Electrochemical behaviour of droxicam: kinetic study in aqueous-organic media.

A kinetic study of the hydrolytic decomposition of droxicam was carried out in order to establish the possible pharmacological action of the drug in the organism of the human being. The electrochemical technique used was scan cyclic voltammetry on a HMDE, giving a well-defined peak. Due to the insolubility of droxicam in water, the working media was a methanol-H(2)O (4:96, V/V) mixture. The decomposition product of droxicam is piroxicam, which is also reduced on a mercury electrode giving a well-defined cyclic voltammetric peak. It is proposed that the decomposition of droxicam is controlled by a kinetic equation: v=(1.93 x 10(-3)+78.9 [D]) [D]=k(D)[D].

Salmonella enteritidis temperature-sensitive mutants protect mice against challenge with virulent Salmonella strains of different serotypes.

The protection conferred by temperature-sensitive mutants of Salmonella enteritidis against different wild-type Salmonella serotypes was investigated. Oral immunization with the single temperature-sensitive mutant E/1/3 or with a temperature-sensitive thymine-requiring double mutant (E/1/3T) conferred: (i) significant protection against the homologous wild-type Salmonella strains; (ii) significant cross-protection toward high challenge doses of S. typhimurium. Significant antibody levels against homologous lipopolysaccharide and against homologous and heterologous protein antigens were detected in sera from immunized mice. Moreover, a wide range of protein antigens from different Salmonella O serotypes were recognized by sera from immunized animals. Besides, primed lymphocytes from E/1/3 immunized mice recognized Salmonella antigens from different serotypes. Taken together, these results indicate that temperature-sensitive mutants of S. enteritidis are good candidates for the construction of live vaccines against Salmonella.

Intramammary immunization with live-attenuated Staphylococcus aureus protects mice from experimental mastitis.

Female mice were immunized by the intramammary route with live-attenuated Staphylococcus aureus according to different schedules and challenged with virulent S. aureus. Immunization in late pregnancy or early lactation induced a significant decrease (P <0.05) in the number of S. aureus CFU recovered from glands after the challenge and a significant increase (P <0.05) in the levels of milk and serum specific IgG and IgA antibodies. Mice immunized before pregnancy were not protected from S. aureus challenge. Immunization did not increase the number of somatic cells in milk when compared with control mice. Protection from S. aureus intramammary infection may be achieved if mice are locally immunized during late pregnancy or early lactation.

Attenuated mutants of Staphylococcus aureus with two temperature-sensitive lesions: isolation and characterization.

The feasibility of constructing attenuated mutants of Staphylococcus aureus with two temperature-sensitive (ts) lesions for ultimate development of a live-attenuated strain was investigated. Temperature-sensitive S. aureus strain G/1/2, which grows well at 31 degrees C but does not replicate at 37 degrees C, was subjected to chemical mutagenesis. After two enrichment cycles, fifteen mutants able to grow at 25 degrees C but unable to grow at 31 degrees C, were identified. Growth curves with temperature shifts from 25 to 31 degrees C, and from 31 to 37 degrees C confirmed that these were mutants with two lesions (dts), each with a different cut-off temperature. The reversion frequency of mutant G/1/2 at 37 degrees C was 2 x 10(-6) whereas those of several dts mutants were much lower (dts7: 7 x 10(-9) and dts12: 1 x 10(-9)). There was no increase in ts mutation reversion rate in response to prolonged incubation at 37 degrees C. The data support the further development of these mutants for use as a stable attenuated vaccine.

Angina bullosa hemorrhagica.

Angina bullosa hemorrhagica is characterized by acute blood blisters, mainly on the soft palate. Elderly patients are usually affected and lesions heal spontaneously without scarring. The pathogenesis is unknown, although it may be a multifactorial phenomenon. Trauma seems to be the major provoking factor and long term use of steroid inhalers has also been implicated in the disease. No underlying hematologic or immunopathogenic disorder has been found. Treatment is symptomatic. We present a 67-year-old patient with recurrent oral blood blisters which were diagnosed as angina bullosa hemorrhagica. Trauma by dental injections and use of steroid inhalers were identified as etiologic factors in this case. Erosions healed with a week. Although this is a benign condition, it may result in acute airway obstruction. Recognition is, therefore, of great importance for dermatologists.

Plasma levels of 8-methoxypsoralen after bath-PUVA for psoriasis: relationship to disease severity.

Plasma levels of 8-methoxypsoralen (8-MOP) were determined by high-pressure liquid chromatography in 19 patients with psoriasis who were receiving bath-PUVA treatment, at different time points after the psoralen bath. The levels of 8-MOP varied between < 5 ng/ml (lower limit of detection) and 34 ng/ml, and we found a relationship between the plasma psoralen levels and the severity of the disease.

[Ambulatory surgery. Experience at a pediatric surgery department].

In October 1987 we opened an Outpatient Surgical Unit, since January 1988 to December 1993 we have treated 8232 patients with an mean age (range of 2 weeks to 17 years). All the patients were given general anesthesia, loco-regional anesthetics or deep sedation. 33.5 percent of the total number of patients operated in our hospital were treated in this Unit, being 41.4 percent if related to programmed surgery. Only 2.5 percent of them were admitted in the inpatient area with no significant complications.

Pachydermodactyly: a case report.

Pachydermodactyly is an uncommon variant of digital fibromatosis that has previously been related to knuckle pads. It is a benign condition that needs no investigation for an underlying systemic disease. We present a new case of pachydermodactyly and comment on the available literature.

Amphibian cross-fertilization and polyspermy.

A string of oocytes from Bufo arenarum (Ba) was inseminated with spermatozoa from Ba, Leptodactylus chaquensis (Lch), or Bufo paracnemis (Bp). Homologous insemination resulted in monospermic eggs and normal development, while in the case of heterologous fertilization both polyspermy and abnormal development were the rule. Oocytes from Ba inseminated with homologous spermatozoa, when reinseminated 30 sec after the first insemination with heterologous spermatozoa, exhibit a high rate of polyspermy and abnormal development. A lower rate of abnormalities was observed when reinsemination was carried out 60 sec after the first insemination. Normal development and an absence of polyspermy were observed in the case of eggs reinseminated 300 sec after initial insemination. This result indicated that the electrophysiological blockage of fertilization either was not triggered or was ineffective. The establishment of the permanent blockage of polyspermy, either by artificial activation of oocytes or as a result of their incubation with the products of the cortical granules, inhibited penetration of Ba eggs by heterologous sperm.

Interaction between granulocytes and antibodies in the enhancement of lung defenses against Staphylococcus aureus after intranasal immunization of mice with live-attenuated bacteria.

Immunization with live-attenuated Staphylococcus aureus induced measurable levels of specific IgG and IgA in the lungs, but the pulmonary clearance of S. aureus in immunized mice did not differ from that of control mice. Aerosol exposure of mice to Pseudomonas aeruginosa induced a significant recruitment of polymorphonuclear leukocytes (PMNL) to the lungs in both immunized and control mice, whereas S. aureus challenge did not. However, challenge with a mixture of P. aeruginosa-S. aureus or exposure to an aerosol of Escherichia coli lipopolysaccharide (LPS) before S. aureus challenge induced PMNL migration and a significant enhancement of pulmonary clearance of S. aureus in immunized mice. The presence of both antibodies and PMNL was required for enhancement of S. aureus pulmonary clearance.

Exocytosis of cortical granules from activated oocytes of the toad, Bufo arenarum.

Observation of the cortical region of oocytes of Bufo arenarum by transmission electron microscopy reveals modifications on their surface and in the contents of the cortical granules (CG) during activation. In non-activated oocytes only amorphous cortical granules (ACG) can be observed. Activated oocytes display ACG, intermediate cortical granules containing both amorphous and membranous material (ICG), and a third type containing only membranous material (MCG). During exocytosis, CG release their contents into the perivitelline space, where the amorphous and membranous materials are found. The three types of CG found during oocyte activation suggest transformation of ACG to MCG and indicate that the different components of the cortical granules, when released into the perivitelline space, might play different roles in prevention of polyspermy.

Carbon tetrachloride activation, lipid peroxidation and liver necrosis in different strains of mice.

Males from three different strains of mice (GXF; CF1, and Swiss) were compared in their liver response to CCl4 effects. They were capable to intensively activate CCl4 to .CCl3. No CCl4 induced lipid peroxidation was detected in the liver microsomes from any of them. All the strains remakably responded to the CCl4 induced liver damage. Results further strengthen our previous hypothesis that covalent binding of CCl4 reactive metabolites to cellular constituents is more important in relation to liver cell injury than early changes in lipid peroxidation.

Carbon tetrachloride induced polysome breakdown. Relative importance of lipid peroxidation and of binding to ribosomal components in the process.

In vivo, CC1-4 causes an intense polysome breakdown; CHC1-3 effect is much less intense and CH2C1-2 effect is negligible. Previous administration of either diphenyl-p-phenyelnediamine or alpha-tocopherol to the rats does not prevent CC1-4-induced polysome breakdown. Promethazine and cystamine were able to prevent to a good extent the CC1-4 induced polysome breakdown process. 14CC1-4 irreversibly binds to ribosomal proteins but not to ribosomal RNA. Several hepatocarcinogens which are effective alkylating agents and cause in vivo polysome breakdown do not induce a lipid peroxidation process.
