Mechanism of dexamethasone-mediated interleukin-8 gene suppression in cultured airway epithelial cells.

The effects of dexamethasone, a glucocorticoid analog, on interleukin 8 (IL-8) gene expression were studied in cultures of primary human tracheobronchial epithelial cells and an immortalized human bronchial epithelial cell line, HBE1 cells. Dexamethasone inhibited IL-8 mRNA and protein expression in a concentration- and time-dependent manner. The inhibition did not occur at the transcriptional level since both nuclear run-on activity and IL-8 promoter-reporter gene expression assay revealed no significant effect. Instead, there was a change in IL-8 mRNA stability in dexamethasone-treated cultures. Under actinomycin D treatment, IL-8 mRNA was quite stable in dexamethasone-depleted cultures, while in dexamethasone-pretreated cultures, IL-8 message was rapidly degraded within the first hour, then leveled off. When dexamethasone and actinomycin D were added simultaneously to dexamethasone-depleted cultures, IL-8 mRNA remained rather stable. When cycloheximide was used to inhibit new protein synthesis, dexamethasone-dependent inhibition was not observed. These results suggest that a posttranscriptional mechanism, which requires dexamethasone-dependent new protein synthesis, is involved in the regulation of IL-8 mRNA by dexamethasone in airway epithelial cells.

The variety of relations between spatial frequency and reaction time.

While stimuli of lower spatial frequency often result in faster responses, this is not invariably the case. Some individuals respond faster to high frequency stimuli on tasks for which most others respond faster to low frequency stimuli. Past experience can also determine the degree to which a low frequency stimulus will mask a high frequency stimulus. These observations suggest that the effects of spatial frequency on visual information processing are under the control of higher level cognitive operations.

Sleep duration and eating behaviors of college students.

31 short-sleeping college students tended to eat more small meals or snacks than 37 long sleepers, all of whom were satisfied with their sleep. This disrupted pattern of larger meals was predicted from work of Elomaa and Johansson with rats who were partially REM-sleep deprived.

Effect of chronic ethanol treatment and withdrawal on mitochondrial redox activity in brain cortical slices.

The effect of chronic ethanol treatment on cytochrome a-a3 redox activity was studied in rat brain cortical slices. The slices were depolarized by electrical pulses and the level of reduced cytochrome a-a3 was directly measured within the slice by dual wavelength spectrometry. Cortical slices cut from the brains of rats withdrawn from ethanol demonstrated less reduction of cytochrome a-a3 following electrical depolarization compared both to controls and ethanol addicted rats. It is suggested that this effect of ethanol withdrawal on cytochrome a-a3 redox activity is due to a change in the regulation of mitochondrial activity.

A strategy to enhance caring and community in the learning environment.

The authors describe the implementation and evaluation of faculty-facilitated small groups that encourage dialogue between nursing faculty members and students about teaching/learning concerns and events. These groups promoted student perceptions of caring behaviors from their faculty and peers and developed students' sense of cohesiveness within the learning community.
