The analysis of the hypotensive action of new piperazine derivatives in the experimental hypertension in rats.

The correlation of hypotensive action and chemical structure of some new piperazine derivatives was studied in normotensive rats and those with experimental hypertension. In the latter, all the tested derivatives, administered both once and repeatedly, appeared to have hypotensive action of various degree. In normotensive animals, there was no evidence of such action.

Catecholamines concentration in brain and heart tissue of rats and its blood pressure during chronic treatment with a new tricyclic antidepressant 10-(3-dimethylaminopropyl)-7-chlorobenzo-(b), (1,8)-naphthyridone-5 monohydrochloride (IF-C-45).

DA concentration in brain tissue decreased statisticaly after 3, 6 and 9 weeks, NA concentration also statisticaly decreased but after 3 weeks only. The NA concentration in heart tissue decreased statisticaly significant in all investigated animals. There were no statisticaly significant changes in blood pressure.

The analysis of cardiovascular action of 10-(3-dimethylaminopropyl)-7-chlorobenzo-(b)-(1,8)-naphthyridone-5 monohydrochloride (IF-C-45) with antidepressant properties.

The effects on cardiovascular system of a new compound, 10-(3-dimethylaminopropyl)-7-chlorobenzo-(b)-(1,8)-naphthyridone-5-monohydrochloride (IF-C-45) were investigated in rats, dogs and cats and compared to the effects induced by imipramine. Within the range of the analysis of peripheral cardiovascular effects IF-C-45 acts in a similar way as imipramine does. Its hypotensive and cardiodepressive action is significantly weaker than that of imipramine.

Administration, Oral↗

A comparison of antihypertensive properties of captopril and enalapril after single and multiple oral administration in spontaneously hypertensive rats.

Oral dose of enalapril or captopril moderately reduced the blood pressure of the spontaneously hypertensive rats (SHR) but not those of normotensive rats. The single daily dose of captopril (30 mg/kg) produced antihypertensive response equivalent to enalapril (3 mg/kg). Chronic administration of these drugs for 2 weeks revealed that systolic blood pressure was reduced more by captopril than enalapril. We did not find differences in hypotensive activity between Enalapril IF and Renitec Merck as well as Captopril JZF Polfa and Capoten Squibb.

Cardiovascular action of a new carbenoxolone derivative.

The circulatory effects of oleanoic acid sodium hydrogen succinate (OSS), an analogue of the anti-ulcer drug, carbenoxolone, were investigated. As carbenoxolone produces such adverse effects as sodium retention and a subsequent elevation of the arterial blood pressure in man, the present study was aimed at determining whether OSS is similar or different from it in this respect. Carbenoxolone, (43,3 mg/kg po) and OSS (66,6 mg/kg po) were given to rats twice daily for 4 weeks. The systolic blood pressure was elevated already after the first week of treatment. The hypertension was accompanied by bradycardia, which increased with the time of treatment. In the blood an increase in the creatinine level, a decrease in the urea level, and a slight elevation in sodium concentration were found after the treatment, while the potassium concentration during the whole treatment period (4 weeks) remained unchanged. Although the principal aldosterone-like effects of carbenoxolone were attributed to the oxygen presence in position 11 of the glycyrrhetinic acid ring, [8], the absence of an oxygen at that position in OSS did not cause the loss of the adverse circulatory effect.

Synthesis and some biological properties of new derivatives of 1-S-phenylthioglycolonitrile.

A series of new S-phenylthioglycolic acid derivatives was synthetized via common transformations of alkylated S-phenyl-thioglycolonitrile. The compounds were submitted to pharmacological screening. N,N-Diethylaminoethyl ester of 1-S-phenylcyclopentane carboxylic acid was submitted to detailed studies and showed high local anesthetic properties.
