[Effect of molsidomine on the regional distribution of cardiac output in the anaesthetized dog (author's transl)].

In anaesthetized dogs blood pressures were measured in the following parts of the circulation: A. femoralis, aorta, left ventricle and A. pulmonalis. By means of particle distribution technique blood flow was measured in 25 different organs. By additional determination of arterio-venous O2 difference (AVDO2) between heart and brain oxygen consumption could be calculated. All data were measured before, 15, 30, 45 and 60 min after injection of 0.05 or 0.1 mg/kg, respectively, N-carboxy-3-morpholino-sydnonimine ethylester (molsidomine, Corvaton) i.v. 1. Molsidomine reduces blood pressures in all parts of the circulation measured. The duration of effects exceeds the experimental period. 2. The compound is no coronary vasodilator in the dose range selected, no dilation of cerebral occurs. Blood flow in kidneys remains constant. Blood flow values of all other organs do not give any indication of an essential peripheral dilatory effect of the drug. 3. Oxygen consumption of left ventricle is significiantly reduced by 25% with molsidomine (0.05 mg/kg i.v.). Oxygen consumption of brain remains constant. 4. All data are in accordance with the hypothesis that molsidomine exerts its effect predominantly by acting on capacitance vessels (venous pooling) of the circulation.

[Autoradiographic and scintillationspectrographic studies of 14C-zolimidine in rats after i.v. and oral application (author's transl)].

The relative distribution of radioactivity after i.v. (5 mg/kg) and oral (25 mg/kg) application of 14C-Zolimidine [2-(p-methyl-sulfonylphenyl)-imidazo-(1,2-a)-pyridine-2-14C] was examined in male rats by whole-body autoradiography and scintillation fluid spectrometry. 14C-Activity was remarkably concentrated in the stomach of i.v. treated animals, probably as a result of secretion from the pyloric and/or fundic part of the mucous membrane. 14C-Zolimidine also accumulated in the aortic vascular walls, the adrenal gland, and in the excretory organs, liver, kidney, and intestine, after both routes of drug administration. Much less radioactivity could be measured in brain and spinal cord. The estimation of nearly 80% gastrointestinal absorption of 14C-zolimidine and the suggestion of one or more metabolites were in accordance with previously reported results. The elimination of radioactivity from brain occurred more rapidly than from other organs. No striking results were found in the reproductive organs of the rats.

New fuzzy logic strategies for bio-molecular recognition.

The concepts of molecular similarity and molecular complementarity, playing important roles in the broad field of molecular recognition, are chemical problems, in which the eyeball technique used by a human observer is very successful but which are very hard to code into a computer algorithm. Based on the model of molecular surfaces, our new approach defines overlapping surface patches with similar molecular properties. These patches are used to represent local features of the molecule in a way, which is beyond the atomistic resolution but can nevertheless be applied in partial similarity as well as complementarity analyses in a very general sense. It is shown that this molecular description can be used as the first step in a docking algorithm for complexes, where the structures of both molecules are known, as well as for the identification of possible active sites without the knowledge of specific molecules binding to this site.

Glutamic acid concentration in human semen--its origin and significance.

Glutamic acid content of semen was determined photometrically in over 400 semen specimens. Glutamatic acid content was found to increase proportionately to temperature in the first few hours post ejaculation. The glutamate estimation should therefore be performed on semen at 30 min. after ejaculation. The frequency distribution of glutamic acid concentration with 400 unselected specimens, 50 "normozoospermias" as well as 42 azoospermias is shown. The mean value of normozoospermias was 10.6 mg% (+/- 4.6 mg%) that of azoospermias 7.7 mg% (+/- 4.7 mg%). There was no correlation found between glutamic acid content and pH value. A correlation however, was demonstrated between glutamic acid and following: sperm count, citrate concentration, gamma-GT and carnitine. No correlation to fructose content was detectable. That suggests that glutamate gets to the ejaculate with the secretions from the prostate gland and epididymis whereas the seminal vesicles do not play a role in the level in the total semen.
