Relationship between pharmacological induction of estrous and/or ovulation and twin pregnancy in the Thoroughbred mares.

The aim of this study was to evaluate the possible relationship of pharmacological induction of estrous and/or ovulation with the occurrence of twin pregnancies in Thoroughbred mares. Out of 680 mares, 356 received one of the following treatments during the estrous cycle in which they became pregnant: injection of 0.5mg of cloprostenol at the ultrasonographic detection of a CL (n=86); injection of 5000 IU human chorionic gonadotropin (hCG) immediately before mating (n=221); injection of 0.5mg of cloprostenol at the ultrasonographic detection of a CL plus injection of 5000 IU hCG immediately before mating on cloprostenol-induced estrous (n=49). The other 324 mares, not treated for induction of estrous or ovulation in the estrous cycle resulting in pregnancy, were used as control group. The occurrence of twin and single pregnancies in treated and control mares underlines that the percentage of twin pregnancy in treated mares (16.6%) was statistically significantly higher (P<0.0001; odds ratio, OR=2.87) than the percentage of twinning in the control group (6.5%). Comparison of the occurrence of twins between treatments revealed a statistically significant difference between mares treated with hCG alone compared to animals given prostaglandin F2alpha (PGF2alpha) plus hCG. The results show a statistically significant difference for each treatment compared to controls, with the least difference (P<0.05; OR=2.18) for the comparison between hCG treatment group and controls, a significance of P<0.01; OR=3.05 for the comparison between PGF2alpha treatment and controls, and a highly statistically significant difference (P<0.0001; OR=6.37) for the comparison between PGF2alpha plus hCG-treated animals and controls.

Cytochemical analysis of peripheral blood mononuclear cells following allogeneic bone marrow transplantation: correlation of hydrolase expression with graft-versus-host disease.

Peripheral blood mononuclear cells of 21 patients undergoing allogeneic bone marrow transplantation (BMT) were monitored post-BMT for immunologic markers (E rosettes OKT3, OKT8, DR, and BT5/9, a monoclonal antibody which stains helper T cells), cytochemical markers (acid phosphatase [AP], beta-glucuronidase [BGLU], and acid alpha naphthyl acetate esterase [ANAE] ), and morphology. The cytochemical T score, was obtained from typical AP, BGLU, and ANAE reactivity. The same was done for the cytochemical non-T score and macrophage (Mo) score. All patients received cyclosporine A (CyA) for graft-v-host disease (GvHD) prophylaxis. In univariate analysis there was no significant correlation between the proportion of E rosettes, OKT3-, OKT8-, DR-, and BT5/9-positive cells, and GvHD. The first three showed instead a positive correlation with time from transplant: E rosettes (P = .02), OKT3 (P = .01), and OKT8 (P = .003). In contrast, a significant negative correlation was found in univariate analysis, between the cytochemical T score and GvHD (P = .0001), and a positive correlation between non-T score and GvHD (P = .0008), as well as between the Mo score and GvHD (P = .03). There was no influence of time from transplant on the T (P = .8), non-T (P = .8), or Mo score (P = .4). In multivariate analysis comparing E rosettes, OKT3, T score, non-T score, GvHD, and time from BMT, the only variable associated with GvHD was the T score (P less than .05). These results suggest that T cell activation during GvHD is associated with a loss of hydrolase expression in T cells, but does not imply relevant modifications of immunologic surface markers. In addition, lysosomal enzymes appear early (before day 10) after transplantation, indicating that T cells at this stage are well differentiated.

Expression of three immunoglobulin isotypes by individual B cells during development: implications for heavy chain switching.

Previous studies in mice and humans have shown that the first IgA+ and IgG+ B cells appearing during ontogeny also bear IgM on the surface. The acquisition of IgD seems to occur at a later stage in differentiation. In this study we have combined autoradiography with two-color immunofluorescence to directly detect human B cells expressing three surface isotypes. We report that the vast majority of IgA+ cells in the neonate also bear IgM and IgD; the same holds true for IgG+ cells. This phenotype is peculiar of newborns, while in the adult the majority of IgA+ and IgG+ cells are single. We discuss the genetic implications of such a finding for heavy chain switching.

A comparative study between visible-light-activated and autopolymerizing sealants in relation to retention.

This investigation was conducted with the object of comparing the retention capacity of two visible-light-activated sealants (Concise-3M and Sealite-Kerr) with the more commonly used autopolymerizing sealant (Delton-Johnson & Johnson). The study included sixty-two children, thirty-five boys and twenty-seven girls, between six and eleven years of age. Two hundred twenty-nine first permanent molar occlusal surfaces were treated with sealants (118 maxillary molars and 111 mandibular molars). Sealants were applied according to the manufactures' instructions and cotton rolls were used for partial isolation. After six, twelve, eighteen and twenty-four months, the surfaces treated with the sealants were examined and evaluated. Three categories were used to evaluate each sealant. During each evaluation period the sealants were graded according to complete, partial, or failed retention. No significant difference was noted between Concise and Delton sealants for C when comparing maxillary and mandibular teeth. Significant differences were observed between Concise and Sealite in relation to C on the maxillary and mandibular arches and also between Sealite and Delton on the maxillary arch. There were no significant differences in retention between the maxillary and mandibular teeth, when the same type of sealant was used.
