Inhibition of five lipoxygenase activating protein (FLAP) by MK-886 decreases atherosclerosis in apoE/LDLR-double knockout mice.

BACKGROUND: Recent reports point to an important role of leukotrienes in atherogenesis. Leukotrienes are produced by 5-lipoxygenase co-operating with five lipoxygenase activating protein (FLAP). We hypothesized that MK-886, an inhibitor of FLAP, could attenuate the development of atherosclerosis in the atherogenic apolipoprotein E/low density lipoprotein receptor (apoE/LDLR) double knockout (DKO) mouse model. MATERIALS AND METHODS: Female apoE/LDLR-DKO mice at the age of 8 weeks were put on Western diet. The experimental group (n = 10) received the same diet as the control group (n = 10), but mixed with MK-886 (Merck, Rahway, NJ) at a dose of 4 microg per 100 mg of body-weight per day. At age 6 months the mice were sacrificed under anaesthesia. RESULTS: Measured by the en face method, the percentage of area occupied by lesions in aortas in the control group was 25.15 +/- 2.9%, whereas in the MK-886-treated group it was 11.16 +/- 0.7% (P < 0.05). Lesion area measured by cross-section of aortic roots was 455,494 +/- 29,564 microm(2) in the control group versus 263,042 +/- 20,736 microm(2) in the MK-886-treated group (P < 0.05). The MK-886 did not change the plasma cholesterol lipoprotein profile as compared with the control mice. Finally, we show that MK-886 may increase plaque stability by decreasing the macrophage content as well as increasing the collagen and smooth-muscle cell content. CONCLUSIONS: Our results show for the first time that inhibition of FLAP by MK-886 reduces development of atherosclerosis in gene-targeted apoE/LDLR-DKO mice.

Corn hybrid affects in vitro and in vivo measures of nutrient digestibility in dogs.

Corn is a commonly used ingredient in dry pet foods because there is a stable supply and it is a relatively inexpensive source of nutrients. Corn hybrids are available that are higher in CP and amylose and lower in phytate concentration than conventional hybrids. Approximately 500 mg of high-protein (HP), high-protein, low-phytate (HPLP), and high-amylose (HA) corn were compared with conventional (CONV) corn and amylomaize starch (AM) in triplicate and exposed to pepsin/hydrochloric acid and pancreatin to simulate hydrolytic digestion. Substrate remaining after this was used to determine in vitro colonic fermentation. Organic matter disappearances as a result of hydrolytic digestion were >80% for CONV, HP, and HPLP, whereas HA (60.7%) and AM (43.7%) were lower (P < 0.05). Total digestion (TD) values after hydrolytic digestion and 8 h of fermentation using canine fecal inoculum were greater (P < 0.05) for CONV, HP, and HPLP vs. HA and AM. The residue left after hydrolytic digestion of all substrates was poorly fermented. Five ileal-cannulated dogs were fed each corn hybrid at approximately 31% of the diet in a 5 x5 Latin square design. Dogs fed diets containing HP corn had higher (P < 0.05) ileal OM digestibility (70.3%) and tended (P < 0.10) to have higher DM digestibility (64.6%). Ileal starch digestibilities were lower (P < 0.05) for dogs fed HA (64.0%) and AM (63.0%). Ileal digestibilities of essential (71.2%), nonessential (67.4%), and total (69.0%) AA tended to be higher (P < 0.10) for HP diets compared with CONV (66.4, 62.4, and 64.0%, respectively). Total-tract DM, OM, CP, and GE digestibilities (77, 82, 77, and 84%, on average, respectively) were higher (P < 0.05) for dogs fed CONV, HP, and HPLP than for those fed AM (66.9, 71.6, 72.6, and 76.5%) and HA (60.6, 65.7, 69.7, and 71.5%). Total-tract fat digestibilities were lower (P < 0.05) for dogs fed HA diets (86.6%) than for all other treatments (91.0%, on average). Total-tract starch digestibilities were higher (P < 0.05) for dogs fed CONV, HP, and HPLP (98%, on average) compared with HA (72.8%) and AM (76.5%). No differences were detected among treatments in fecal bifidobacteria, lactobacilli, or Clostridium perfringens concentrations. The experiments demonstrated that HP and HPLP corn had hydrolytic digestion and fermentation characteristics similar to those of CONV corn, whereas HA resulted in similar responses to AM, a well-established resistant starch ingredient.

Inhibition of nuclear factor-kappaB attenuates artherosclerosis in apoE/LDLR - double knockout mice.

Nuclear factor - kappaB (NF-kappaB) is a good therapeutic target for cardiovascular disease and numerous efforts are being made to develop safe NF-kappaB inhibitors. Nowadays many authors address NF-kappaB as a major therapeutic target in atherosclerosis, especially for preventive measures, in the light of two main hypothesis of atherosclerosis: oxidation and inflammation. We hypothesized that ammonium pyrrolidinedithioocarbamate (PDTC) - a well-known inhibitor of NF-kappaB could inhibit the development of atherosclerosis in this experimental model. We used apoE/LDLR - DKO mouse model, which is considered as a one of the best models to study the anti-atherosclerotic effect of drugs. In this model PDTC inhibited atherogenesis, measured both by "en face" method (25,15+/-2,9% vs. 15,63+/-0,6%) and "cross-section" method (565867+/-39764 microm2 vs. 291695+/-30384 microm2). Moreover, PDTC did not change the profile of cholesterol and triglycerides in blood. To our knowledge, this is the first report that shows the effect of PDTC on atherogenesis in gene-targeted apoE/LDLR - double knockout mice.

Effect of curcumin on atherosclerosis in apoE/LDLR-double knockout mice.

It is widely appreciated that inflammation and oxidant stress contribute to atherogenesis. Curcumin, a polyphenolic natural compound has been reported to possess anti-inflammatory and anti-oxidant actions. We hypothesized that curcumin could inhibit the development of atherosclerosis in the apoE/LDLR-double knockout mice fed with Western diet (21% fat, 0.15% cholesterol w/w, without cholic acid). Curcumin (purity>or=98%), premixed with diet, was given for 4 months at a dose of 0.3 mg/ per day/ per mouse. In this model curcumin inhibited atherogenesis, measured both by "en face" method (25,15+/-2,9% vs. 19,2+/-0,6%, p<0,05) and "cross-section" method (565867+/-39764 microm2 vs. 299201+/-20373 microm2, p<0,05). Importantly, curcumin influenced neither the concentrations of cholesterol and triglycerides in blood nor animal body weight. To our knowledge, this is the first report that shows the anti-atherogenic effect of low dose of curcumin in fine model of atherosclerosis: gene-targeted apoE/LDLR-double knockout mice.

Soliton trains in Bose-Fermi mixtures.

We theoretically consider the formation of bright solitons in a mixture of Bose and Fermi degenerate gases. While we assume the forces between atoms in a pure Bose component to be effectively repulsive, their character can be changed from repulsive to attractive in the presence of fermions provided the Bose and Fermi gases attract each other strongly enough. In such a regime the Bose component becomes a gas of effectively attractive atoms. Hence, generating bright solitons in the bosonic gas is possible. Indeed, after a sudden increase of the strength of attraction between bosons and fermions (realized by using a Feshbach resonance technique or by firm radial squeezing of both samples) soliton trains appear in the Bose-Fermi mixture.

Multimode dynamics of a coupled ultracold atomic-molecular system.

We analyze the coherent multimode dynamics of a system of coupled atomic and molecular Bose gases. Starting from an atomic Bose-Einstein condensate with a small thermal component, we observe a complete depletion of the atomic and molecular condensate modes on a short time scale due to a significant population of excited states. Giant coherent oscillations between the two condensates for typical parameters are almost completely suppressed. Our results cast serious doubts on the common use of the 2-mode model for the description of coupled ultracold atomic-molecular systems and should be considered when planning future experiments with ultracold molecules.

[Adult multilocular rhabdomyoma as etiology of a space-occupying lesion in the area of the neck].

Real rhabdomyomas belong to the rarest tumors in the human body. The origin of these benign neoplasms is in the cardiac or skeletal muscle, in the beginning, there are no significant symptoms and the tumor grows very slowly. Extracardiac rhabdomyomas seldom occur and are mostly located in the head and neck region. A multilocular occurrence represents a rarity. We describe a 77-year-old female patient with a multilocular adult rhabdomyoma in the parapharyngeal and paratracheal space. The tumor was accidentally found; the patient had not complained of any specific symptoms. After histological examination, complete resection was carried out. On discharge the patient showed no functional deficits. Long-term follow-up is necessary because local recurrences have been described.

Urine cytology in patients with urinary tract calculosis.

Urinary cytological examination was performed on 375 patients suffering from urolithiasis at the Department of Urology. University School of Medical Sciences, Bydgoszcz between 01.01.1991 and 31.12.1992. 189 were female (mean age: 47.5 years), 186 were male (mean age: 53.5 years). We found dysplastic cells in urinary sediments in 25.3% of patients before therapy and in 5.3% of patients after conservative or traditional operation and interventional therapy (ESWL. PCNL). Degenerated cells were found in 72% of patients before therapy and in 30.4% after therapy only. In our group neoplasmatical cells were not found. Inflammatory background was found in 44.5% before and in 21.9% after therapy, pyuria in 19.7% and 0.8% respectively. In urinary sediments we found concrements in 24% of patients before and 7.7% after therapy. However, 20 (51.3%) of 39 patients after interventional therapy (ESWL, PCNL) had concrements in urinary sediments. In conclusion, no abnormality was seen in 85.1% before therapy and in 31.2% after therapy. The significance of cytological examination in management of calculous diseases was discussed.

Functional implications of extensive new data on the innervation of pulmonary neuroepithelial bodies.

Pulmonary neuroepithelial bodies (NEBs) are innervated organoid groups of neuroendocrine cells, present in the epithelial lining of intrapulmonary airways of man and all air-breathing vertebrates studied so far. NEBs receive a vagal nodose sensory innervation that is considered by other authors as their main, if not their only innervation, although apparently not needed for the normal development and maintenance of NEBs. In the present study of NEBs in the developing rat lung (gestational day 16 - adult), immunoreactivity (IR) for calcitonin gene-related peptide (CGRP; a marker for NEBs), protein gene product 9.5 (PGP9.5; a marker for NEBs and neuronal elements), calbindin D28k (CB; a calcium binding protein), and for the growth-associated protein 43 (GAP-43; a marker for growing or remodelling nerve fibers) was combined with vagal denervation experiments. GAP-43 and CB IR revealed that the vagal sensory innervation of airways precedes the prenatal development of NEBs by several generations of branching. Unlike several other nerve fiber populations innervating NEBs, the vagal sensory component apparently does not express GAP-43 IR in mature lungs. GAP-43 labelling, however, did reveal newly ingrowing intraepithelial vagal sensory fibers, specifically reinnervating NEBs in adult rats 2 weeks following a cervical vagal crush. In conclusion, details of the innervation of pulmonary NEBs were disclosed that had been invisible so far, shedding new light on its complexity and probable involvement in the normal development and functional maintenance of NEBs throughout life.

[Diagnosis and treatment of necrotizing fasciitis of the head and neck region].

BACKGROUND: Necrotizing fasciitis of the head and neck is a rare occurrence. It spreads rapidly along fascial planes causing extensive necrosis; it often results in gangrene of the overlying skin. Mostly involved are immunodeficient patients with banal infections of the upper aerodigestive tract, small traumas, but also after surgical procedures. The "Federal Health Agency" of the Federal Republic of Germany estimates a number of 40 cases a year in Germany for all regions of the body. The mortality is about 20%-50% dependent on the localisation of this soft tissue infection. PATIENTS: Four cases of necrotizing fasciitis of the head and neck region, which were treated at the ENT-Department of the Martin Luther University Halle-Wittenberg since 1995, were described. RESULTS: Despite aggressive surgical debridement and intensive care medicine two patients died because of streptococcus-associated-toxic-shock-syndrome. In all patients we found a diabetes mellitus as a known risk factor for this necrotizing soft tissue disease. In two of four patients with cervical involvement mediastinitis was diagnosed as a complication. The CT-scan showed this mediastinal spread only in one case. CONCLUSIONS: Only rapid diagnosis and surgical treatment with radical debridement can influence the disease positively. Thoracal CT-scan is necessary in all cases of cervical necrotizing fasciitis because of the high risk of mediastinal involvement. A strong complication is a streptococcus-associated-toxic-shock-syndrome, which should be prevented because it is often associated with a lethal outcome. Penicillin G and clindamycin are advocated for antibiotic treatment.

Development of the innervation of long bones: expression of the growth-associated protein 43.

It has been known from clinical and experimental observations that the peripheral nervous system is involved in the development of long bones. Expression of growth-associated protein 43 (GAP-43/B-50) was found in axonal growth cones during embryonic and postnatal ontogeny as well as in regenerating axons after nerve injury. The aim of the present study was to examine the occurrence of growing nerve fibers in rat tibia from gestational day 16 (GD 16) to postnatal day 28 (PD28). An indirect immunoenzymatic reaction using antibodies raised against GAP-43 was applied to detect outgrowing nerve fibers penetrating into the developing bone. On GD 16 and GD 17 no GAP-43-immunoreactive (IR) fibers were observed in the close vicinity of bone rudiments. On GD19 GAP-43-IR fibers were scarcely present within the periosteum of the central portion of the diaphysis. In the perichondrium surrounding the proximal epiphysis, nerve fibers were first detected around birth. From PD1 onward, numerous fibers were seen in the fibrous buds of the perichondrium at the epi-metaphyseal junction (Ranvier's grooves), some of them being adjacent to the blood vessels. Nerve fibers penetrating into the bone and located in the bone marrow, predominantly associated with blood vessels, were first observed on GD21 and their number increased with further development. They were initially located in the central portion of the diaphysis and later extended towards the metaphyses. On PD4 an increased number of GAP-43-IR fibers appeared in the perichondrium of proximal and distal epiphyses. In the fibrous strands penetrating into the epiphyses and in the secondary ossification centers, nerve fibers were first observed on PD10. From PD14 onward the pattern of tibial innervation remained unchanged but the intensity of GAP-43 immunostaining visibly decreased. The present study demonstrates that developing long bones of rat hindlimbs are supplied by growing nerve fibers immunoreactive for GAP-43 from GD 19 onward. Time and location of their appearance were at least partially correlated with known events taking place during long bone development, e.g. formation of primary and secondary ossification centers. Decreased expression of GAP-43 immunoreactivity in later developmental stages is believed to reflect nerve fiber maturation.

Endometriosis of the urinary bladder.

Although every seventh or eighth case of endometriosis is found to have urinary involvement, this type of endometriosis is still rarely recognized. The symptoms are not specific and are frequently suggestive of a neoplastic process. On the basis of our two cases of histopathologically two types of endometriosis as well as literature review the practical significance of endometriosis in urology is described. In each patient who previously underwent a gynaecological or surgical procedure in the abdominal cavity, endometriosis of the bladder is likely to occur. This possibility should not be ruled out, otherwise it could lead to misdiagnosis and inappropriate treatment.

[Diagnostic value of cytologic examination of urine sediment in patients with non-neoplastic diseases].

The results are presented of urine sediment cytological examination of all patients treated in the Department of Urology for non-neoplastic diseases from March 01, 1989 to October 01, 1991. Out of the total number of 936 newly admitted patients in 88 cases the urine cytological examination demonstrated dysplastic cells which accounted for 9.4%, while in 11 cases cells suspected of malignancy were found which accounted for 1.2%. These patients are under constant urological control.

Diagnostic value of urine cytology and urine carcinoembryonic antigen (CEA) level in the distinction between bladder cancer and urinary tract infection.

In the retrospective investigation we estimated the CEA levels in urine and urine cytology in 293 patients with urinary bladder cancer (group I) and in 129 patients with urinary tract infection (group II). An increased CEA level was found in 212 cases of bladder cancer and in 109 cases of urinary infection. Neoplastic cells in urine sediment were identified in 254 cases in the first group, while in the second, multiple inflammatory cells and pyuria were diagnosed in 55 and 56 cases, respectively. The combined examination (increased CEA level and/or positive cytology of the urine sediment) allowed for the diagnosis of neoplastic disease in 270 patients in the first group and infection diseases in 123 patients in the second group. After surgical therapy an increased urine CEA level and neoplastic cells in the urine cytology were found in 1 case which was then confirmed histopathologically. In the group with urinary infection an increased CEA level in urine, after antiphlogistic therapy, was found in 5 cases, inflammatory cells in 12 cases, pyuria in 8 cases and suspect cells were found in 6 cases in the group with urinary infection. In specimens from these cases, after electroresection, bladder cancer was diagnosed in 2, and cystitis cystica type changes in 2 cases.
