Assessment of immunomodulating activity of ofloxacin and three other quinolones in human plasma.

A time-dependent degree of immunosuppression or immunostimulation which may be correlated to pharmacokinetic variables was obtained by administering a single dose of ofloxacin, norfloxacin, pipemidic and piromidic acid to healthy volunteers. The plasma samples collected before drug administration and then at various time intervals were tested for their immunomodifying activity by employing a modified 2-way mixed lymphocyte reaction and a PMN chemotactic assay. Our results show that some of the quinolones tested--norfloxacin, pipemidic acid, piromidic acid--have an immunosuppressive activity.

Evaluation of official instrumental methods for the determination of particulate matter contamination in large volume parenteral solutions.

The distribution pattern of particle contamination in nine different types of LV parenteral solutions and the possibility of correlating the counts made with two official instruments (Coulter Counter and HIAC) were studied. Two hundred containers of LV parenteral solutions (corresponding to 40 batches) produced in Italy, were sampled. Each bottle was submitted to HIAC and Coulter Counter countings, for particle sizes ranging between 2 and 25 micron. For about 50% of the products, the two straight lines that represent the distribution of particle contamination obtained with the two methods did not cross-over within the studied size range, the Coulter Counter counts always proving higher than the HIAC ones. In the other cases, the cross-over point of the two lines occurred at varying size levels. Statistical analysis of the results pointed to a relationship between the contamination values obtained with the two counting methods for sizes ranging between 2 and 5 micron, but there was no correlation for sizes equal to, or higher than, 10 micron. From the maximum contamination levels established by the BP and the FU IX for the HIAC method, the corresponding values were calculated for the Coulter Counter method. Similarly the values were calculated the HIAC method based on the maximum values set for the Coulter Counter.

Activity of human serum lipoproteins on the infectivity of rhabdoviruses.

The inhibiting activity of various human serum lipoprotein classes and their lipid components on the infectivity of rhabdoviruses has been studied. The research has been carried out according to different experimental procedures on both the vesicular stomatitis virus (Indiana strain), and the fixed rabies virus (CVS strain). The results obtained have shown an inhibition of the infectivity of VSV and CVS, mainly linked to the very low and low density lipoprotein classes. It has also been demonstrated that the inhibitors act directly on viruses causing a decrease in their attachment to host cells.

Preliminary study on fixed rabies virus strain "Roma". I: Adaptation on BHK21/S13 cells.

In this study the attempts to adapt Roma fixed rabies virus to BHK21/S13 cells and human diploid fibroblasts, for a better characterization of the virus, are reported. BHK21 cells were infected with the virus and maintained in culture for more than 100 passages. A persistently infected culture by rabies virus was obtained. Attempts to propagate virus in human diploid fibroblasts were unsuccessful.

[Prevention of ischemic heart disease: a feasibility study of health education in the primary school].

To verify the level of participation of children and teachers in an educational program, the reliability of the program itself and to determine whether children can modify family's behavioral patterns, a feasibility study was undertaken among the pupils of a primary school. Seventy-two children, belonging to different socio-economic classes, were studied. Children were given a questionnaire before and after the course, which was held by previously trained teachers. Very good participation of children and teachers in the program was observed. On the contrary very low involvement of the parents was achieved. This experience emphasizes the difficulties arising when actions to modify adult's life styles are attempted. On the contrary, a standardized education program, lasting for a whole school year, could be a feasible, useful and not too costly program not only for the primary prevention of coronary artery disease, but for any program of extensive health care.
