Results of radiotherapy in control of stage I and II non-Hodgkin's lymphoma.

We retrospectively analyzed 114 patients with non-Hodgkin's lymphoma, clinical stages I and II, classified by the criteria of Rappaport and treated by radiotherapy alone. Of 84 patients classifiable, one-third were nodular and two-thirds diffuse lymphomas. Berkson-Gage actuarial and relapse-free survivals were determined for these two groups and for subgroups stratified by histology, stage, and by presence or absence of extranodal disease. Five year relapse-free and overall survivals were 83% and 100%, respectively, for the nodular group and 37% and 59% for the diffuse group. Extranodal involvement was less frequent in the nodular (19%) than in the diffuse (52%) group, where it was associated with Stage IE disease and increased relapse-free and actuarial survival. Histopathological subtype in the diffuse group (histiocytic versus combined lymphocytic poorly differentiated and mixed lymphocytic-histiocytic) did not influence survival. Extranodal involvement and stage I disease were associated with better survival in the diffuse histiocytic group. Successful radiotherapy for all stages of disease, all histologies, was not correlated with extended versus involved fields, and 89% of the relapses in the entire series were by wide dissemination.

Nerve growth factor prevents vinblastine destructive effects on sympathetic ganglia in newborn mice.

Vinblastine injections in newborn mice produce severe atrophy of sympathetic ganglia; after a 7-day treatment the ganglia are 80% reduced in volume. Histological examinations show that this effect is due to a marked decrease in the neuronal cell population. The most precocious ultra-structural alterations are localized in the nuclear compartment, followed by axonal swelling and microtubule disappearance. Simultaneous injections of nerve growth factor entirely prevent the noxious effects of the vinca alkaloid, and result in partial appearance of the growth effects of nerve growth factor. Such protective action is not due to inhibition of vinblastine uptake which is the same in control mice and in mice pretreated with nerve growth factor. It is suggested that nerve growth factor prevents the vinca alkaloid action by favoring the assembly or organization, or both, of microtubules, which, from in vitro studies, have been proved to be inaccessible to vinblastine.

Modification of DNA patterns in plasma and nucleated blood cells from systemic sclerosis patients.

OBJECTIVE: To analyze the DNA patterns extracted from plasma and nucleated blood cells (lymphocytes) in systemic sclerosis (SSc) with a new MFC DNA extracting kit. METHODS: Ten SSc patients and 9 healthy controls were studied. Heparin containing blood samples were separated into plasma and buffy coat fractions and subjected to DNA extraction. The DNA pattern was revealed by 0.4% agarose electrophoresis and analyzed in a Gelblot Programme file (UVP Product). RESULTS: In control samples the DNA pattern observed in plasma extract was different from that of the buffy coat. For the plasma a series of peaks ranging from 2-23 Kb were present, and for the buffy coat we usually observed 2 to 3 principle bands, respectively, at around 33 Kb and 0.5 Kb. For SSc patients the DNA patterns that resulted from the plasma and buffy coat were totally different from the control samples, with some exceptions. CONCLUSION: We observed that SSc samples contain a distinctively different DNA pattern compared to healthy controls. Further studies are needed to establish whether or not this DNA pattern might be considered peculiar to SSc, and whether or not the method is a useful tool for pathogenic studies of the disease and for diagnostic purposes.
