5-HT2A promoter polymorphism is not associated with anorexia nervosa in Japanese patients.

Genetic factors have been implicated in playing a significant role in susceptibility to anorexia nervosa (AN). Among many candidate genes for AN, an association with the A allele of the -1438G/A polymorphism in the promoter region of the 5-HT2A receptor has been reported. However, these findings are controversial and all patients studied to date have been Caucasian. This study was designed to determine whether this association is reproducible in Japanese subjects. This case-control study of a cohort of 75 female Japanese AN sufferers and 127 normal female control subjects revealed no significant association between the 5-HT2A promoter polymorphism and AN. Thus, at least for Japanese subjects, the A-allele of the -1438G/A polymorphism in the promoter region of the 5-HT2A receptor gene does not contribute to a predisposition to AN.

Analysis of tumor necrosis factor-alpha gene promoter polymorphisms in anorexia nervosa.

Elevated plasma tumor necrosis factor-alpha (TNFalpha) levels and enhanced spontaneous TNFalpha release from peripheral blood mononuclear cells in patients with anorexia nervosa (AN) have been reported. TNFalpha activates the hypothalamic-pituitary-adrenal axis and reduces food intake, which is characteristic of eating disorders. Recently, three novel polymorphisms in the 5'-flanking region of the TNFalpha gene were reported at positions -1031 (T --> C substitution), -863 (C --> A) and -857 (C --> T). Differences in these alleles are reportedly related to altered TNFalpha-transcriptional promoter activity. Therefore, we performed a case-control association analysis to determine whether any of those three polymorphisms in the TNFalpha promoter region were involved in a predisposition to AN. The results of our analysis of a cohort of 79 female Japanese AN sufferers and 127 normal female control subjects provide no support for the hypothesis that -1031T/C, -863 C/A and -857C/T polymorphisms in the TNFalpha gene promoter region influence the susceptibility to AN.

Lactose malabsorption. Optimalization of investigational methods.

Although the prevalence of lactose malabsorption is high and the importance of this diagnosis is established in gastroenterological practice, still a variety of poorly standardized tests is used to investigate the ability of the small bowel to digest lactose. In this report the various most commonly used lactose malabsorption tests are described and compared with each other knowing that a 'gold standard' does not exist. The hydrogen breath test is the test most used in clinical practice. On the basis of recent investigations the recommendation is made to improve the accuracy of this test by prolonging it to 4 hours and by simultaneous measuring of the oro-coecal transit time.

The central inhibitory effect of interleukin-1 on gastric acid secretion.

The effect of interleukin-1 (IL-1) on gastric secretory functions was examined in pylorus-ligated conscious rats. Intracisternal (i.c.) injection of IL-1 beta (1-100 ng) induced dose-related, long-lasting inhibition of gastric acid output, which was due to the reductions of both the amount and the acid concentration of the gastric juice. A much higher dose of IL-1 alpha was required to achieve identical effects on gastric acid secretion when it was given by intravenous routes. The i.c. injection of IL-1 alpha also had an inhibition of gastric secretion. This inhibitory effect of i.c. applied IL-1 beta on gastric acid secretion was completely abolished in indomethacin-pretreated animals but not in reserpine-pretreated ones. These results suggest that IL-1 may have an inhibitory action on the regulation of gastric secretory functions by its central action which is dependent on the eicosanoid metabolism.

[Evaluation of super-elasticity characteristics of orthodontic Ni-Ti alloy wire].

Mechanical properties of orthodontic Ni-Ti alloy wires, especially their super-elasticity characteristics, were investigated in order to use them effectively in the clinics. In this study, twenty commercial orthodontic Ni-Ti wires were tested and two parameters were devised for the load-deflection curve and introduced to evaluate the practical mechanical properties, one being the "super-elastic index: SEI" for super-elasticity, and the other being the "E-load" for the load in the super-elastic region. There was a "flat area" in the load-deflection curve of the super-elastic type of Ni-Ti alloy wires, whose SEI was above seventy and a continuous force could be used through the wide range of deflection. The load of the work-hardened type of Ni-Ti wires, SEI below thirty, was nearly proportional to the deflection. And the middle type showed intermediate properties of the two. The E-load values of the super-elastic type wires are thought to indicate the load level in the super-elastic region. The quantitative evaluation of the super-elasticity characteristics of the orthodontic Ni-Ti wires became possible by the introduction of these parameters.

The super-elastic Japanese NiTi alloy wire for use in orthodontics. Part III. Studies on the Japanese NiTi alloy coil springs.

Closed and open Japanese nickel titanium (NiTi) alloy coil springs were fabricated from the Japanese NiTi alloy wire. The closed coil springs were subjected to a tensile test and the open coil springs were subjected to a compression test to evaluate the mechanical properties. At the same time, a test with the commercially available steel coil springs also was done. It was clearly established that the Japanese NiTi alloy coil springs exhibited superior springback and super-elastic properties similar to the properties of the Japanese NiTi alloy arch wires. In addition, it was shown that the load value of super-elastic activity can be effectively controlled by changing the diameter of the wire, the size of lumen, the martensite transformation temperature, and the pitch of the open coil spring. The most important characteristic of the Japanese NiTi alloy coil springs is the ability to exert a very long range of constant light, continuous force. It is possible to use this coil selectively to obtain optimal tooth movement.

[Interferon production in vitro].

Interferon production in vitro was induced by poly I: C and PHA in leukocytes obtained from patients with cancer, benign diseases and control subjects. The interferon response per lymphocyte was relatively constant in all groups. However, interferon production was slightly inhibited in patients receiving cancer chemotherapy. The peak response of interferon occurred at 48 hours after the initiation of the combined culture in each specimen. From our observations, it was suggested that interferon production in vitro seems to be useful marker for evaluation of effect of chemotherapy and or immunotherapy, and, furthermore, in the treatment of cancer patients with interferon inducers, there seems to be optimum BRM dose and optimum timing of administration.
