Free H2O2 released from human blood leukocytes during phagocytosis into the extracellular medium was highly reactive with the ferric form of HRP, forming an enzyme-substrate complex which was identical to HRP-H2O2 compound II. The formation of HRP-H2O2 compound II was employed for assaying the rates of H2O2 release by leukocytes upon addition of bacteria or myristate. The treatment of normal human blood leukocytes with myristate resulted in a marked stimulation of H2O2 release compared to phagocytizing cells. The activity of H2O2 release in response to myristate was found to be deficient in the leukocytes of two patients with chronic granulomatous disease. This assay method with myristate supplementation is so sensitive and specific that it should be useful for the diagnosis of chronic granulomatous disease.
Possible mutagenic activity of captan was investigated by in vitro and in vivo cytogenetic studies and by the dominant lethal study in mice. In vitro cytogenetic study with cultured human diploid cells revealed a significant increase in the frequency of cells showing stickiness and a severe mitotic inhibition at concentrations of 3.0 and 4.0 microgram of captan per ml. although no chromosomal aberrations were observed. In in vivo cytogenetic study, no chromosomal aberrations were induced in the bone marrow cells of rats treated orally with captan at a single dose of 500, 1000 or 2000 mg/kg or at five consecutive doses of 200, 400 or 800 mg/kg/day. Dominant lethal study also failed to show any mutation induction after treatment of male mice with daily oral dose of 200 or 600 mg of captan per kg bw for five days.

51 cases of chronic hepatitis were followed up by serial liver biopsies. 6 cases, all of which were of the active type, showed transition to liver cirrhosis within 5 to 10 months after the initial biopsy which had indicated chronic hepatitis. In 109 cases of liver cirrhosis, the antecedent acute hepatitis had been found within a relatively short period, usually 2 to 3 years before the establishment of the diagnosis of liver cirrhosis. Whether these cases which showed rapid transition to liver cirrhosis after acute hepatitis should be included in the group of chronic hepatitis or in another group remains to be settled.

A proteinase from the sarcocarp of melon (Cucumis Melo L. var. Prince) was purified by a three-step procedure involving batch-wise treatment with CM-cellulose fibers, column chromatography on CM-cellulose powder and gel filtration on Sephadex G-75. The final enzyme preparation was homogeneous on acrylamide gel electrophoresis. Its molecular weight was estimated by two different methods to be about 50,000. Anlayses indicated tha presence of 475 amino acid residues and at least 7 moles of hexose. The maximum activity was found in the alkaline pH region against casein as a substrate. The optimum temperature against casein was 70 degrees at pH 7.1. The enzyme was strongly inhibited by diisopropyl fluorophosphate, partly inhibited by HgCl2 and not inhibited by EDTA, p-chloromercuribenzoic acid, N-tosyl-L-lysine chloromethyl ketone, N-tosyl-L-phenylalanine chloromethyl ketone, and soybean trypsin inhibitor. The reduced and carboxymethylated insulin B-chain was cleaved at the peptide bonds of Asn3-Gln4, Cm-Cys7-Gly8, Glu13-Ala14, Leu15-Tyr16, Cm-Cys19-Gly20, Phe25-Tyr26, Pro28-Lys29, and Lys29-Ala30 by the enzyme.
