Effects of relative metabolic rate and heart rate variation on the performance of flight attendants.

The main work of the cabin attendants in an actual flight in service for passengers. The effects of flight attendant duties in flight differ from the effects of the same tasks performed on the ground. In this study, the relative metabolic rate (RMR) and heart rate (HR) of cabin attendants in a cruising aircraft galley and cabin are compared with those of a crew working in a mock-up apparatus on the ground. The types of work tested are: (a) oshibori (steamed towel) service, (b) soft drink service, (c) setting meal tray, (d) putting casserole on tray, (e) meal tray service, (f) walking on aisle. The RMR at each type of work during flight is as indicated: (a) 1.07-2.10, (b) 1.08-1.54, (c) 1.37-1.82, (d) 2.57-3.50, (e) 2.11-3.10 and (f) 1.84. The range of HR was: (a) 105-120, (b) 90-110, (c) 90-120, (d) 100-130 and 100-140 beats/min. In most cases, the RMR and HR levels of work done in the mock-up were lower than those recorded in flight. These results suggest that the oxygen intake of work done in flight is greater than that on a mock-up. One of the reasons might be that the cabin barometric pressure (ca. 660 torr or cabin altitude ca. 1,500 m) or an aisle inclination of about 3 degrees caused a decrease in the efficiency of oxygen intake during flight.

Intraprostatic lymphatics in man: light and ultrastructural observations.

Despite reports that human prostate is without lymphatics, electron microscopic studies of prostatic curettings of human prostate have demonstrated unequivocal evidence that lymphatics exist in prostate proper. Such channels are found in two locations: between the muscle cells and in the fibrous stroma supporting the muscle bundles. The lymphatics between the muscle cells are of smaller caliber and probably represent initial lymphatics. They are intimately connected by anchoring filaments and microfibrils to the smooth muscle cells. The functional relationship between these has been discussed. No lymphatics were seen in subepithelial location. We saw only blood capillaries in that area.

A light and electron microscopic study with comments on their relation to malignant fibrous histiocytoma and angiosarcoma.

Analysis of 365 cases of malignant soft tissue tumors revealed 83 cases of liposarcoma. The ages of the patients were distributed between 18 and 86 years with a mean average of 54.3 years. Approximately 50% of the cases were located in the lower extremity. Histologically, they consisted of 5 types; well differentiated (17 cases), myxoid (49 cases), round cell (3 cases), pleomorphic (11 cases), and mixed (3 cases). The appearance of lipoblasts of either the signet-ring type or mulberry (multivacuolated) type with displaced nuclei was the common characteristic feature for all types. Electron microscopically, lipid droplets were found inside of endoplasmic reticulum in lipoblasts, and abundant glycogen granules were seen in the cells containing only few lipid droplets. The lipoblasts were usually located close to the vascular wall in the interstitium, and findings suggesting an intimate relation between pericytes and lipoblasts were encountered. Although the differential diagnosis of malignant fibrous histiocytoma and liposarcoma was made possible by the appearance of lipoblasts in the latter, storiform pattern and histiocyte-like cells sometimes appeared in poorly differentiated liposarcoma. This tendency was also found in angiosarcoma. A close follow-up of recurrent cases revealed that there are actually cases of liposarcoma with malignant fibrous histiocytoma-like pattern and angiosarcoma with malignant fibrous histiocytoma-like pattern. It was considered that malignant fibrous histiocytoma-like figures can appear in various poorly differentiated sarcomas.

Stereoscopic scanning electron microscopy of the red pulp of dog spleen with special reference to the terminal structure of the cordal capillaries.

In order to obtain direct morphological information about the three-dimensional fine structure of the splenic terminal vascular bed, especially the terminating mode of the cordal capillaries, stereoscopic scanning electron microscopy of perfusion-fixed and freeze-fractured red pulp of a normal dog spleen was undertaken. An improved method of perfusion-fixation was utilized in which the hydrostatic pressures of the splenic artery and vein were maintained at approximately the same levels as those in the living state. Stereoscopic observations of scanning electron micrographs clearly demonstrated the three-dimensional fine architecture of the splenic sinuses, the spongy cordal reticular tissue and the intracordal vasculature. The cordal capillaries terminate in the labyrinthine cordal space according to a certain mode in which the walls of the terminals are transformed into a meshwork structure continuous with the cordal reticular tissue owing to an increase in number and size of fenestrations. No evidence could be detected to prove or suggest any direct continuity of the capillary end with the splenic sinus. These results strongly support the concept of an "open circulation", at least in the red pulp of the dog spleen, with the possibility of a "functionally closed circulation" under some physiological conditions.

Ultrastructure and histochemistry of the medullary interstitial matrix of rat kidney.

Medullary tissue of the normal rat kidney was perfused with 3 percent glutaraldehyde (GA), incubated in 0.5 percent cetyl pyridinium chloride and postfixed in 1 percent OsO4. In comparison with the ordinary fixation with GA and OSO4, the medullary interstitium represented abundant matrical substance that is rich in acid mucopolysaccharides (AMPS) and morphologically represents a diffuse reticular structure consisting of 30 to 150 a thick microfibrils and granular structures of 300 to 500 A in diameter. When chondroitinase was applied before OsO4 treatment, the dense granes disappeared and the microfibrils were replaced by loosely textured 30 A thick microfilaments. After hyaluronidase treatment the microfibrils disappeared and most granules changed into a ring-shaped structure with an electronlucent central portion. These results suggest that the reticular structure consists of microfilaments of hyaluronates and amorphous masking substance of chondroitin sulfates. In the dense granule, hyaluronates become concentrated in the central portion and chondroitin sulfate in the peripheral zone. When perfused with a CPC-containing GA, the medullary interstitium was diffusely filled with a large amount of fine granular substances suggesting the presence of water soluble free AMPS filling the reticular space.

Morphological and biochemical alteration in the rat liver induced by maprotiline.

The fatty change in the liver induced in male rats by a new antidepressant, maprotiline, given at high-dose was studied morphologically and biochemically. The accumulation of triglyceride in the liver was related to accelerated synthesis of fatty acid in the liver in males, which liver triglyceride was decreased in spite of increased synthesis of fatty acid from acetate in females. Secretion of lipoprotein granules was noted in both sexes. The sexual differences of triglyceride contents in the liver and serum was influenced by sexual hormone. Proliferation of smooth endoplasmic reticulum and "fingerprints" were found in treated rats and these were correlated with the induction of hepatic microsomal drug metabolizing enzymes. The "myeloid bodies" induced in hepatocytes were considered to be of lysosomal nature. Both morphological and biochemical alterations in the hepatocytes induced by maprotiline was reversible upon cessation of treatment.

Giant mitochondria in the epithelial cells of the proximal convoluted tubules of diseased human kidneys.

Electron microscopy of 100 renal biopsies obtained at random from patients with various renal disorders revealed gigantic mitochondria in the cytoplasm of the epithelial cells of the proximal tubules in 36 cases (36 per cent). These giant mitochondria are usually round, ovoid, spindle-shaped or rod-shaped often reaching the size of the cell nucleus, and appear singularly and sporadically in the epithelial cell lining of the tubules. Once recognized by electron microscopy they are easily identified in routine histologic preparations and show the same staining characteristics as other mitochondria. In electron microscopy, giant mitochondria have a double limiting membrane, abundant matrical substance, and few remnants or cristae. In addition, they are characterized by (1) intramatrical irregularly branching fibrils approximately 35 A thick, (2) intramatrical bundles of straight filaments approximately 60 to 70 A thick, (3) intramatrical electron-opaque globules up to 0.6 mum., and (4) helical filaments in focally dilated spaces of the outer compartment. The significance of such a remarkable morphologic change of tubular mitochondria and the nature of the unusual components are at present unknown, but an abnormal metabolic condition is considered in its pathogenesis. Nothing specific has yet been found in the relationship between the positive appearance of giant mitochondria and types of renal disorders or clinical and laboratory data. It may be postulated from the present study that "giant mitochondrial tubulopathy" is a rather common, characteristic, and noteworthy change in the histopathology of renal tubule.

Prognostic status of p53 gene mutation in canine mammary carcinoma.

BACKGROUND: The p53 gene mutations have been associated with the development of human breast and canine mammary neoplasms; breast carcinoma patients with alterations of p53 gene are considered to have a poor prognosis. Mammary carcinoma represents the most common malignant tumor in female dogs. However, the prognostic significance of p53 gene mutation in the dog has been unclear. STUDY DESIGN: The alteration in exons 5-8 of p53 gene in 69 canine mammary carcinomas were investigated by PCR-SSCP with direct sequence analysis and statistically analyzed to compare with other clinicopathological parameters including age, neuter, tumor size, stage, histology, p53 expression, recurrence and death from carcinoma. RESULTS: 12 out of 69 (17%) carcinomas showed p53 gene mutations. After a follow-up period of 30 months, multivariate regression analysis revealed that p53 gene mutation was only an independent risk factor for increased risk of the recurrence and death from mammary carcinoma. CONCLUSION: The p53 gene alterations might contribute to the prognostic status in canine mammary carcinomas, in a way comparable to that of human tumors.
