[Effect of excess D,L-tryptophan on the level of aminoacylation of tRNA by 14C-tryptophan].

The effect of the D,L-tryptophan excess in the rat organism was studied as applied to the level of amino acylation of tRNA by 14C-tryptophan. It is established that the introduction of tryptophan for three days in doses of 50 and 75 mg per 100 g of the living weight evokes a 24 and 40% decrease, respectively, in the level of amino acylation of the rat livel tRNA by 14C-tryptophan. The ability of accepting 14C-tryptophan in total preparations of the experimental rats liver tRNA considerably lower as compared to norm. A short-term heating in the presence of magnesium ions partially restores their decreased acceptor ability. This gives ground to suppose that the molecules of tRNAtr in the total preparation of the rat liver tRNA with a great tryptophan excess in the organism partially change their conformation and this is one of the reasons of the decrease in the level of tRNA amino acylation by 14C-tryptophan. The decrease in the protein synthesis at the first stage with the presence of a great tryptophan excess in the organism is evident to be also connected with the inhibition of tryptophanyl-tRNA-synthetase activity.

[Changes in the primary structure of rabbit muscle aldolase under the influence of valine against a background of prolonged fasting].

The primary structure of the muscle aldolase molecule was studied as affected by semilethal doses of valine administered the abdominal cavity of the rabbits after a long fasting. It is established that in spite of differences in the amino acid composition of the protein, uniformity of the peptides distribution in the process of bromo-cyanogen fragments elution and the total amount of amino acid residues in the identical fragments are maintained. Changes are found only in the point-replacements by amino acids in C-fragment of the molecule (asparagine is replaced by valine and threonine by serine).

[Biosynthesis of rat skin collagen under amino acid imbalance].

Protein differing from the normal one by the content of S-S-bonds and by the amount of the carbohydrate component was found in the acid-soluble fraction of skin collagen in rats which were fed the protein-free diet and the protein-free diet with the glycine loading for 3 and 14 days and tryptophan for 1, 2 and 3 days. The result testifies to the fact that under extremal states in the animal skin there occurs the synthesis of collagen the structure of which differs from that in the normal animal skin.

[Features of metabolic disorders in the liver during morphine poisoning of various duration].

Administration of morphine into rats at a dose of 30 mg/kg within 6 days led to a decrease in total rate of tRNA aminoacylation in liver tissue. Content of lactate, pyruvate, malate and alpha-ketoglutarate was decreased within 6 days-long course of morphine administration, while content of lactate was only altered after 5 weeks of the intoxication. Adaptation reactions appear to be increased with time in long-term intoxication with morphine.

[Change in the intensity of biosynthesis of proteins, lipids and glycogen in animal tissues during long-term administration of morphine].

The intensity of biosynthesis processes in animal organism has been studied as affected by long-term administration of morphine. It was established that morphine administration to rats for five weeks intensified protein biosynthesis in the brain, kidneys, skeletal muscles: specific radioactivity of blood serum proteins also increased. Incorporation of 2-/14C/glycine label to the brain, cardiac and skeletal muscles increased as affected by morphine: the label incorporation to the liver lipids decreased and that to the kidney and spleen lipids did not change. Specific radioactivity of glycogen multiply increased in the rat liver as affected by morphine.

[Structure and properties of collagen in leukemia].

Collagen I was isolated from human bone tissue and from mice bone tissue of the AKR-50 strain at the pronounced stage of leukosis. Dissimilarity of native and leukemic collagens was exhibited after evaluation of their amino acid composition, electrophoretic mobility in polyacrylamide gel containing SDS, content of the carbohydrate moiety as well as of isopoints, elution profiles in reverse-phase chromatography and gel filtration and electron microscopy of SLS-crystallites. Impairments of collagen processing in leukemia appear to be responsible for its alterations in structure and properties.

[Effect of carbostimulin and its mixture with vitamin D3 on the mineralization process of the bone callus in rabbits].

Mineralization of rabbit bone callus was studied within 15 days after resection of the upper part of radius bone inder conditions of treatment with carbostimuline and its mixture with vitamin D3. Content of calcium in the bone regenerate was increased by 40.8% after administration of carbostimuline and--by 70.5% after treatment with mixture containing vitamin D3. These preparations caused only slight effects on the phosphorus content. Dry residue in the regenerate os f animals, treated with the preparations, was increased approximately 2-fold (43% and 23%, respectively, p less than or equal to 0.001) as compared with control group. Content of citric acid was distinctly higher in the bone tissue of the treated animals. Within 15 days after the operation content of sialic acids wanormalized in blood serum of the animals. The data obtained were corroborated by X-ray and histologic examinations.

[Correction with formate of metabolic disorders in alcoholic intoxication].

Formate was studied for its effect on the content of acetaldehyde, activity of the total aldehyde dehydrogenase, content of substrates of glycolysis and tricarbonic-cycle and pool of free amino acids of rat tissues during alcohol intoxication. The introduction of formate during the acute alcohol intoxication lowers the acetaldehyde content in the blood; the ethanol load being prolonged--it increases the activity of aldehyde dehydrogenase and normalizes the content of pyruvate, glutamate and malate in the liver and glutamate and oxaloacetate in the brain, that evidences for the correction of metabolic disturbances in the organism.

[Reaction of glycine with acetoacetate and various properties of the catalyzing enzyme from the rat liver].

The enzymic interaction between acetoacetate and glycine which are added outside is established to occur in the tissue homogenates of the liver, kidneys and spleen of rats. The interaction leads to a decrease of the both components in the equivalent quantities. Homogeneous crystalline preparations of the enzyme catalyzing the above mentioned reaction are isolated from the liver tissues and purified. Certain properties of the enzyme are studied.

Acetoacetates

[Various metabolic reactions of formate in animal tissues].

A new metabolic reaction of the aldolase condensation between formic acid and acetaldehyde proceeding with the formation of milk acid is detected in the liver of rats. Milk acid has been determined by chemical, enzymic and autoradiographic methods. Homogeneous preparations of the enzyme which catalyzes the mentioned reactions and is called lactate synthase are obtained in the crystalline form. The method for obtaining the lactate synthase from the rat liver is described as well as certain properties of the lactate synthase.

Acetaldehyde

[Effect of different levels of carbonic acid on activity of dehydrogenases].

A dependence is established for the activity of NADP-dependent isocytrate dehydrogenase and malate dehydrogenase as well as of NAD-malate dehydrogenase on the carbonic acid level. When the biocarbonate concentration in rabbit blood and tissues rise, the activity of cytoplasmic NADP-isocytrate dehydrogenase and NADP-malate dehydrogenase of the liver lowers. The analogous regularity is observed in the in vitro experiments when the bicarbonate concentrations in the medium rises up to 40 mM (within the physiological limits). The activity of NAD-malate dehydrogenase in the soluble fraction of the rabbit liver and kidney increases with the bicarbonate level within the same limits. The found dependence was confirmed in experiments with the enzyme crystalline preparations.

[Effect of different concentrations of HCO3- and CO2 in aquarium water on metabolism in the Cyprinus carpio L. organism].

The level of pCO2 and bicarbonates in blood and tissues of Cyprinus carpio L. increases with concentration of carbonic acid (HCO3- and pCO2), the other indices of the hydrochemical composition being constant. The value of blood pH is unchanged. In fish with a higher content of carbonic acid in blood the amount of lactate, pyruvate, oxaloacetate, malate, citrate, alpha-ketoglutarate, ammonia, glutamin, free amino acids, total and residual nitrogen rises. Simultaneously the content of urea increases in kidneys, but decrease in gills, whereas in blood and live it does not differ from the corresponding indices of fishes from the control group.

[Immobilization of Penicillium vitale glucose-oxidase on aminosilochrome and properties of immobilized enzyme].

Penicillium vitale glucose-oxidase modified by means of the carbohydrate component oxidation is added covalently to aminoorganosylochrome. The activity of the immobilized preparations is 20-38% depending on the protein-carrier ration in immobilization. Comparison of some properties of native and immobilized glucose-oxidase showed that the rH optimum of the immobilized glucose-oxidase is slightly widened towards the alkaline regions; the immobilized glucose-oxidase possesses a considerably higher pH-stability at pH alkaline values; the immobilized glucose-oxidase preparations are characterized by a significantly greater thermostability: their thermoinactivation constant at 65 degrees C is 8-10 times lower than that of the native enzyme.

[Effect of amino acids on transformation of glucose oxidase as antigen in tissues of immunized animals].

The effect of certain amino acids on transformation of glucoseoxidase as an antigen in different tissues of the animals immunized with it showed that the used glucoseoxidase of the fungus Penicillium vitale Pidopl. et Bilaj possesses antigenic properties peculiar to this enzyme isolated from other sources. 1.5 minutes after a single administration of the antigen to nonimmunized rabbits it is determined in them in descending amounts in such an order: blood, lungs, liver, kidneys, lymphatic nodes. The same order is for immunized animals, but the enzyme quantity 1.5 minutes after the last injection is lower considerably in the kidneys, spleen, lymphatic nodes and in blood serum and changes slightly in the lungs and liver. The preliminary loading of animals under experiment immunized with amino acids accelerates essentially disappearance of the antigen under determination (glucoseoxidase in the given case) in the studied tissues. The data obtained testify to the fact that acceleration of antigen disappearance may be not related to interaction with antibodies.

[Structure of cytoplasmic glycerol-3-phosphate dehydrogenase in experimental atherosclerosis in rabbits].

Differences in the ratio of molecular forms of crystalline glycerol-3-phosphate dehydrogenase (GPD, EC 1.1.1.8) from sceletal muscle and in their primary structure were found in rabbits with experimental atherosclerosis as compared with normal animals. In atherosclerosis the molecular weight of GPD and of its fragments was increased; the amino acid composition of these GPD molecules differs from that of control rabbits as shown by estimation of content of 8 amino acids. Experimental atherosclerosis is apparently related to alteration in the GPD primary structure.
