Benign perineal masses in the male are rare. To date 11 cases of benign tumors of the male perineum have been reported. Two unusual cases, both benign tumor masses, are reported. A brief review of the pertinent pathology is included. No similar encapsulated lesions have been previously reported.
The value of induced diuresis, employing furosemide, on blood loss during and after prostatectomy is evaluated. Two series of 100 patients (consecutive) undergoing prostatectomy, one series with furosemide and one without, are compared. The conclusion drawn from critical analysis is that induced diuresis with furosemide is beneficial in reducing blood loss.
An unusually rare case of chronic hematuria, secondary to an intraparenchymal renal aneurysm is presented. The triad of normal blood pressure, intrarenal aneurysm, and hematuria can be treated conservatively by partial nephrectomy with good results. This treatment is recommended since in a significant percentage of patients with these lesions spontaneous rupture can occur, which is a life-threatening disaster.

Because human casein does not precipitate from milk at its isoelectric point as does bovine casein, there is no easy method of quantitation. Casein represents only approximately 30% of the protein fraction in human milk, and the complex methods necessary for isolation cannot be used easily with small samples in a survey of a large number of mothers. Two-dimensional electrophoresis coupled with computer imaging has the potential to compare and quantitate proteins expeditiously using a small sample size. IsoDalt, a denaturing methodology, separates the casein micelle into its component parts, beta-casein, kappa-casein, parakappa-casein and casomorphins. Identification of these spots was made by immunoassay of a Western blot with monoclonal anti-human casein. Two spots at 24 kDa and 26 kDa, thought to be phosphorylated isomers of beta casein, were selected for quantitation. Milk samples from 20 mothers, 8 weeks post partum, were run on two-dimensional (2-D) gels; a slide was taken of each silverstained gel with a Kodak control strip; the slide was scanned into powerMac Photoshop 3 with a Polaroid-Sprintscan; spots were isolated using "threshold", "mask" with IPTK (Imaging Processing Tool Kit, Reindeer Games) a Photoshop plug-in, and transferred to the NIH-Image program. Using an NIH-Image gel macro (Thomas Seebacher), the area and integrated density of the spots were measured. The Kodak control scale provided calibration and conversion to OD units. Visual scanning of the gels and computer units indicated a wide range of concentrations. To understand the range in units of weight, a standard was generated using bovine alpha casein (Sigma). Measurements will be used in a statistical program, Statview (Abacus), in an attempt to correlate information from a questionaire with casein concentration.
Two-dimensional (2-D; Iso Dalt method) electrophoresis was done on lipid membrane fractions from human milk samples collected in polypropylene tubes at home by the mother. The samples represent milk from two weeks to ten months post partum. Identification was made by immunoassay of Western blots. Apolipoprotein E was not easily seen on silver-stained gels because concentration is small and spots overlap another protein. Blot assays show a more complex pattern than 34 kDa plasma apo E with molecular size isoforms at 36 kDa, 34 kDa, 20 kDa and 15 kDa. One sample was negative for apo E. Apo E plays an important role in lipid transport in tissue and in the brain. It is synthesized in a variety of tissues by specific cells. Milk may reflect several sources.

The homologue of the viral Kirsten ras (v-Ki-ras) gene found in the human lung carcinoma cell line, Calu-1, has an intron-exon structure similar to that of the human homologue of the viral Harvey ras (v-Ha-ras) gene. A second, potential fourth coding exon is present in the human Ki-ras gene and similar sequences are found in the Kirsten murine sarcoma virus. Cysteine is encoded at the twelfth amino acid position, suggesting that the Calu-1 Ki-ras gene has undergone a mutational activation at the same position as the human Ha-ras gene of the bladder carcinoma cell line, T24. A comparison of their predicted amino acid sequences suggests that ras proteins have a 'constant' region and a 'variable' region. Here we propose a common modular structure for ras gene products in which the variable region forms a physiologically important combining site.
