[Population genetics of HLA polymorphism in blood donors of Bremen and Hannover].

501 blood donors from Bremen have been typed for HLA-ABC and -DR. The results are compared with HLA data obtained on 474 blood donors from Hannover. The gene frequencies do not differ significantly between these two population samples. Comparisons with population samples from Kiel, Hamburg, Essen, Frankfurt/M., Mainz, Mannheim, Freiburg/Br., Munich and Vienna did also not reveal any remarkable differences concerning the gene frequencies. Analysis of linkage disequilibrium of two-factor and three-factor haplotypes could show that the typical Caucasoid allele combination A1/B8 is not a constituent part of three-factor haplotype combinations. Between the population samples from Bremen and Hannover no marked differences in the distribution of two-factor and three-factor haplotype frequencies could be found.

Adenylate cyclase in membrane fractions of RIN-A2-cells: studies with forskolin, NaF, GppNHp and NEM.

In crude membrane fractions of rat pancreatic islets and of RIN-A2-cells, forskolin and NaF stimulated adenylate cyclase activity. Basal and stimulated enzyme activity was approximately 3 to 6 fold higher in membranes of RIN-A2-cells than in membranes of islet cells. In RIN-A2-cells GppNHp and NEM inhibited forskolin-stimulated enzyme activity. The inhibitory effect of GppNHp could be reduced by NEM. It is suggested that the adenylate cyclase system of RIN-A2-cells contains inhibitory and stimulatory N-proteins and that there are critical thiols related to Ni, Ns and/or the catalytic unit. Thus, membrane fractions of RIN-A2-cells may be an appropriate model for studies on the adenylate cyclase system of insulin-producing cells.

[Uric acid determination in dilute serum with an enzyme electrochemical and enzyme-free sensor].

The paper describes the development of electrochemical methods for analytical determination of uric acid basing on amperometric sensors: with an uricase enzyme electrode, termed "electrochemical-enzymatic" method; with an enzyme-free electrode by indication of the anodic oxidation of uric acid, termed "electrochemical" method. The comparison of analytical quality of both methods shows a sufficient specificity, precision, and accuracy. They show a good correlation with regard to the specific uricase-catalase reference method (UCM): yelectr./enz. = (0.943 xUCM + 19.8) mu mol/l; r = 0.9948 yelectr. = (0.964 xUCM + 6.31) mu mol/l; r = 0.9917 The analysis by the "electrochemical" method can be done with a higher frequency of samples per hour completely without need of enzyme. Both methods are principally suitable for an application in analytic biochemical laboratories.

[Pathologico-anatomical and histological studies with special reference to the tracheal mucus membrane of calves following exposure to an aerosol disinfectant (Wofasteril)].

Various morphological methods were used in examining calves, with the view to determining the action of Wofasteril, an aerosolic disinfectant, on their organism, particularly on their trachea. Calves were found to differ from animals with pneumonia and other pulmonary inflammations, in that no transformation and quality change occurred to the mucus of their tracheal goblet cells in response to peracetic acid. Pathologico-anatomic and histological alterations in various organs were not attributable either to the disinfectant. A contribution is made to a more general description of the normal tissue structure, in that data are presented on the length, width, and number of goblet cells in the trachea of calf.

[The interaction of methemoglobin and modified poly-(hydroxyethylmethacrylates) studied by spectroscopic methods].

The interaction between methemoglobin (MetHb) and macroporous matrices on the basis of polymethacrylates was investigated by means of optical and e.p.r. spectroscopy. The spectroscopic data show that the adsorption of MetHb to imidazole-containing matrices occurs by complex formation between matrix-bound imidazole and the iron of the prosthetic group, with all 4 polypeptide chains of the MetHb molecule being included in the interaction. The adsorption to hydrophobic side chains containing matrices leads, via the protein-matrix interaction, to considerable disturbances of iron protoporphyrin IX in equilibrium or formed from protein-contacts, which are of general importance with respect to the functional variablity and control, respectively, of iron porphyrins in hemoproteins. In case of matrix containing n-hexyl groups deoxyHb is oxidized by O2 to MetHb, instead of being oxygenated to HbO2. Not all prosthetic groups are able to bind N-3. With the increase in hydrophobicity of the matrix a conformational change is enforced leading in the beta-chains to the direct interaction between iron and sulfur of cysteine (beta-cys 92), as it is proved in all cytochrome P-450 and other model compounds.

[Glucose determination in diluted whole blood using an enzyme electrode].

The construction and operation of a measuring device containing an enzyme electrode is described. Glucose oxidase is enclosed in an acrylamide film by means of photopolymerization. The maximal increase of the anodic current of the H2O2 being formed in the enzyme reaction serves as measuring signal. The device gives for diluted serum and blood correlation coefficients above 0,99 according to the methods given in the German Pharmacopeia 7. The measuring time for each specimen is roughly 90 s, the serial variation coefficient 2%.

Comparison of the peroxidatic activity of cytochrome P-450 with other hemoproteins and model compounds.

The H2O2 dependent catalysis of cytochrome P-450 was compared with the catalytic mechanism of horse radish peroxidase, methemoglobin and iron protoporphyrin complexes. A relatively stable intermediate being comparable to compound I of horse radish peroxidase is formed in the case of iron porphyrin complexes, methemoglobin and probably cytochrome P-450. In the case of peroxidase compound II is the more stable intermediate. This could be the reason for the different catalytic properties of peroxidase on the one hand and iron porphyrin complexes, methemoglobin and cytochrome P-450 on the other hand.

Aspects of application of cytochrome P-450 and related systems in substrate hydroxylation.

Extrapolating the recent progress in the near future the extensive utilization of cofactor-dependent enzymes (enzymes of the 3rd generation) for solving economic or medical problems will be restricted by the difficulties of cofactor regeneration. Real possibilities exist in analytical systems, for instance enzyme electrodes. In the present paper a special case of overcoming the cofactor regeneration in P-450 catalyzed substrate hydroxylation is demonstrated: The peroxide-dependent reaction gives the same products as obtained under physiological conditions; that is why in an electro-enzyme-reactor producing hydrogen peroxide by cathodic oxygen reduction a considerable simplification of the multi-enzyme complex is possible by omitting electron transfer proteins. At present the main problem is the instability of the terminal oxidase. Attempts are being made to solve these problems by immobilizing the protein or substituting P-450 by other hemoproteins or iron porphyrin derivatives.

[Chemical and immunogenic properties of the capsular antigen of Staphylococcus aureus. III. Protective activity of capsular antigen in experiments on mice].

Experiments were conducted on 1500 albino mice; protective activity of 7 batches of S. aureus capsular antigen isolated from 5 strains was studied. The animals were immunized subcutaneously with different doses of the preparation (from 0.001 to 10000 microgram) once, twice, and thrice, and infected intraperitoneally with 6 encapsulated cultures. Capsular antigen proved to possess weak protective properties. The following regularities were revealed: 1) high antigen doses (over 5 microgram) increased mouse sensitivity to staphylococcus infection; 2) the greatest protective activity was shown by doses within 0.01-1 microgram; 3) there was no distinct "dose-effect" dependence; 4) single immunization was less effective than double and triple; 5) antigenic specificity of the capsular material was not connected with its protective properties.

CGS 9343B and W7 (calmodulin antagonists) inhibit KCl-induced increase in cytosolic free calcium and insulin secretion of RINm5F cells.

CGS 9343B:1,3-Dihydro-1-[1-[4-methyl-4H,6H-pyrrolo[1,2-a]-[4,1] benzoxazepin-4-yl)methyl)-4-piperidinyl]-2H-benzimidazol-2-o ne maleate and W7:N-6(aminohexyl)-5-chloro-1-naphthalenesulfonamide) are calmodulin antagonists with different specificities. The effects of CGS 9343B and W7 on cytosolic free calcium concentration ([ Ca2+]i) and insulin release were investigated in rat insulinoma cells (RINm5F). As measured with the Quin-2 technique, preincubation with CGS 9343B (0.3-10 microM) and W7 (5-50 microM) concentration dependently decreased KCl (25 mM)-mediated accumulation of cytosolic calcium. Both, CGS 9343B (10 microM) and W7 (50-100 microM) almost abolished the alanine- and KCl-induced increase in [Ca2+]i and significantly inhibited KCl (25 mM)- and alanine (10 mM)-mediated insulin release. W5 (100 microM), the chlorine-deficient analogue of W7 with decreased affinity for calmodulin, did not inhibit the KCl-induced increase in [Ca2+]i and enhanced basal and KCl-mediated insulin release by 56% and 189%, respectively. Our data suggest that CGS 9343B and W7 inhibit the depolarization-induced calcium uptake and subsequent increase in [Ca2+]i.

Plasma levels of cytokines and soluble cytokine receptors in psychiatric patients upon hospital admission: effects of confounding factors and diagnosis.

It has been hypothesized that the immune system plays a pathogenetic role in psychiatric disorders, in particular in major depression and schizophrenia. This hypothesis is supported by a number of reports on altered circulating levels and in vitro production of cytokines in these disorders. However, the respective evidence is not consistent. This may be in part due to an incomplete control for numerous confounding influences in earlier studies. We investigated the plasma levels of cytokines and soluble cytokine receptors in psychiatric patients (N = 361) upon hospital admission and compared the results to those obtained in healthy controls (N = 64). By multiple regression analysis we found that circulating levels of interleukin-1 receptor antagonist (IL-1Ra), soluble IL-2 receptor (sIL-2R), tumor necrosis factor-alpha (TNF-alpha), soluble TNF receptors (sTNF-R p55, sTNF-R p75) and IL-6 were significantly affected by age, the body mass index (BMI), gender, smoking habits, ongoing or recent infectious diseases, or prior medication. Cytokine or cytokine receptor levels were significantly increased in patients treated with clozapine (sIL-2R, sTNF-R p75), lithium (TNF-alpha, sTNF-R p75, IL-6) or benzodiazepines (TNF-alpha, sTNF-R p75). Taking all these confounding factors into account, we found no evidence for disease-related alterations in the levels of IL-1Ra, sIL-2R, sTNF-R p75 and IL-6, whereas levels of TNF-alpha and sTNF-R p55 in major depression and sTNF-R p55 in schizophrenia were slightly decreased compared to healthy controls. We conclude that, if confounding factors are carefully taken into account, plasma levels of the above mentioned cytokines and cytokine receptors yield little, if any, evidence for immunopathology in schizophrenia or major depression.

[Use of cytochrome P-450 and ferroporphyrin as catalyzers in hydroxylation reactions jointly with electrochemical systems].

The mechanism of P-450 catalyzed reactions is discussed. They are shown not to be true peroxidase reactions. Liver microsome P-450 is capable of oxidizing such substrates as benzphenamine, amidopyrine and p-nitroanisole via molecular oxygen reduced on the cathode. The methods of stabilizing the enzymic system by means of immobilization of microsomes or isolated components are described. The possibility of coupling the enzymic and the electrochemical system is considered. The approaches to the modelling of the cytochrome P-450 catalytic activity with the aid of ferroporphyrine are proposed.
