Thyroid hormone profiles in experimental acute renal failure.

Thyroid hormone profiles were determined in two groups of dogs made uremic, either by i.v. uranyl nitrate 10 mg/kg BW injection or by bilateral ureteral ligation, and in one group of sham-operated animals. Each group consisted of 6 dogs and served as its own control. From blood samples taken in 12-h intervals up to 144 h for uranyl nitrate-injected dogs and 96 h for operated dogs, serum levels of T4, T3, fT4, fT3, rT3, and cortisol were measured by radioimmunoassays. The results obtained in both groups of uremic dogs showed an initial sharp fall of T4, T3, fT4, and fT3 followed by a plateau or retarded decrease. In sham-operated dogs the fall of these hormones was slight and of short duration. Reverse T3 had a tendency to increase in all groups examined, but a significant elevation was recorded only after bilateral ureteral ligation. In this group cortisol serum levels were found the highest, being also significantly increased in the other two groups. The temporal coincidence of the most marked alterations in T3, rT3, and cortisol serum concentrations indicates a significant role of stress in thyroid dysfunction. Although serum creatinine rise and weight loss were not parallel with thyroid hormone alterations, the involvement of uremic compounds and malnutrition in this process is also quite clear. Thus, the data presented suggest simultaneous influences of uremic toxins, stress, and malnutrition on the induction of thyroid dysfunction in dogs made uremic by uranyl nitrate injection or bilateral ureteral ligation.(ABSTRACT TRUNCATED AT 250 WORDS)

Acute Kidney Injury↗

Circadian rhythms in the urinary excretion of phosphate and cyclic 3',5'-adenosine monophosphate in kidney graft recipients.

Circadian rhythms of urinary phosphate excretion in 13 supine and 8 ambulatory kidney graft recipients in the late post-transplant period were studied. A comparison of serum phosphate level and urinary phosphate excretion revealed two groups of patients: a) with normal serum phosphate level and a high urinary phosphate excretion, and b) patients with subnormal serum phosphate and a moderate phosphate excretion. Compared to control group of 9 healthy volunteers much greater variations in phosphate and cyclic 3',5'-AMP excretion in grafted patients were recorded. An inversed phosphate excretion rhythm was observed in 5 out 13 supine patients.

The effect of five days calcium chloride treatment on the somatostatin cells of the antral mucosa in white rats.

The effects of a five day treatment with calcium chloride (1 ml 0.11 M CaCl2/100g body weight i.p., twice daily) on somatostatin cells in gastric antral mucosa of white rats were investigated. Somatostatin cells of experimental and control animals were identified by immunoperoxidase method using rabbit antihuman antisomatostatin (1:1000), and counted on perpendicularly cut sections taken from four transversal levels going from pylorus to antrofundic border. The results obtained have demonstrated the occurrence of somatostatin cell hyperplasia in animals treated with calcium chloride, established on the ground of highly significant rise in the number of somatostatin cells and a change in their topography.

[Diurnal rhythm and the effect of hemodialysis therapy on blood prolactin levels in patients with chronic renal insufficiency].

In 97 patients with different stages of chronic renal failure prolactin serum level was determined, and its diurnal rhythm and dependence on haemodialysis treatment were studied. Diurnal rhythm was investigated in 6 men and 6 women with advanced chronic renal failure on diet and other symptomatic treatment. A normal diurnal rhythm of prolactin was recorded only in 3 patients, two of them being with elevated basal prolactin levels; an abnormal prolactin rhythm was documented in 8 patients. In 4 patients no evident diurnal changes in prolactin level were observed, though 3 of them had a normal basal prolactin level. In one patient slight fluctuations of the prolactin level were recorded, while in 4 other patients an abnormal type of diurnal rhythm was observed. Prolactin determinations in patients on maintenance haemodialysis showed increased values in 45 men (719 + 122 mIU/l) as well as 34 women (1251 + 171 mIU/l). The analysis of the influence of haemodialysis on cuprophane and polyacrylonitrile membrane was made in 6 patients and no difference was shown between predialysis and postdialysis serum prolactin values, as well as in values obtained for non-dialysis and dialysis days. The results obtained in this study showed an absent or abnormal diurnal rhythm of prolactin in 75% of patients with chronic renal failure. In patients on maintenance haemodialysis increased prolactin levels were found. Haemodialysis procedure on cuprophane and polyacrylonitrile membrane failed to affect the serum level of prolactin.
