Specific immunotherapy stimulates the binding of histamine H2 receptor antagonist by lymphocytes.

The binding of antagonists of histamine receptors H1 (promethazine) and H2 (ranitidine) by peripheral blood lymphocytes from pollinotics was determined before and after the course of immunotherapy. We found that lymphocytes from atopic subjects showed significant decrease in the binding of H2 receptor antagonist as compared to control subjects. Specific immunotherapy induced statistically significant increase in H2 receptor antagonist binding, which correlated with the improvement of clinical symptoms.

Binding of antagonists of H1 and H2 histamine receptors to peripheral blood lymphocytes of atopic and healthy subjects.

The binding of the antagonists of histamine H1 and H2 receptors by peripheral blood lymphocytes from atopic and healthy subjects was investigated. We found that lymphocytes from atopic subjects showed statistically significant decrease in the binding of H2 receptor antagonist - ranitidine. In addition, lymphocytes from atopic and control subjects had similar capacity of binding of H1 receptor antagonist - promethazine. The ratio of the amount of H1 and H2 antagonists, bound to lymphocytes from atopic and healthy subjects, was calculated. The difference between the values in the group of atopic (2.55) and control subjects (1.55) was statistically significant.

Histamine upregulates Th1 and downregulates Th2 responses due to different patterns of surface histamine 1 and 2 receptor expression.

Histamine, which acts via G protein-coupled receptors, is an important mediator of immediate hypersensitivity and is also able to influence the nature of T cell responses. We demonstrated that TH1 and Th2 cells express distinct surface histamine receptor patterns and that Th1-type responses are enhanced by histamine, whereas Th2-type responses are negatively regulated, due to different intracellular signals generated by histamine stimulation. These findings account for negative feedback regulation in a wide variety of pathologies.

[Mixed connective tissue disease in a male patient chronically exposed to toxic chemicals].

A case of the mixed disease of the connective tissue (MCTD) in male patient occupationally exposed to PVC and other toxic agents is presented. Clinical symptoms consisted of the typical signs of SLE, rheumatoid arthritis and lupoid hepatitis. MCTD diagnosis was confirmed serologically by the presence of autoantibodies anti-RNP. Prednisone administered in the daily dose of 60 mg produced remission.

CAST-ELISA test in diagnosis of mite allergy.

The recently developed CAST-ELISA method was applied to determine allergen-induced leukocyte stimulation. This method is based on the measurement of sulfidoleukotriene secretion by peripheral blood leukocytes stimulated with specific allergen in the presence of interleukin 3.23 patients allergic to Dermatophagoides pteronyssinus were included in this study. All subjects received no medication 2 weeks before the testing. Leukocytes were isolated by dextran sedimentation. After incubation with allergen the cells were centrifuged and the concentration of sulfidoleukotrienes in supernatant was determined. The results were expressed in pg/ml after subtracting the values of spontaneous sulfidoleukotriene production in portions incubated without allergen. We observed a wide range of sulfidoleukotriene secretion upon allergen stimulation. Concentrations of leukotrienes ranged from 0 to 3440 pg/ml. The mean level in the whole group was 1521.3 pg/ml at the optimal allergen concentration. Positive results were observed in 21 subjects. In 2 subjects we could not demonstrate leukotriene secretion by stimulation with allergen. 15 healthy control subjects were also included in this study. 12 subjects had negative skin prick tests (SPT) as well as CAST results. In 2 healthy subjects with positive SPT, we also observed positive CAST results. However, leukotriene secretion was lower than 300 pg/ml in these subjects. We conclude that CAST-ELISA is a valuable method for determining the allergic status in patients with mite allergy.
