Antinociceptive activity of the novel fentanyl analogue iso-carfentanil in rats.

A large number of fentanyl analogues have been synthesized so far, both to establish the structure-activity-relationship (SAR) and to find novel, clinically useful antinociceptive drugs. In this study, the newly synthesized fentanyl analogue 3-carbomethoxy fentanyl (iso-carfentanil) was compared to fentanyl for its antinociceptive activity (tail-immersion test) in rats. It was revealed that the introduction of a 3-carbomethoxy group in the piperidine ring of fentanyl skeleton reduced the potency and shortened the duration of action of the parent compound, i.e., fentanyl. The antinociceptive potency of 3-carbomethoxy fentanyl is influenced mainly by the steric factor (voluminosity of the carbomethoxy group and the cis/trans isomerism), while the chemical nature of the group is probably irrelevant. This is in agreement with SAR studies of other 3-substituted fentanyl analogues. In contrast to potency, the duration of action is not affected by cis/trans isomerism. It is assumed that the time course of action of 3-carbomethoxy fentanyl is influenced by the nature of the carbomethoxy group. Since the potency and the duration of action of this novel antinociceptive compound are interesting from the aspect of SAR studies and have potential promise for clinical application, 3-carbomethoxy fentanyl deserves to be extensively evaluated.

Higher environmental temperature potentiates cataleptic effect of fentanyl in rats.

The influence of higher environmental temperature (HET=30/-1 degrees C) on fentanyl-induced behavior was studied in unrestrained rats. Subacute exposure (3 days) of rats to HET significantly (P<0.01) increased the cataleptic effect of fentanyl citrate (0.5 mg/kg), in comparison to the corresponding exposure to normal environmental temperature (NET=22+/-1 degrees C). Also, the hyperthermic response of rats to a low dose of fentanyl citrate (0.2-0.5 mg/kg) was significantly (P<0.01) potentiated, and the hypothermic response to a high dose of fentanyl citrate (1.5 mg/kg) was significantly (P<0.05) attenuated after exposure to HET. Fentanyl-induced hyperexcitability, loss of righting reflex, loss of corneal reflex and analgesia were not significantly affected by HET. This study provides the first evidence on the influence of environmental temperature on drug-induced catalepsy. HET-induced potentiation of the cataleptic response to fentanyl could be the result of an interference with behavioral thermoregulation.

Expressed emotion in the families of patients with schizophrenia and its influence on the course of illness.

This paper presents the results of a study carried on an urban population in Belgrade investigating the connections between relapse in schizophrenia and the expressed emotion (EE) status of families where at least one of the patient's parents was a member of the household. The overall rate of high EE was just under 50%, in the middle of the range of values reported in studies carried out elsewhere. Relapse was found to be 10 times more frequent in patients whose families were rated high in EE. The sample consisted of 30 patients with hebephrenic schizophrenia and 30 with paranoid schizophrenia. The results suggested that these subtypes may be associated with different attributes of EE. Criticism was more frequent in families of patients with paranoid schizophrenia, while emotional overinvolvement was more frequent in families having a hebephrenic offspring. The different components of EE, suitably combined, may differentiate between the two subtypes of schizophrenia in terms of relapse rate. The relative odds for high criticism were similar for both subtypes, but for maternal overinvolvement (EOI) the odds were 10.5 for hebephrenic as against 2.3 for paranoid schizophrenia. Thus, EOI was more common in the families of those with hebephrenia and also led more frequently to relapse. There was also an inverse relationship between relapse rate and warmth, whether paternal or maternal. This was significant in both subtypes, and indicated that the threshold for the positive effect of warmth was higher for fathers than for mothers. [Antibody titer in saliva and serum of experimental animals].

This paper reveals the antibody titer of plaque microorganisms in experimental gingivitis rats of differents ages and immunization. Before sacrificing experimental animals status of gingival tissue was assessed. Further on speciments of saliva and serum were taken for antibody titer measurements. None of the young rats developed gingivitis during experiment, where as the adult immunized rats bled on probing. Low values of antibodys point out the absence of intensive synthesis of specific antibodys in saliva upon the antigen stimulation of plaque maicroorganisms. Serum antibody titers were low in young rats, moderate in adult, and high in adult immunized rats. These results indicate that adult rats react stronger to plaque antigents than young rats and that previous contact with the antigens increases the reaction.

Isodicentric X chromosome in a woman with characteristics of gonadal dysgenesis.

This paper presents a female patient with primary amenorrhea in whose karyotype an aberrant X chromosome was present. The chromosome resulted from the fusion of two X chromosomes at distal parts of the long arm and from the loss of the segment q24 leads to qter. The clinical and cytogenetic picture is compared with that of patients with the same aberration reported by other authors.

[Relation between dental plaque fluoride content and caries].

The purpose of this study was to investigate the effect of plaque fluoride content on dental caries in children aged 12 years, from the area with minimal (Valjevo) and optimal and/or higher fluoride concentrations (Vranjska Banja) in drinking water. The caries prevalence was determined by the standardized epidemiological method. The fluoride concentration in dental plaques was measured with the Ion-selective electrode. The fluoride content in the plaque of children from Valjevo (7.8 ppm) was significantly smaller than in children from Vranjska Banja (36.5 ppm). In 12-year old individuals with the lowest fluoride concentrations in the plaque the greatest values of the caries prevalence were found. The increase in plaque fluoride content provoked the decrease in caries prevalence. The lowest values of caries index were found in children with 21 to 30 ppF in dental plaque.

[Relation between plaque index and gingival cell infiltrate in experimental animals].

The aim of this study was to determine the relation between plaque and gingival cell infiltrates in highly imbreaded laboratory rats during experimentally provoked gingivitis. In this experiment 24 Lewis rats, aged 1-3 months, were used. From the zero day of inflammation plaque was scored by Sillnes and Löe index, while the presence of gingival cell infiltrate was determined with the stereological method. The results point out that the elder animals manifested early plaque accumulation, while the young group of animals presented a more intensive gingival cell infiltration. On the basis of the obtained results it can be concluded that within the equal amounts of plaque the young animals had a more intensive gingival cell infiltration as the result of distincet defensive reaction to the presence of microbial antigens.
