those fed 30 ppm sterigmatocystin and 27 out of the 51 mice (52.9%) fed 120 ppm developed angiosarcomas in the dorsal brown-fat tissue. Benign vascular lesions in the liver and a few angiosarcomas of the ovary and lung were also observed in mice of both test groups. No vascular changes were observed in control mice except for peliosis-like lesions in the spleens of two mice. Increases in the incidences of lung and hepatocellular adenomas were also observed in sterigmatocystin-treated mice.
