Plasma and skin blister fluid concentrations of trimethoprim following its oral administration.

Plasma and skin blister fluid concentration-time curves following a single oral dose of trimethoprim have been evaluated. Skin blisters were produced by the cantharides technique, using patches with cantharidin ointment. Trimethoprim concentrations in plasma following multiple doses of 200 mg were also determined. The maximal concentration in plasma after a single oral dose of 400 mg trimethoprim was 3.95 +/- 1.08 mg/l, and it was observed after 2 h, whereas in skin blister fluid the level was 2.21 +/- 0.62 mg/l, and it was delayed for up to 6 h. This means that a certain time is required for drug transfer from the capillaries via the basal membrane into blister fluid. Penetration of the drug into blister fluid, defined as the ratio of the areas under the trimethoprim level time curve in skin blister fluid to that of plasma, was 0.826 +/- 0.096. The steady-state concentration of trimethoprim in plasma during routine treatment with 200-mg doses ranged between 2 and 3.5 mg/l.

Distribution of trimethoprim and sulphamethoxazole in blood during treatment with co-trimoxazole.

Changes in the distribution of sulphamethoxazole and trimethoprim in whole blood, plasma and erythrocytes at steady-state in patients treated with cotrimoxazole have been studied. Unlike sulphamethoxazole, trimethoprim was weakly bound to erythrocytes and was partially liberated when the erythrocytes were rinsed with isotonic saline. The maximal steady-state concentration of trimethoprim in whole blood was 3 mg/l, but calculated on the basis of the concentration determined in erythrocytes it was 1.8 mg/l. Erythrocytes may be of great significance in trimethoprim distribution as carriers of a readily liberated reservoir of the drug. Acetylated sulphamethoxazole derivatives occurred in a higher percentage in erythrocytes at the maximal steady-state concentration (9.9%) than at the level (7.3%), which may help in interpreting the behaviour of this metabolite in other cells in the organism.

Penetration of trimethoprim and sulfamethoxazole into skin blister fluid.

In 32 patients the concentrations of sulfamethoxazole and trimethoprim in whole blood, plasma and skin blister fluid were studied in the course of treatment with co-trimoxazole and trimethoprim alone. Measurements were taken on the fourth day of treatment, 3 h after administration of the morning dose of the drug. The blood contained a lower concentration of sulfamethoxazole than plasma. About 70% of the sulfonamide penetrated into the exudate from plasma. Trimethoprim administered conjointly with sulfamethoxazole to a higher degree penetrated skin blister fluid to a greater extent than when given alone.

[ECG picture in rats exposed to the action of petroleum].

The male rats of Wistar strain were exposed to the vapours of arabic petroleum Abu-Dhaki Zakun at concentration 1500 mg/m(3) during 16 weeks, 6 times weekly per 90 minutes daily. The ECG examinations revealed progressive bradycardia, increase of the percentage of conduction disturbances appearing as the bundle-right branch block, changes of the shape of P deflection, decrease of the angle of inclination of QRS vector, and increase of the percentage of nonspecific changes of ST-T syndrome. The obtained results can suggest a noxious effect of petroleum on the heart muscle when given by inhalation in a determined concentration.

Effects of "essential" phospholipids (EPL) on experimental ethanol-induced cardiomyopathy.

15 percent ethanol alone and together with "essential" phospholipids (EPL) was applied orally (as the only source of drinking fluid) ad libitum to white rats over 8 months. The evaluation of effects of EPL was based on biochemical assays of the blood serum, including creatine phosphokinase (CPK), a-hydroxybutyrate dehydrogenase (HBD) and free fatty acids (FFA) as well as on histological and histochemical studies of lactic dehydrogenase (LDH) of the heart muscle. In rats treated with ethanol alone we found increased CPK and HBD activities, elevation of FFA and lowered activity of LDH. EPL seem to protect against the above mentioned changes.
