Effect of interferon and poly I: C on DNA fiber propagation in L 929 cells.

Autoradiographic analysis of DNA fiber replication was used to measure the rate of DNA propagation, the distances between initiation points of adjacent replicons and the direction of DNA propagation in L929 cells treated with either interferon or poly I: C, or untreated. Poly I:C ceased by 44% the rate of DNA propagation while the interferon had no effect. Poly I:C but not interferon reduced the mean distance between initiation points by 32%.

Effect of streptovirudin on parainfluenza 3 virus multiplication.

Streptovirudin (SV) in a concentration of 100 microng/ml reduced the multiplication of parainfluenza 3 virus by about 3.5 log units. When uninfected WISH cells were treated for 2 and 12 hours with 100 mug/ml of SV, 3H-thymidineincorporation was decreased, but no marked differences were observed in the incorporation of 3H-uridine. The level of intracellular cyclic adenosine monophosphate (c-AMP) was found to decrease in SV-treated cells.

Effect of poly I:C on cell proliferation in vivo.

Female Swiss albino mice were injected intraperitoneally with poly I:C1 microgram/g body weight. The animals were killed 2, 12, and 24 hr after the injection of poly I:C. Half an hour before the death of animals they were reinjected intraperitoneally with 3H-thymidine. Fragments of ileum were excised. The quantification of autoradiographs included the counting of labeled cells, mitotic cells as well as the recording of cell position along the ileal crypts. The spatial distribution of both the mitotic and DNA-synthesizing cells of colonic crypts were affected by poly I:C at neither of time intervals between the injection and the death of animals. On the other hand, the parameters of cellular proliferation, i.e. mitotic and labeling indices, decreased significantly 12 and 24 hr after the injection of poly I:C, being unaffected 2 hr after the injection.

Purification of Clostridium toxoids.

A two-step fractionation procedure was applied for purification and concentration of the individual Clostridium toxoids. The toxoids were precipitated with hydrochloric acid in the presence of sodium sextametaphosphate, then antigenic fractions were separated from inactive contaminants by Sephadex G-75 filtration. Specific activity of the preparations thus obtained, as determined by Mancini radial immunodiffusion, was 150--565 binding units per mg of protein nitrogen for Clostridium perfringens toxoid, 204--352 binding units for Clostridium oedematiens toxoid and 26.6 -- 51.2 binding units for Clostridium septicum toxoid.

An attempt to determine the relation between levels of IgG, IgA and IgM immunoglobulins and titers of antibodies against RS virus.

In a group of 126 children aged 5-24 months with respiratory infections, the relation between levels of IgA, IgG and IgM immunoglobulins and titers of antibodies against the RS virus were studied. In children aged 5-12 months with high titers of antivirus antibodies, elevated levels of IgG were more frequent, but high antibody titers were not related to IgA and IgM immunoglobulins.

Effect of prostaglandins on cell function and multiplication of parainfluenza 3 viruses in WISH cells.

Cytotoxic effect of prostaglandins E2 and F2alpha on cells grown in vitro and the influence of these compounds on multiplication of myxovirus parainfluenza 3 were investigated. The prostaglandins were added to culture medium (0-01-10 mug/ml) 24 hr before virus infection, or for 2 and 48 hr after inoculation with viruses. WISH cells and monkey kidney cell cultures were used. No direct cytotoxic effect of prostaglandins at concentrations 0-01-1 mug/ml was found (viability, supravital staining, phase-contrast system, Nitro-BT reduction and succinic dehydrogenase tests), whereas the concentration of 10 mug/ml within 48 hr led to reduction and succinic dehydrogenase tests), whereas the concentration of 10 mug/ml within 48 hr led to partial injury of the cell population with symptoms of damage to mitochondria. Prostaglandins E2 and F2alpha inhibited multiplication of parainfluenza 3 virus at concentrations 0-1-10 mug/ml. The inhibitory effect was most pronounced if prostaglandins were added to medium for the whole period of virus multiplication i.e. for 48 hr but little or no effect was found if they were added prior to inoculation or for 2 hr after it. Inhibitory effect of prostaglandins on replication phase of viruses is suggested.

Growth of rubella virus in primary rabbit kidney cell cultures maintained with and without calf serum.

Rubella virus growth was tested in primary rabbit kidney cell cultures, maintained with or without calf serum. In fluid phase of the cultures maintained with 1.5% of heat-inactivated calf serum, the infectious virus titers were much higher than in the system without serum. The differences reached 1.5-2.0 log10 TCID50/ml. Virus multiplication curves also ran different courses. In serum-containing system virus titer increased up to the 9th day post infection, while without serum--only up to 4-5 days. The time of appearance and the type of cytopathic effect were not affected by the absence of serum or by its presence in a concentration of 1.5%.

Inhibition of multiplication of parainfluenza 3 virus in prostaglandin-treated WISH cells.

Influence of prostaglandins E2 (PGE2) and F2alpha (PGF 2alpha) on multiplication of myxovirus parainfluenza 3 was investigated. At concentrations of 0.01--1 mug/ml prostaglandins had no direct cytotoxic effects. PGE2 and PGF2alpha inhibited multiplication of parainfluenza 3 virus at concentrations 0.1--10 mug/ml. The inhibitory effect was most pronounced when the prostaglandins were added to medium for the whole period of virus multiplication (48 hours) while little or no effect was found when prostaglandins were added before virus inoculation or for 2 hours after infection. It is suggested that prostaglandins exert an inhibitory effect on the replication phase of the virus by influencing the growth of the WISH cells.
