Some properties of glucose-6-phosphate dehydrogenase from rat and chick brain: a comparative study.

1. Glucose-6-phosphate dehydrogenase (G6PDH) has been purified to homogeneity from rat and chick brain by affinity chromatography with Sepharose bound 2',5' ADP. 2. Some properties of the two enzymes are studied and the effects of hydrogen ion concentration, Mg2+ ions, temperature and urea on the initial rate of enzyme are described. 3. G6PDH from chick brain differs from the rat enzyme in affinity for 2',5' ADP Sepharose, in pH optimum, in heat stability and it is differently affected by Mg2+ ions. No effect is detectable after urea treatment on enzymes from both sources.

Effect of prostaglandins in vivo on enzymes involved in thyroid carbohydrate metabolism.

Treatment of adult guinea pigs with prostaglandins produces changes in the levels of enzymes involved in carbohydrate metabolism of the thyroid gland. A decrease in glucose-6-phosphate dehydrogenase activity is observed with a concomitant increase in 6-phosphogluconic dehydrogenase; the glycolytic enzymes appear unaffected by the same treatment. The results indicate that prostaglandins do not have the biochemical effects obtained with thyrotropin and cAMP administration, showing that these compounds play an antagonistic role in comparison with the above mentioned stimulating agents.

Molecular aspects of the regulation of rat kidney alkaline phosphatase.

The mechanisms by which phosphate regulates the activity of alkaline phosphatase (orthophosphoric monoester phosphohydrolase, EC 3.1.3.1) in rat kidney were investigated. Measurements of incorporation of [(14)C]leucine into kidney alkaline phosphatase in rats fed on complete or phosphate-free diet provide evidence of a twofold increase in the rate of synthesis of the enzyme in diet-treated animals. Cycloheximide experiments indicated that control and diet-adapted enzyme decreases in activity according to first-order kinetics with a calculated half-life of 10.3 and 6.5h after complete and phosphate-free diet administration respectively. Basal and diet-adapted enzymes exhibit similar K(m) values for several phosphomonoesters and an identical degree of inhibition is produced by cysteine. In addition, the enzyme from both sources is the same with regard to heat inactivation at 45, 56 or 64 degrees C, to the profile of elution from Sephadex and to electrophoretic properties on polyacrylamide gel. A failure of rat kidney alkaline phosphatase to respond to cortisol (hydrocortisone) was also observed.
