Contraction followed by relaxation in response to hypoxia in the sheep isolated middle cerebral artery.

In rings of sheep middle cerebral artery, precontracted with 5-hydroxytryptamine (5-HT) 10 microM, hypoxia (pO2 = 10 mm Hg) caused contraction and re-oxygenation caused a transient further relaxation. The hypoxic contraction was abolished and the re-oxygenation relaxation was reduced by removal of the endothelium. BW755C 20 microM reduced the hypoxic contraction and re-oxygenation relaxation. Haemolysate 1 microliter/ml and methylene blue 10 microM reduced the hypoxic contraction and re-oxygenation relaxation and enhanced the hypoxic relaxation phase. It is concluded that the endothelium of isolated rings of sheep middle cerebral artery releases vasoconstrictor substances during hypoxia and releases vasodilator substances during re-oxygenation.

Nucleotide sequence of a cDNA coding for the NADPH-protochlorophyllide oxidoreductase (PCR) of barley (Hordeum vulgare L.) and its expression in Escherichia coli.

The primary structure of the NADPH-protochlorophyllide oxidoreductase of barley has been deduced from the nucleotide sequence of a cloned full-length cDNA. This cDNA hybridizes to a 1.7 kb RNA whose steady-state level in dark-grown seedlings is drastically reduced upon illumination. The predicted amino acid sequence (388 residues in length) includes a transit peptide of 74 amino acids whose end point has been delimited by sequencing the N-terminus of the mature protein. Expression of the cDNA in Escherichia coli leads to the synthesis of an enzymatically active precursor of the NADPH-protochlorophyllide oxidoreductase. Activity of this protein in bacterial lysates is completely dependent on the presence of NADPH and protochlorophyllide and requires light.

DNA Methylation is Reduced in DNasel-Sensitive Regions of Plant Chromatin.

Pea, barley, and corn chromatin was isolated and subjected to limited digestion with DNasel. The preferentially degraded, small molecular weight fraction, presumably consisting of sequences in an active state of expression, was isolated and its 5-methylcytosine content determined. The DNasel-sensitive chromatin fraction from all three plant species investigated contained a markedly reduced level of DNA methylation compared to total DNA.
