152Eu depth profiles in granite and concrete cores exposed to the Hiroshima atomic bomb.

Two granite and two concrete core samples were obtained within 500 m from the hypocenter of the Hiroshima atomic bomb, and the depth profile of 152Eu was measured to evaluate the incident neutron spectrum. The granite cores were obtained from a pillar of the Motoyasu Bridge located 101 m from the hypocenter and from a granite rock in the Shirakami Shrine (379 m); the concrete cores were obtained from a gate in the Gokoku Shrine (398 m) and from a pillar top of the Hiroshima bank (250 m). The profiles of the specific activities of the cores were measured to a depth of 40 cm from the surface using low background germanium (Ge) spectrometers. According to the measured depth profiles, relaxation lengths of incident neutrons were derived as 13.6 cm for Motoyasu Bridge pillar (granite), 12.2 cm for Shirakami Shrine core (granite), and 9.6 cm for concrete cores of Gokoku Shrine and Hiroshima Bank. In addition, a comparison of the granite cores in Hiroshima showed good agreement with Nagasaki data. Present results indicates that the depth profile of 152Eu reflects incident neutrons not so high but in the epithermal region.

Prevalence of goiter and urinary iodine excretion levels in children around Chernobyl.

The prevalence of goiter among children living in areas affected by the Chernobyl accident was investigated by analysis of data on approximately 120,000 children examined at five medical diagnostic centers in Belarus, Russia, and the Ukraine. Examinations of thyroid gland were conducted with an arch-automatic ultrasonographic instrument at the five centers under the same protocol. The diagnosis of goiter was established when the thyroid volume exceeded a limit calculated from age, height, and body weight of a child. A considerable variation by region was noted in the prevalence of goiter. Highest in the Kiev region, the prevalence in the five regions was 54% in Kiev, 38% in the Zhitomir regions of the Ukraine, 18% in Gomel, 22% in the Mogilev regions of Belarus, and 41% in the Bryansk region of Russia. Urinary iodine content was measured in approximately 5700 children, and an endemic iodine deficient zone was confirmed in the Bryansk, Kiev, and Zhitomir regions. A significant negative correlation was observed between the prevalence of goiter and the median level of urinary iodine content (Spearman's rank correlation coefficient was -0.35, P = 0.025).

Distinct difference in relative biological effectiveness of 252Cf neutrons for the induction of mitotic crossing over and intragenic reversion of the white-ivory allele in Drosophila melanogaster.

The relative biological effectiveness (RBE) of 252Cf neutrons was determined for two different types of somatic mutations, i.e., loss heterozygosity for wing-hair mutations and reversion of the mutant white-ivory eye-color, in Drosophila melanogaster. Loss of heterozygosity for wing-hair mutations results predominantly from mitotic crossing over induced in wing anlage cells of larvae, while the reverse mutation of eye color is due to an intragenic structural change in the white locus on the X-chromosome. For a quantitative comparison of RBE values for these events, we have constructed a combined mutation assay system so that induced mutant wing-hair clones as well as revertant eye-color clones can be detected simultaneously in the same individuals. Larvae were irradiated at the age of 80 +/- 4 h post-oviposition with 252Cf neutrons or 137Cs gamma-rays, and male adult flies were examined under the microscope for the presence of the two types of clonal mosaic spots appearing. The induction of wing-hair spots per dose unit was much greater for 252Cf neutrons than for 137Cs gamma-rays, whereas the frequencies of eye-color reversion were similar for neutrons and gamma-rays. The estimated RBE values of neutrons were 8.5 and 1.2 for the induction of mutant wing-hair spots and revertant eye-color spots, respectively. These results indicate that the RBE of neutrons is much greater for mitotic crossing over in comparison to the intragenic white-ivory reversion events. Possible causes for the difference in RBE are discussed.

Regulation of mitochondrial pyruvate dehydrogenase activity by tau protein kinase I/glycogen synthase kinase 3beta in brain.

According to the amyloid hypothesis for the pathogenesis of Alzheimer disease, beta-amyloid peptide (betaA) directly affects neurons, leading to neurodegeneration and tau phosphorylation. In rat hippocampal culture, betaA exposure activates tau protein kinase I/glycogen synthase kinase 3beta (TPKI/GSK-3beta), which phosphorylates tau protein into Alzheimer disease-like forms, resulting in neuronal death. To elucidate the mechanism of betaA-induced neuronal death, we searched for substrates of TPKI/GSK-3beta in a two-hybrid system and identified pyruvate dehydrogenase (PDH), which converts pyruvate to acetyl-CoA in mitochondria. PDH was phosphorylated and inactivated by TPKI/GSK-3beta in vitro and also in betaA-treated hippocampal cultures, resulting in mitochondrial dysfunction, which would contribute to neuronal death. In cholinergic neurons, betaA impaired acetylcholine synthesis without affecting choline acetyltransferase activity, which suggests that PDH is inactivated by betaA-induced TPKI/GSK-3beta. Thus, TPKI/GSK-3beta regulates PDH and participates in energy metabolism and acetylcholine synthesis. These results suggest that TPKI/GSK-3beta plays a key role in the pathogenesis of Alzheimer disease.

Identification and synthetic pathway of sialyl-Lewis(x)-containing neolacto-series gangliosides in lens tissues. 2. Enzymatic synthesis of sialyl-Lewis(x) gangliosides in monkey and rat lenses.

In Japanese monkey lenses, 3H-labeled fucose and N-acetylneuraminic acid were enzymatically transferred to neolactotetraosylceramide (nLc4) and III 3 FucnLc4, respectively, suggesting the presence of a synthetic pathway of IV3 NeuAcIII3 FucnLc4 via III3 FucnLc4 in monkey lenses. Six rat strains, Wistar, Sprague-Dawley and pigmented strains, contained sialyl-Lewis(x) gangliosides in non-cataractous lenses in a strain-specific manner. Glycosyltransferase assay revealed that the transfer of 3H-labeled fucose to nLc4 occurred in all the strains, but that the transfer of 3H-labeled N-acetylneuraminic acid to III3 FucnLc4 was strain-specific. These results suggested that sialyl-Lewis(x) gangliosides were generally synthesized from neolactotetraosylceramide via Lewis(x) glycolipid (III3 FucnLc4) in lens tissues, differing from other tissues. Combining our results, we propose two synthetic pathways of sialyl-Le(x)- containing neolacto-series gangliosides and A-pathway ganglio-series gangliosides in human senile cataractous lens: one to sialyl-Lewis(x) gangliosides from nLc4 via Lewis(x) glycolipid, and the other to GD1a from GM3, via GM2 and GM1.

Exposure of rat hippocampal neurons to amyloid beta peptide (25-35) induces the inactivation of phosphatidyl inositol-3 kinase and the activation of tau protein kinase I/glycogen synthase kinase-3 beta.

Exposure of rat hippocampal neurons to the peptide amyloid beta (A beta) (25-35) as well as A beta (1-40) peptides enhances phosphorylation of tau to a paired helical filament (PHF)-state through activation of tau protein kinase I (TPK I)/glycogen synthase kinase-3 beta (GSK-3 beta) [Busciglio, J., Lorenzo, A., Yeh, J. and Yankner, B.A., Neuron, 14 (1995) 879-888; Takashima, A., Ishiguro, K., Noguchi, K., Michel, G., Hoshi, M., Sato, K., Takahashi, M., Hoshino, T., Uchida, T. and Imahori, K., Neurosci. Meeting Abstr., 671 (1995) 17]. In order to examine the effects of A beta treatment on intracellular signaling mechanism, we have investigated the role of phosphatidyl inositol-3 (PI-3) kinase in the phosphorylation of tau. A beta (25-35) exposure induced an inactivation of PI-3 kinase and an activation of TPK I/GSK-3 beta in rat hippocampal culture. Wortmannin, an inhibitor of PI-3 kinase, also activated TPK I/GSK-3 beta, leading to an enhancement of tau phosphorylation and neuronal death in hippocampal culture. These results suggest that A beta (25-35) inhibition of PI-3 kinase results in the activation of TPK I/GSK-3 beta, the phosphorylation of tau, and resultant neuronal death in rat hippocampal neurons.

Histopathological characteristics of childhood thyroid cancer in Gomel, Belarus.

We reviewed histopathologically 19 cases of childhood thyroid cancer occurring between 1991 and 1994 among 14,396 screening subjects in Gomel, Republic of Belarus, the region most severely radio-contaminated by the Chernobyl nuclear power plant accident in 1986. The patients were 13 girls and 6 boys with a mean age of 10.6 years. The mean age at the time of the accident was 3.2 years. Mean tumor diameter was 16 mm, and all cases were papillary carcinoma with various amounts of solid component. Psammoma bodies and stromal fibrosis were encountered to some extent in almost all cases. The tumors were highly prone to local invasion and regional lymph-node metastasis. No morphological evidence for radiation-induced cancer was obtained in these cases. 137Cs levels were relatively high in the patients' bodies and in the soil at the places of domicile. However, there was no dose-response relationship between cancer prevalence and radioactivity. These facts suggest that the incidence of aggressive pediatric thyroid cancer is extremely high in Gomel, where most of the children were exposed to a low level of radioactivity over a long time after the accident. At present, however, no definite conclusion can be drawn on the relationship between cancer occurrence and radioactive contamination.

Structure and function of asterosaps, sperm-activating peptides from the jelly coat of starfish eggs.

Jelly coat of starfish eggs has the capacity to activate homologous spermatozoa and induce the acrosome reaction. We have isolated 12 sperm-activating peptides (SAPs) from the egg jelly of the starfish, Asterias amurensis. Eleven SAPs were structurally identified by sequence analysis and electro-spray ionisation mass spectrometry. All of them are glutamine-rich tetratriacontapeptides with an intramolecular disulphide linkage between Cys8 and Cys32. They are much larger than sea urchin SAPs and do not show any significant sequence similarities to known proteins. Thus we have collectively named them asterosaps. The amino terminal region, where structural diversity of asterosaps is observed, is not important for their activity, whereas the disulphide linkage is essential. Asterosaps do not induce the acrosome reaction by themselves, but are able to induce the acrosome reaction in combination with an egg jelly glycoconjugate named ARIS. Furthermore, anti-asterosap rabbit antibody significantly decreased the acrosome reaction-inducing activity of the jelly solution and the activity was restored by addition of excess asterosap. These results support our hypothesis that the main physiological role of SAPs is the induction of the acrosome reaction in cooperation with two other jelly components, ARIS and Co-ARIS.

Determination of the relative neutron sensitivity of a C-CO2 ionization chamber.

The neutron sensitivity of a C-CO2 ionization chamber has been experimentally determined in the neutron energy range of 0.1-1.2 MeV. Eleven kU data were obtained which ranged from 0.04 to 0.1 as a function of neutron energy. There were two peaks in the spectrum of the kU factor, and their positions corresponded to the resonance energies of the kerma factors of carbon dioxide, of which the energies are about 0.4 and 1 MeV. The present data agree with the data obtained at the National Institute for Radiological Sciences in Chiba, Japan (NIRS). The energy dependences of the sensitivity were calculated by using the kerma and the W-value ratios of gas to soft tissue for comparison. The experimental data were reproduced well by the calculations.

Cloning and sequencing of mouse complementary DNA for heme oxygenase-2.

A mouse cDNA encoding a human homologue of heme oxygenase-2 (HO-2) was isolated. The deduced protein contains 315 amino acids and has a calculated molecular mass of 35.8 kDa. The nucleotide sequence is 85.6% identical and the amino acid sequence 87% identical to those of the human protein. The corresponding mRNA is present in brain and testis, but not in ovary, kidney, liver, or spleen.

Description of new composite tissue transfer for salvage of a complex hand defect.

Leiomyosarcoma of the soft tissue involving bones is an extremely rare tumor. In the past, a hand tumor of this type required amputation. Presently, with improved chemotherapy and reconstruction, the hand and its dexterity sometimes can be saved. In this case, after chemotherapy, the authors opted for wide excision including the skin, tendons, muscles, and bones. The large defect was reconstructed with an iliac crest autograft and a dorsalis pedis free flap including tendons. Functional result was excellent in this patient. The pathologic results also were reviewed by histology, immunohistochemistry for muscle marker, and electron microscopy.

Estimation of radiation doses for atomic-bomb survivors in the Hiroshima University Registry.

The present study presents the Hiroshima University Registry of atomic bomb survivors, of which the total number is about 270,000, and application of absorbed doses. From this registry, we picked up 49,102 survivors and applied organ doses based on the dosimetry system 1986 (DS86), which is named the Atomic Bomb Survivor 1993 Dose (ABS93D). The applied dose data are based on the tables listed in the DS86 final report such as the free-in-air kermas, the house shielding factors, and organ dose factors for the active bone marrow and the breast. Calculations for the 13 other organs provided in DS86 are possible. To obtain the organ doses for each survivor, it is necessary to obtain information concerning (1) place exposed, (2) whether they were shielded or not, and (3) age. ABS93D body transmission factors for active bone marrow for neutrons and gamma rays agreed with DS86 to within a few percent. Of the survivors studied, 35,123 of them were used for the relative risk estimation of leukemia mortality, adopting the same method as the Radiation Effects Research Foundation (RERF) for comparison. For the observation period from 1968 to 1989, the analyzed relative risks for leukemia mortality at 1 Gy by shielded kerma and by active bone marrow dose are 2.01 and 2.37, respectively, which are consistent with the RERF results.

137Cs concentration in soil samples from an early survey of Hiroshima atomic bomb and cumulative dose estimation from the fallout.

Low background gamma-ray measurement has been performed to determine the 137Cs content in soil samples collected in a very early survey of the Hiroshima atomic bomb. These soil samples were collected just 3 d after the explosion within 5 km from the hypocenter and were not exposed to the global fallout from nuclear weapon tests. Out of 22 samples, 137Cs was detected for 11 samples, and their radioactivities ranged from 0.16-10.6 mBq g-1 at the time of the measurement. A comparison of the 137Cs deposition with the rainfall area within Hiroshima city indicates that the rainfall area was wider than the previously proposed one. Cumulative exposure by the fallout has been estimated to be 0.31 mC kg-1 (0.12 R) in Hiroshima city except for the heavy fallout area and at most 1.0 mC kg-1 (4 R) in the heavy fallout area.

Dosimetry of mixed neutron and gamma radiation with paired Fricke solutions in light and heavy water.

Paired Fricke solutions, made up from light water or heavy water and 0.8N in H2SO4 and 1 mM in Fe(NH4)2(SO4)2 and NaCl, were calibrated with 60Co gamma rays and with mixed neutron and gamma radiation from a 252Cf source. Absorbance increases, AL and AH, in light- and heavy-water Fricke dosimeters, respectively, increased with fast-neutron and gamma-ray tissue doses, Dn (GY) and D gamma (GY), of the mixed radiation as follows: AL = 0.00178Dn + 0.00371D gamma; AH = 0.00121Dn + 0.00442 D gamma. G-values of 7.2 and 5.5 were obtained for 252Cf neutrons in light- and heavy-water Fricke dosimeters, respectively. When we applied the pair of equations to AL and AH values observed after exposure to mixed radiation in a nuclear reactor, resulting Dn and D gamma values agreed within 10% to doses measured with paired ionization chambers. Doses required for Fricke dosimeters were 5 Gy or more. In contrast, we found that micronuclear yields in onion roots can measure the neutron component of mixed radiation fields at the order of 10 cGy with reasonable accuracy even if the neutron to gamma-ray dose ratio is unknown.
