Isolation and structure of the Streptococcus faecalis sex pheromone, cAM373.

The Streptococcus faecalis sex pheromone, cAM373, which induces a mating response of donor cells harboring plasmid pAM373 and is also produced by Staphylococcus aureus, was isolated and its structure determined. Supernatant from an overnight culture of a recipient strain was subjected to successive purification procedures, and 4.4 micrograms cAM373 was obtained. The isolated pheromone showed activity at a concentration as low as 5 X 10(-11) M. Sequence analysis indicated that cAM373 was a heptapeptide, H-Ala-Ile-Phe-Ile-Leu-Ala-Ser-OH, and that its Mr was 733. A synthetic replicate of the peptide showed the same biological activity and chromatographic behavior as the native cAM373.

New optical spike in frog twitch muscle fibers injected with a potential probe dye NK2935.

Absorbance responses posterior to action potentials were recorded from frog twitch fibers loaded by an impermeant potential probe dye NK2935 that was chemically identical with WW781. Extracellularly loaded NK2935-related responses (EOS) increased in absorbance around 640nm wavelengths and were like action potential in waveform but were slightly preceded by action potential. Model analysis showed that EOS consisted of two components, namely 39% non-delay component reflected with sarcolemma action potential and 61% first-order delay component reflected with transverse tubule (T) action potential (delay-constant tau 1 = 0.48 ms). The T-disrupted fibers showed only the non-delayed response. Intracellularly injected NK2935-related responses (IOS) were also composed of a single spike of the same direction swing as in the EOS case described by the term "ipsidirection," but were clearly preceded by EOS and followed by Ca transient, birefringence, transparency and latency relaxation responses. The T-disrupted fibers showed a small "sidedness" response characterized by both opposite direction swing, namely "contradirection" and non-delay against EOS. Model analysis showed that IOS consisted of two main components, namely second-order delay component with ipsidirection (tau 2 = 0.8 ms) and the further high-order delay component with contradirection. Based on spatio-temporal heterogeneity of NK2935 binding inside fibers and potential-absorbance relation in sarcolemma, the former origin was thought to be in junctional sarcoplasmic reticulum (jSR) responding to T excitation with a transient "positive polarization" estimated roughly 20 mV. Similarly, the latter was possibly in free SR where a slow "negative polarization" estimated -0.9 mV might appear associated with Ca release.

Rearrangement of immunoglobulin kappa gene in a variant-type Burkitt lymphoma cell line, KOBK101, carrying a translocation between chromosomes 2 and 8.

The immunoglobulin kappa light-chain gene was rearranged in a Burkitt lymphoma cell line with the t(2;8) (p11;q24) translocation, KOBK101, showing recombination between the variable and the joining regions on one allele and an aberrant substitution of the region upstream of the J5 segment with an unknown sequence on another allele. Because DNA segments homologous to the consensus sequences of joining and switching regions of immunoglobulin genes were detected in the unknown sequence, we speculate that the system for either V-J or switching recombination in immunoglobulin genes may be involved in forming this unusual rearrangement.

Identification of new gene products coded from X regions of human T-cell leukemia viruses.

Antibodies were raised against oligopeptides deduced from the nucleotide sequence in the conserved region located between env and the 3' long terminal repeat in human T-cell leukemia virus type I (HTLV-I) and type II (HTLV-II) to detect a protein coded from this region in virus-infected cells. Two of these antibodies precipitated a protein of 41 kilo-daltons in HTLV-I-infected cell lines and a protein of 38 kilo-daltons in HTLV-II-infected cells. The protein in HTLV-I-infected cells was precipitated by plasma from patients with adult T-cell leukemia but not by plasma from a normal adult. These results indicate that these proteins were translated from new coding regions (X) present in HTLV-I and HTLV-II.

Chromium-induced pulmonary cancer. Report of a case and a review of the literature.

A 57-year-old man, whose occupational history was that of a chromate worker for about 35 years, suffered nasal perforation during and died due to pulmonary cancer. In the autopsy materials, the concentration of chromium in the lung tissues was very high, being about 90 times the amount found in normal lungs. The chromium particles in the lung tissues could not be detected by analytical electron microscope, but they could be found in the anthoracotic lesions of the lungs by means of an X-ray microanalyser. The present case was judged to be a case of chromium-induced pulmonary cancer since all the pathologic data obtained were similar to those found in similar cases reported in the literature.

Isolation and structure of the bacterial sex pheromone, cAD1, that induces plasmid transfer in Streptococcus faecalis.

The Streptococcus faecalis sex pheromone cAD1, which is involved in the conjugative transfer of the hemolysin plasmid pAD1, has been isolated and its structure determined. Its Mr is 818 and its amino acid sequence is H-Leu-Phe-Ser-Leu-Val-Leu-Ala-Gly-OH. A replicate of the pheromone synthesized by the liquid-phase method showed the same biological activity and chromatographic behavior as the isolated cAD1. Pheromone activity was detectable at a concentration of approximately 5 X 10(-11) M.

Effect of an intracellular calcium antagonist (TMB-8) on carbamylcholine-induced amylase release from dispersed rat pancreatic acini.

During 10-min incubation with increasing concentrations of carbamylcholine (carbachol), amylase release from dispersed rat pancreatic acini increased, became maximal at 2 X 10(-6)M and then decreased. In the concentration range of 10(-7)M to 10(-4)M, 8-(N,N-diethylamino)-octyl 3,4,5-trimethoxybenzoate hydrochloride (TMB-8) caused a dose-dependent inhibition of amylase release induced by a submaximal concentration of carbachol. No inhibitory effect was observed on basal and secretin-stimulated amylase release. TMB-8 showed a significantly greater ability of blocking the action of carbachol than verapamil and diltiazem. TMB-8 could reverse the submaximal stimulation of amylase release caused by supramaximal concentrations of carbachol to a maximal stimulation, while verapamil and diltiazem could not. These results confirm the hypothesis that mobilization of intracellular calcium is the primary step in the action of carbachol on pancreatin acinar cells and contributes to the submaximal secretory response of acinar cells induced by high concentrations of carbachol.

The activity of synthetic analogs of serum thymic factor (FTS) to convert mouse pre-T cells into Thy-1 positive cells.

Serum thymic factor (FTS) and its 33 analogs were compared with regard to the ability to induce Thy-1 antigen on mouse pre-T cells. The pre-T cells were prepared from spleen cells of athymic nu/nu mice by passing them through a nylon wool column, and the induction of Thy-1 antigen was analysed using a fluorescence-activated cell sorter. Thy-1 negative cells were converted into Thy-1 positive cells by FTS and some of its analogs. Deletion of pGlu at the amino terminus of FTS did not alter the activity. Des ( pGlu1 , Ala2)-FTS and des ( pGlu1 , Ala2, Lys3 )-FTS were inactive and deletion of the carboxyl terminal residue, Asn, also resulted in the loss of activity. Substitution or modification of pGlu1 and Asn9 with an amino acid, an acyl group or an amine caused a marked reduction of the activity. The residue 3, Lys, could be substituted without any loss of the activity by Arg, but not by Leu. These results show that the octapeptide moiety, Ala-Lys-Ser-Gln-Gly-Gly-Ser-Asn, is the minimum essential part of the FTS molecule for the expression of activity and they suggest that the positive charge at the residue 3 is also important for its activity.

Echocardiographic assessment of left ventricular function in patients with complete left bundle branch block.

To elucidate the mechanism, cardiac function and prognosis of complete left bundle branch block (CLBBB), 32 patients with CLBBB were studied using echocardiographic and vectorcardiographic methods. They were classified into 5 types: according to the presence or absence of early systolic notch and paradoxical motion of interventricular septum. Type I, an early systolic notch and paradoxical motion of the interventricular septum; type II, an early systolic notch and a flat septal motion, type III, an early systolic notch but no paradoxical motion; type IV, an early systolic notch of a slight degree but no paradoxical motion; type V, showed neither early systolic notch nor paradoxical motion. Type I patients had enlarged left ventricular dimension, severe cardiac damage and poor prognosis while type IV patients had a good ventricular function and prognosis with no complications. In type V patients, intraventricular conduction disturbances were apparent; a wide QRS loop in horizontal plane with no significant terminal delay. In type II and III, there were no typical findings. The histology revealed that the left bundle branch block was located at the bifurcation.

[Multiple duodenal carcinomas associated with multiple gastric cancers].

Because of the rarity of primary duodenal cancer, the coexistence of this disease with gastric cancer is very rare. The case presented herein suffered from two carcinomas of the duodenum and six gastric cancers. The patient was 71-year-old man who underwent subtotal gastrectomy and pancreatoduodenectomy. Pathologic studies showed that all of the gastric and one of the duodenal cancers were limited to the mucosal layer and that the other duodenal tumor invaded to the pancreas. Several cases of double cancers in the duodenum and the stomach have been reported in Japan, and this is first of multiple carcinomas of the duodenum associated with multiple gastric cancers.

[Two cases of malignant cardiac tumors attached on the anterior mitral valve].

Two cases were reported in which malignant cardiac tumors attached to the mitral valve were diagnosed during their lifetimes using two-dimensional echocardiography. Case 1 was a 29-year-old man with speech disturbance and left hemiparesis. Histological examination of the specimen excised from the cardiac tumor during the operation revealed mesenchymal chondrosarcoma, which is extremely rare etiologically and has never been reported so far. Case 2 was a 62-year-old woman complaining of paroxysmal nocturnal dyspnea and chest pain. The diagnosis of malignant histiocytosis was made from the pathological examination of biopsy specimen taken from the rib metastasis. Using two-dimensional echocardiography, characteristic findings for the cardiac tumor were obtained. The tumor echo in Case 1 showed, unlike to that reported for myxoma, two different echogenic layers; the outer dense and the inner light in the echo density. On surgery, tumor echo was revealed to reflect the cystic lesion. In Case 2, two-dimensional echocardiography on admission revealed two separate tumor echoes which attached to the mitral valve and left atrial wall region, respectively. Within two months, they grew rapidly and finally fused into one mass resulting in so-called ball valve syndrome. Phonocardiographically, the tumor plop in Case 1 was high-pitched in quality, and was extinguished completely after the tumor was resected. The tumor plop in Case 2 was not audible on admission, but became evident after fusion of the tumor echoes and was associated with a presystolic murmur.

The role of the N-terminus in the active conformation of the substance P analogues.

The role of the N-terminus in the activity of substance P analogues was studied with the isolated rat spinal cord and the guinea-pig ileum. When the C-terminal Leu10 or Met11 was replaced by the D-isomer, the analogues without the N-terminal tetrapeptide were 40-100 times less potent than those with the N-terminus in both motoneuron-depolarizing and ileum-contracting actions. These results that the N-terminus may stabilize the active conformation of the substance P analogues.
