Preparation and some properties of human serum kallikrein immobilized on ARM-Sepharose.

A highly purified human serum kallikrein immobilized on CH-Sepharose 4-B was obtained. KM values for N-benzoyl-L-arginine ethyl ester and N-tosyl-L-arginine methyl ester hydrolysis of this preparation were 1.10 x 10(-3) M and 3.6 x 10(-4) M, respectively; pH optimum of hydrolysis of these esters were found to be 8.2 and 8.5, respectively. The immobilized kallikrein possessed kininogenase activity and was capable of activating prekallikrein.

In vivo activity of spleen cells from untreated syngeneic mice against Ehrlich ascites carcinoma.

Inbred CFW/L1 or BALB/c male mice were injected intraperitoneally with 1 X 10(5) EAC cells. They died in 98.6 or 100%, respectively, within 100 days of observation. Intraperitoneal injection of 6 X 10(7) syngeneic spleen cells from 6--9-week-old CFW/L1 or BALB/c donors 2.5--4 h after tumour inoculation significantly prolonged MST and 16.7 or 23.5% of mice, respectively, survived for 100 days. Smaller amounts of spleen cells, thymus cells, mixed spleen and thymus cells, or spleen cells from older donors did not show this effect. Homogenized spleen cells were also inactive. Antitumour activity of spleen cells was still evident when they were isolated from irradiated (2 000R) donors or were given 24 h after tumour inoculation. Spleen cells used for in vivo studies showed spontaneous cytotoxic activity in vitro against EAC cells in the 51Cr-release cell-mediated cytotoxicity test. The possible participation of NK cells in the observed in vivo antitumour activity of normal spleen cells is discussed.

Changes in the relative amount of surface antigens on the living cells after treatment with insoluble protease derivatives.

Relative amount of surface antigen was compared on L 1210 leukaemia cells treated with soluble or insoluble derivatives of trypsin and papain. Trypsin or trypsin insoluble derivative do not change the amount of antigen significantly as compared with control. However, papain insoluble derivative decrease the relative amount of antigen within 45 min to the value of 0.43 or 0.55 respectively as compared with the control specimen.

Radioiodination of L 1210 cells.

The lymphoid leukaemia L 1210 cells of mice were labelled with 125I. The cell homogenates were fractionated and from the microsomal fraction 90 per cent of the radioactive material could be precipitated with perchloric acid, whereas only 4 per cent was precipitated from the soluble fraction. Papain bound with Enzacryl AH released 31 per cent of radioactivity. It was concluded therefrom that the surface proteins of the cells were labelled. Electrophoretic separation of these proteins in polyacrylamide gel with sodium dodecyl sulphate was performed and 6--8 radioactive fractions of surface peptides were found.

Cytochemical characterization of mouse L1210 leukemia.

Mouse lymphatic leukemia L1210 cells were characterized with polyclonal and monoclonal antibodies and lectins. The cells were found to have the phenotype IgG-, Thy-1.2-, Lyt-1-, Lyt-2-, asialo-GM1-, TL-, I-Ad-, IL-2R+, peanut agglutinin+, and Helix pomatia lectin +/-. They retained expression of H-2Kd and H-2Dd. Thus, these cells resemble "null" cells.
