Characterization of the alpha-amylase-encoding gene from Thermomonospora curvata.

The nucleotide sequence of a 3007-bp DNA fragment from Thermomonospora curvata CCM3352 containing the coding and regulatory region of the alpha-amylase-encoding gene (tam) was determined. Primer extension mapping was used to determine the 5' end of the transcript, and it was demonstrated that the gene is transcribed from a unique promoter which is also functional in Streptomyces lividans TK24. Transcription of tam in T. curvata was induced by maltose, even in the presence of glucose. In S. lividans, tam was expressed constitutively. The deduced amino acid sequence reveals a considerable similarity with alpha-amylases from streptomycetes.

[Lactic acid production and urease activity in strains of Enterococcus faecium found in the rumen and their genetic stability].

Lactic acid production, urease activity and genetic stability were investigated in five selected rumen strains of Enterococcus faecium. The average value of urease activity in the tested strains was 16.5 +/- 0.953 nkat per ml, two strains were urease-negative. The values of E. faecium strains produced lactic acid ranged from 1.087 +/- 0.134 to 1.787 +/- 0.213 mmol per 1 l. Cultivation of the strains in ethidium bromide (EB) eliminated urease activity of these strains already in the first subculture, but the elimination effects of sodium dodecyl sulphate (SDS) and acridine orange (AO) were manifested later on (1-8 subcultures). Lactic acid production was eliminated in all strains from 1st to 8th subculture after cultivation in SDS, EB and also AO.

Physical properties and biochemistry of lung surfactant following vagotomy.

Surface tension and the biochemical composition of lung washings were studied in rats 24 h after bilateral cervical vagotomy. A significant decrease in body weight, rate of breathing, stability and hysteresis shape indexes and hysteresis loop areas were noted. The gamma max, gamma min, gamma stat, indexes of compressibility, the number of cells and the protein content of washings were significantly increased. There were only minor changes in the absolute amount of phospholipids but their relative rate was significantly lower. Interstitial lung edema and early bronchopneumonia were confirmed histologically.

[Surface tension in rat lungs after quartz dust inhalation (author's transl)].

The influence of quartz dust (1000mg/m3) on surface tension in rat lungs was investigated. The exposure-times were 5, 15 and 30 hours. The animals were killed, 1, 7, 14, 28 and 90 days after quartz dust exposure and the surface tension was measured in a Wilhelmy balance. There was no significant difference between control animals and quartz dust animals in gammamax, gammamin and S. But there were significant differences in the same groups in hysteresis shape index and compressibility-coefficient.

Pattle's bubble-stability method in the rat.

The following report deals with Pattle's alveolar bubble-stability method for surfactant studies in rats. There was no difference in stability ratio of bubbles obtained from separate lung lobes without gross pathological lesions. Bubble stability was not influenced by microscopic illumination (used only briefly for photographing) and by 1-day storage at plus 4degrees C. The stability ratio was significantly lower in distilled water in comparison with the value obtained in isotonic saline. If the time interval passing from squeezing of the bubbles to the time of the first measurement is strictly retained, the reliability of the method is satisfactory. The use of cummulative distribution curves of individual stability ratios' values and non-parametric testing of such results is recommended.
