Involvement of the area postrema in experimental hypertension has been investigated. Ablation of the rear apex of the fourth brain ventricle (=the region of the area postrema) elevated blood pressure, heart rate and plasma angiotensin II level. The same characteristic changes were seen in the two kidney Goldblatt rat model of hypertension. A possible involvement of the area postrema in this model of hypertension is discussed. Intraventricular perfusion of angiotensin II elevated blood pressure without a significant change in heart rates. This pressor response appeared to be dependent on release of antidiuretic hormone. The results are discussed in relation to the intrinsic brain angiotensinogenase system.
Cultured cells (chicken embryo liver cells and rat embryo fibroblasts) were treated with triparanol (MER-29) for various lengths of time. Both types of cells have developed numerous membranous whorls-myeloid bodies in the cytoplasm. Various stages in myeloid bodies development are described. Acid phosphatase activity was cytochemically demonstrated within the myeloid bodies, indicating their lysosmal nature. This activity appeared only at a late stage of the myeloid bodies formation.
Distribution of acid phosphatase as a marker enzyme for lysosomes was investigated in the isoprenalin stimulated rat parotid gland. The enzyme was localized in lipofuscin-like bodies as well as in non-discharged granules. The appearance of these bodies was correlated in time to the appearance of smooth vesicles and reduction of the acinar lumen. Ferritin, used as a tracer and introduced into the stimulated gland via cannulated parotid ducts, was found in smooth vesicles, vacuoles and lipofuscin-like bodies throughout the cytoplasm of the acinar cells. Very often ferritin-containing vesicles were found in the vicinity of the Golgi complex. In most cases the vesicles containing ferritin also showed acid phosphatase reaction product. A possible correlation between the lysosomal system and the process of recycling and degradation of membranes in the stimulated gland is discussed.
The specific adherence to target fibroblasts of lymphocytes differentiated in vitro from blast cells after stimulation with pokeweed mitogen was studied under the electron microscope. Lymphocyte pseudopods were seen to penetrate into the target cells forming a close contact between the membranes of the two interacting cells. Microfilaments were the only cytoplasmic components that could be detected within or in the vicinity of the penetrating pseudopods. No morphological signs of secretion could be observed in the contact region. Morphological aspects of the contact region are discussed as related to the lytic mechanism.
Interaction between pokeweed mitogen-stimulated secondary lymphocytes (PWM-lymphocytes) and target fibroblasts resulted in over 80 per cent adherence of the sensitized lymphocytes to the target cells. Adherence is by pseudopod penetration into target fibroblasts. The only lymphocyte cellular components found in the contact region were microfilaments. Cytochalasin B completely inhibited the specific adsorption of the PWM-secondary lymphocytes to the target cells. What adhesion did take place in the presence of cytochalasin B was found to be nonspecific. Ultrastructurally, the contact between lymphocyte and target cells was altered by the drug, when pseudopods were not observed. Possible effects of cytochalasin B on lymphocyte-mediated cytolysis, mainly by its effect on microfilament function, is discussed.
