Nitric oxide synthase type 1 expression in human lung cancer and its relation to p53.

BACKGROUND: The nitric oxide synthases (NOS) have been observed in human tumour cell lines as well as in solid tumours. Neuronal isoform of NOS (NOS1) was particularly abundant in low-differentiated gynaecological, breast and central nerve system tumours, suggesting that it may characterize poorly differentiated tumours. So far, little is known about expression of the neuronal NOS isoform in non-small cell lung cancer. Our aim was to compare NOS1 expression in non-small lung carcinomas with respect to tumor staging and p53 protein expression. MATERIAL AND METHODS: Thirty-two cases of non-small lung carcinomas of all grades of malignancy and ten control lung specimens with neoplastic lesions were examined. Paraffin-embedded tissues were stained with hematoxylin and eosin, or with antibodies to NOS1 and p53, and evaluated under light microscope. RESULTS: No statistical correlation between expression of p53, NOS1 and degree od tumour differentiation was observed. CONCLUSION: Expression of NOS1 can not serve as a marker for highly malignant tumours in the non-small cell lung carcinomas.

Influence of temporary ischemia on energetic functions of rat liver in early and late reperfusion period.

Energetic function impairment is one of the main reasons of liver ischemia-reperfusion injury. The aim of the study was to estimate the influence of temporary rat liver ischemia on energetic functions of the organ in the early and late reperfusion period and to answer the question whether the reduction of destructive factors release in sinusoids in the early phase of reperfusion can prevent liver energetic function impairment. To perform the experiment 55 Wistar rats divided into 5 experimental groups were used. 60 min. partial ischemia of the liver was applied, with 30 min, 72 h and 5 days reperfusion period. Suppression of KC was performed by intravenous administration of GdCl3. In the group of animals subjected to liver ischemia a considerable decrease of reaction to SDH and LDH activity was observed in the reperfusion period. In animals with KC suppression the loss of reaction to SDH intensity in zones I of acini was insignificant and the decrease of reaction to LDH was more orderly and proceeded in a more regular manner. The value of AKBR fell significantly after 30 min. of reperfusion in both groups of animals subjected to ischemia, while the serum level of AspAT and LDH increased rapidly. Temporary ischemia of the rat liver induces a decrease of the organ energetic functions in the early and the reperfusion period. The elimination of Kupffer cell activity in the early reperfusion phase does not prevent liver energetic functions impairment, but reduces the time in which they return to initial values in remote observation.

Circadian changes of cytochrome P-450-dependent monooxygenase system in the rat liver.

Circadian changes in cytochrome P-450 and cytochrome b5 content and activity of NADPH-cytochrome P-450 and NADH-cytochrome b5 reductases have been studied in rat liver microsomes in season autumn. The obtained results indicate, that cytochrome P-450 in 6-month-old animals shows 12 h rhythm, but in older ones 24 h rhythm. NADPH-cytochrome P-450 reductase activity shows 24 h rhythm in oldest animals only. Cytochrome b5 and its reductase has 24 h rhythm in all examined groups of rats.

The influence of submaxillary gland factor upon Golgi complex activity in jejunum epithelial cell of mice.

The effects of submaxillary gland factor on the thiamine pyrophosphatase, 5-nucleotidase (the markers of Golgi complex), adenosine-triphosphatase, NADPH-tetrazolum reductase and acid phosphatase were studied in the intestinal epithelium of mice. A decreased intensity of histochemical reactions for the markers of Golgi's complex after salivectomy and an increased activity after homogenate injection were confirmed. The reaction intensity to NADPH-tetrazolium reductase, adenosine-triphosphatase and acid phosphatase after salivectomy and after homogenate injection were similar to those of the control mice with sham operations. On the basis of investigations performed, this provides support, that submaxillary glands produce a factor which controls Golgi complex activity and may influence on glycocalyx synthesis.

[Role of alcoholism in the development of upper gastrointestinal tumors].

A preliminary report on the results of studies on the relationship between alcohol abuse and upper gastrointestinal tumours is presented. Alcohol abuse was measured with the aid of a questionnaire. GGTP and mean MCV were determined. Within the 1st September, 1986--30th September, 1987, 75 patients with the cancer of the upper gastrointestinal tract and 107 patients with benign lesions to this area were examined. Alcohol abuse was found in 33% of patients with the cancer and 19% of patients with benign lesions to the upper gastrointestinal tract. This difference is statistically significant (p greater than 0.05). The results of GGTP and MCV values were statistically insignificant in both groups. The authors have found more marked correlation of alcohol abuse with the cancer of the upper gastrointestinal tract in relation to the oral cavity and pharynx than the stomach. Further studies including larger groups of patients are recommended.

Influence of isoflurane on enkephalin levels and on some indicatory enzymes in the central nervous system of rabbits.

The objective of this study was to analyse the effects of isoflurane anesthesia (lasting for 15 or 60 min) and isoflurane anesthesia termination (after 1 or 24 h) on met-enkephalin (MENK) and leu-enkephalin (LENK) levels in discrete brain areas and spinal cord segments in rabbits. Moreover histochemical analysis of activities of succinate dehydrogenase, magnesium-dependent adenosine triphosphatase (Mg++ATP-ase) and acid phosphatase in the striatum and hypothalamus were carried out to evaluate the effects of isoflurane anesthesia on energetic, transport and catabolic processes. Throughout anesthesia (15 and 60 min) and after its termination (1 h) the LENK contents were increased in hypothalamus, hippocampus, mesencephalon and lumbar segment of spinal cord. Moreover, during isoflurane anesthesia and after its termination (1 h) MENK and LENK levels decreased in cervical segment and MENK content dropped in thoracic segment of spinal cord. Histochemical data indicated, that isoflurane enhanced energetic processes as well as exchange processes in neurocytes, glial cells, capillary walls and ependymal cells of the third ventricle. Measurements of acid phosphatase activity provided evidence of no signs of toxicity of isoflurane in the examined areas. The changes in enkephalin levels observed during the isoflurane anesthesia and after its termination depended on the type of examined neuropeptides, as well as on parts of the brain and spinal cord studied. The changes observed after isoflurane administration in enkephalinergic system are discussed with regard to our earlier experiments with halothane and enflurane.
