[Relationship between an involvement of Oddi's sphincter in a choledochopancreatic junction and clinical appearances in 120 patients with anomalous arrangement of the pancreatico-biliary ductal system].

We studied relationship between an involvement of Oddi's sphincter in a choledochopancreatic junction and clinical appearances in 120 patients with anomalous arrangement of the pancreaticobiliary ductal system. The 120 composed of 83 with a sphincteric mechanism at the junction of the common bile and pancreatic duct and 37 without the sphincteric mechanism. We found that influences of sphincteric mechanism at the junction closely related to the length of the common duct between an orifice of the papilla of Vater and a junction of the common bile and pancreatic duct. And in all 83 with a sphincteric mechanism at the junction the length of the common duct showed less than 1.5 cm, and in all 37 without a sphincteric mechanism at the junction the common duct showed more than 1.5 cm in length. There was no correlation between a distribution of the sphincter muscle and clinical appearances, association with congenital choledochal cyst, cholelithiasis and bile duct cancer in the 120 patients.

[Para-esophageal anastomosis of left atrium in orthotopic heart transplantation in dogs: possibility for electrophysiologic examination of the transplanted heart with transesophageal atrial stimulation].

Para-esophageal anastomosis of left atrium was examined in orthotopic heart transplantation as a new surgical technique for transesophageal electrophysiologic study of the transplanted heart. Seven orthotopic heart transplantations were performed in mongrel dogs. Left atrial appendage of the donor heart was trimmed as a flap and the sinus venosus of right atrium was incised posteriorly. An upper midline incision was made in the posterior wall of left atrium of the recipient after native ventricles were resected. Left atrial appendage flap of the donor heart was anastomosed to the posterior incision of native left atrium, and the remains of left atrium were sutured continuously. Sinus venosus incision of the donor heart was anastomosed to the right atrium of recipient. Ascending aorta and pulmonary trunk were reconstructed in the standard fashion. Transesophageal atrial pacing was possible during and after weaning from cardiopulmonary bypass in all seven cases. The appendage flap was found to occupy one third of the posterior wall of left atrium at the time of sacrifice. It was suggested that para-esophageal anastomosis of left atrium would be an useful technique for transesophageal electrophysiologic study of the transplanted heart.

[Results of noncurative resection of primary lung cancer].

Out of 1099 cases of primary lung cancer patients who underwent surgical treatment in our department from 1952 to 1988, 374 cases were divided as noncurative by the post surgical examination. We divided the causes of noncurative resection as follows; I) absolute noncurative resection 1) tumor rest in a) lymph nodes (Ly rest), b) chest wall or diaphragma (Ch + Dia rest), c) bronchial stump (Bro rest) 2) intra-pulmonary metastasis (pm), 3) dissemination or pleuritis carcinomatosa (Dis + pl) II) relative noncurative 1) incomplete dissection of lymph nodes (Ro) 2) group 2b lymph node metastasis (2b Ly). Ly rest was the commonnest (103 cases 27.5%) in noncurative resection and Ch + Dia rest (94 cases, 25.1%) and p.m. (54 cases 14.4%) followed thereafter. Two thirds of the noncurative cases were classified into these 3 groups, and the rest of the causes were Dis + pl, Bro rest. RO and 2b Ly groups. p.m. and Ly rest groups were the most common causes of noncurative resection in adenocarcinoma cases whereas Ly rest and Ch + Dia rest groups were common in squamous cell carcinoma cases. Five year survival rate of the total noncurative cases was 12.1% although those of Dis + pl groups was 22.9% and those of p.m. group was 18.8%. Five year survival rate of the other groups were around 10%. A slightly better survival rate was observed when relative noncurative resection was performed in RO (19.7%) and 2b Ly (18.7%) groups.

Molecular cloning and characterization of cDNA for the precursor of rat pituitary adenylate cyclase activating polypeptide (PACAP).

We have isolated and cloned a cDNA for rat pituitary adenylate cyclase activating polypeptide (PACAP) precursor protein from the brain. The nucleotide and the deduced amino acid sequences revealed that the rat PACAP cDNA encoded a 175 amino acid protein highly homologous to the ovine and human PACAP precursors. The rat PACAP precursor protein included a putative signal peptide at the amino-terminus, a PACAP-related peptide (PRP) of 29 amino acid residues and the rat PACAP of 38 amino acid residues flanked by basic processing sites, and a carboxy-teminus amidation signal for PACAP. Furthermore, the amino acid sequence of the rat PACAP was completely identical to that of the ovine and human PACAP. Northern blot analysis of rat brain RNA revealed an approximately 3.0 kb transcript for the PACAP precursor in rat brain.

Adrenal pheochromocytoma PC12h cells respond to pituitary adenylate cyclase activating polypeptide.

An adrenal pheochromocytoma cell line, PC12h, was found to respond to a novel hypothalamic neuropeptide, Pituitary Adenylate Cyclase Activating Polypeptide (PACAP). The cells elevated both intracellular and extracellular cAMP levels on stimulation by PACAP, whereas they showed little response to VIP which is structurally related to PACAP. Using [125I]PACAP27 (a shorter form of the peptide) and [125I]VIP, we found large amounts of specific binding sites for PACAP but few binding sites for VIP in PC12h cells. These results indicate that PC12h cells respond to PACAP via a specific PACAP receptor.

Specific expression of human endothelin-2 (ET-2) gene in a renal adenocarcinoma cell line. Molecular cloning of cDNA encoding the precursor of ET-2 and its characterization.

Analysis of culture medium of human renal adenocarcinoma cells ACHN by RP-HPLC suggested that the cells specifically secreted human endothelin-2 (ET-2). cDNAs encoding human ET-2 precursor were cloned from a cDNA library constructed with mRNA derived from the ACHN cells, and the nucleotide and deduced amino acid sequences were determined. The ET-2 cDNA was revealed to contain 1.3 kb and encode prepro-ET-2 protein consisting of 178 amino acid residues. The ET-like sequences found in the prepro-ET-1 and -ET-3 was conserved in this prepro-ET-2. The Northern blot analysis of mRNA suggested that the transcript of ET-2 gene was 1.4 kb. This is the first direct evidence that human ET-2 gene was expressed specifically in tumor cells.

Insulin-like growth factor-I stimulates endothelin-3 secretion from rat anterior pituitary cells in primary culture.

Since we found relatively high concentrations of immunoreactive (ir-) ET-3 in the rat pituitary gland (190 pg/g tissue), we have investigated the possible ET-3 secretion from the primary culture of anterior pituitary cells and the effects of various growth factors on the ET-3 secretion. The ir-ET-3 was detected in the incubation medium within 2 h, and 24 h of culture attained the concentrations of 1.15 +/- 0.26 pg/well/6 x 10(5) cells. The ir-ET-3 secretion was stimulated by insulin, insulin like growth factor-II (IGF-II), and most effectively by insulin like growth factor-I (IGF-I) in a dose- and time-dependent manner, whereas the production of ir-ET-1 and ir-big ET-1 was slightly inhibited by IGF-I and IGF-II. In reverse-phase HPLC, the ir-ET-3 released into the culture media showed identical retention time with authentic ET-3. Although ir-ET-1 and ir-big ET-1 secretion was stimulated by transforming growth factor-beta (TGF-beta), ir-ET-3 secretion was inhibited. These results indicate that the anterior pituitary cells secrete ET-3 and the secretion is stimulated by IGF-I.

Solubilization of receptor for pituitary adenylate cyclase activating polypeptide from bovine brain.

A receptor for pituitary adenylate cyclase activating polypeptide (PACAP) was solubilized from bovine brain membranes with 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate. Saturable binding experiments using [125I]PACAP27 revealed that the solubilized extract contained a single class of binding sites with a Kd of 200 pM and a Bmax of 0.9 pmol/mg protein. The Kd did not change significantly after the solubilization. Competitive binding experiments confirmed that the solubilized receptor retained a specificity for PACAP. Thus, the solubilization of the receptor in an active form was successfully achieved by the present procedure.

Molecular identification of receptor for pituitary adenylate cyclase activating polypeptide.

Pituitary adenylate cyclase activating polypeptide (PACAP) is a novel hypothalamic peptide with 38 (PACAP38) or 27 (PACAP27) amino acid residues, structurally related to vasoactive intestinal peptide (VIP). Bovine brain membrane has a PACAP specific receptor interacting with both PACAP27 and PACAP38. Affinity-labeling of the receptor with [125I]PACAP27 identified a dominant band of Mr = 60 k. The labeling density of the 60 k band decreased in the presence of unlabeled PACAP27 or PACAP38, whereas the 60 k band remained in the presence of unlabeled VIP. Binding of [125I]PACAP27 to the membrane decreased in the presence of GTP and the labeling density of the 60 k band decreased concomitantly. The results indicate that bovine brain has a specific PACAP receptor, whose apparent molecular weight is 57 k (substracting the molecular weight of [125I]PACAP27 from 60 k).

Isolation of a neuropeptide corresponding to the N-terminal 27 residues of the pituitary adenylate cyclase activating polypeptide with 38 residues (PACAP38).

A novel neuropeptide with 38 residues (PACAP38) was isolated from ovine hypothalamic tissues using the pituitary adenylate cyclase activation in rat pituitary cell cultures as a parameter of the biological activity (Miyata et al, Biochem. Biophys. Res. Commun. 164, 567-574, 1989). From the side fractions obtained during the purification of PACAP38, a shorter form peptide with 27 residues corresponding to the N-terminal 27 amino acids of PACAP38 and amidated C-terminus was isolated and named as PACAP27. Synthetic PACAP27 showed a biological activity of adenylate cyclase stimulation comparable to PACAP38. Moreover PACAP27 which shows a considerable homology with vasoactive intestinal polypeptide (VIP) demonstrated a similar vasodepressor activity as VIP, but the adenylate cyclase stimulating activity was about 1000 times greater than VIP.

Endothelin-3 concentrations in human plasma: the increased concentrations in patients undergoing haemodialysis.

Plasma immunoreactive endothelin-3 (ir-ET-3) concentrations were measured by a sandwich-enzyme immunoassay (sandwich-EIA) for endothelin-3 (ET-3). The assay method consists of two antibodies directed against N-terminal and C-terminal portions of ET-3. It detects as little as 0.1 pg/well of ET-3 without the crossreaction with endothelin-1, endothelin-2 and big ET-3. Plasma ir-ET-3 concentrations were found to be 0.45 +/- 0.07 pg/ml (mean +/- SD) in healthy volunteers, and were increased in patients undergoing haemodialysis (0.83 +/- 0.26 pg/ml, p less than 0.001). In reverse-phase HPLC, ir-ET-3 in normal plasma and in plasma of haemodialysis patients was eluted at the position of authentic ET-3, indicating that ir-ET-3 in plasma detected by the EIA was ET-3 itself. These results suggest that circulating ET-3 exists in normal human plasma and that production and/or metabolism of ET-3 may be altered in patients undergoing haemodialysis.

A 1H-NMR study of the solution conformation of Icaria chemotactic peptide and its [Lys7] analog: effects on the physiological activity of a substitution of proline to lysine at position 7.

Solution conformations of Icaria chemotactic peptide isolated from the Ropalidian wasp venom and its [Lys7] analog have been analyzed by the use of two-dimensional proton nuclear resonance spectroscopy. It has been shown that Icaria chemotactic peptide takes alpha helical conformation in the C-terminal 8-13 segment with the N-terminal 1-7 segment disordered. By contrast, the [Lys7] analog takes alpha helical conformation over a wider range from residues 3 to 13. The present results indicate that the substitution of proline to lysine at position 7 results in a drastic change in solution conformation, providing the Icaria chemotactic peptide with new physiological functions.

Role of lysine residue at 7th position of wasp chemotactic peptides.

Most of chemotactic peptides isolated from various kinds of wasp venom have lysine residue at 7th (or 8th) position, but only a chemotactic peptide from Icaria sp. has no basic amino acid residues in the sequence. The relation of chemotaxis with other biochemical activities such as superoxide generation and lysosomal enzyme release from guinea pig neutrophils was studied by the use of substitution analogs of Icaria chemotactic peptide at 7th position. Findings revealed two distinct ways of chemoattractant signal transduction in neutrophils.

Plasma concentrations of endothelin-1 in spontaneously hypertensive rats and DOCA-salt hypertensive rats.

To investigate the possible involvement of endothelin-1 (ET-1), an endothelium-derived potent vasoconstrictor peptide, in the pathophysiology of hypertension, plasma ET-1 levels in 15-week-old spontaneously hypertensive rats (SHR) and DOCA-salt hypertensive rats were measured with a sandwich-type enzyme immunoassay. The vasocontractile effect of ET-1 in aortic helical preparations was significantly more sensitive in DOCA-salt hypertensive rats than in control sham-operated rats, but plasma levels of ET-1 did not differ between them. Plasma ET-1 levels in genetically hypertensive rats (SHR and stroke-prone SHR) were significantly lower than those in age-matched normotensive Wistar-Kyoto (WKY) rats. The plasma concentrations of big ET-1, a precursor of ET-1, in both SHR and SHR-SP were significantly lower than those of WKY, suggesting that the production of ET-1 is decreased in rats of genetic hypertension. Although the vascular reactivity to ET-1 increased in both DOCA-salt hypertensive and genetically hypertensive rats, present findings of the plasma ET-1 levels suggest that the role of ET-1 in the vascular control system may be different in DOCA-salt hypertensive rats and genetically hypertensive rats.

A sandwich-type enzyme immunoassay to detect immunoreactive big-endothelin-1 in plasma.

A sensitive sandwich-type enzyme immunoassay (sandwich-EIA) has been established for human big endothelin-1 (big-ET-1). A monoclonal antibody AwETN40 which recognizes the N terminal portion of big-ET-1 was used as an immobilized antibody. The Fab' fragment of rabbit antibodies to the human big-ET-1 C terminal peptide (22-38) was used as the enzyme-labeled antibody after being coupled to horseradish peroxidase (HRP). The assay is sensitive enough to detect as little as 0.2 pg/well (5 x 10(-17) mol/well) of big-ET-1 without crossreactivity with ET-1. Immunoreactive big-ET-1 in plasma was determined after being extracted with Sep-pak C-18 cartridges. The average levels were 5.2 +/- 1.0 pg/ml for males and 5.7 +/- 1.6 pg/ml for females (the values were corrected for recovery efficiency). Big-ET-1 was found to be eliminated from the blood stream at a slower rate than ET-1. This may be an important reason for the higher plasma levels of immunoreactive big-ET-1 compared to the levels of immunoreactive ET-1.

One of the endothelin gene family, endothelin 3 gene, is expressed in the placenta.

A cDNA encoding human endothelin 3 (ET-3) precursor was cloned from a cDNA library from the placenta, and its nucleotide and deduced amino acid sequences were determined. This ET-3 cDNA was found to contain 2.3 kb pairs and encode prepro-ET-3 protein consisting of 224 amino acid residues. The putative big-ET-3 seems to consist of 42 amino acid residues. Two of the intron insertion sites were determined with information from nucleotide sequences of the cloned genomic ET-3 gene. This is the first direct evidence that the ET-3 gene is transcribed and expressed in the placenta.
