Coupling between reduced nicotinamide adenine dinucleotide oxidation and metabolite transport in renal brush border membrane vesicles.

In a previous communication [Giménez-Gallego, G., Benavides, J., García, M.L., & Valdivieso, F. (1980) Biochemistry (preceding paper in this issue)] we have reported the occurrence of a NADH oxidase activity in the renal brush border membranes. The brush border membranes can utilize the energy from the oxidation of NADH to drive the transport of amino acids (aspartic and glutamic acids), organic acids, and the lipophilic cation tetraphenylphosphonium (TPP). The coupling between NADH oxidation seems to be due to the formation of a proton electrochemical gradient (delta-mu H+) as indicated by the effect of specific ionophores. This system may be implicated in the reabsorption process in the renal tubules and in the maintenance of the delta-mu H+ (positive and acidic in the luminal side) previously described in the renal tubules "in vivo".

Size and distribution of polyadenylic acid sequences in Drosophila polytene DNA and RNA.

[3H]Poly(U) hybridizes very rapidly to polytene DNA from Drosophila hydei. When hybridization is performed at 30 degrees C in 2 X SSC to a large excess of DNA, 95% of the poly(U) becomes ribonuclease resistant. Also, complementary RNA transcribed in vitro from polytene DNA hybridizes to poly(U). 023--0.25% of the DNA is composed of (dA)-rich sequences and 0.23--0.31% of cRNA hybridizes to [3H]poly(U). The length of the (dA)-rich sequences on the DNA and cRNA is 40 nucleotides. The Tm values of these hybrids formed between DNA or cRNA-poly(U) is 45 degrees C. The poly(A) fragments from cytoplasmic RNA ranged from 80 to 170 nucleotides in lenght, and migrated in polyacrilamide gels as a broad peak. The average sizes of the poly(A) fragments from the poly(A)-containing RNA transcribed by nuclei isolated from salivary glands in vivo or in vitro were 40, 70, 170 and 70 nucleotides, respectively. Hybridization in situ of [3H]-poly(U) to chromosome squashes indicated that the (dA)-rich sequences are randomly distributed over the whole genome.

In vivo survival rate of rabbit morulae after vitrification in a medium without serum protein.

The in vivo survival rate of rabbit morulae after vitrification in a mixture of dimethyl sulphoxide and ethylene glycol solution without protein supplement (WPS) was compared with two types of protein supplements: rabbit serum (RS) and bovine serum albumin (BSA). Significant differences were observed in the percentage of transferable embryos (undamaged embryos after devitrification, 80.4% versus 93.2 and 92.1%, WPS, BSA and RS, respectively, P < 0.05) and live born rate (40.9% versus 56.1%, WPS and BSA, respectively, P < 0.05). Non-significant differences were, however, observed in the percentages of implanted embryos at 12 days post-ovulation induction (56.7, 69.7 and 68.6%), post-implantation survival rate (82.3, 74.2 and 77.2%) and live born rate in pregnant does (54.6, 56.1 and 50.5%) with different vitrification media (RS, BSA and WPS). We conclude that rabbit embryos can be vitrified and stored using protein-free vitrification medium with moderate losses of viability.

Cyclophosphamide-loaded nanospheres: analysis of the matrix structure by thermal and spectroscopic methods.

This study reports on the preparation and evaluation of cyclophosphamide loaded-polyalkylcyanoacrylate nanospheres obtained by emulsion polymerization. Characterization by differential scanning calorimetry, infrared spectroscopy and X-ray diffraction can reveal state dispersion of the drug inside the nanospheres. Such information predicts the stability of the particles and the drug release behaviour. The study has indicated the prescence of a molecular dispersed system. The drug release behaviour was also studied.

Aceclofenac-loaded poly-epsilon-caprolactone nanocapsules. Effect of coadjuvants on morphometrical properties and drug entrapment.

The physicochemical properties of aceclofenac nanocapsules, prepared by interfacial precipitation of poly-e-caprolactone at the oil/water interface, have been studied. A Central Composite Design was used to investigate the influence of polymerization adjuvants on these properties on the elaboration of aceclofenac-loaded poly-e-caprolactone nanocapsules. In this way, the effect of polymer, oil and drug concentrations in organic phase on the size and encapsulation efficiency has been analyzed. The optimized nanocapsule formulation leads to use an initial concentration of drug (aceclofenac) of 0.8 mg/ml and 0.6% of oil (Miglyol 812), being the polymer and surfactant concentrations previously fixed.

[Rheumatic activity in patients with heart valve replacements].

We made a comparative analyses of the clinical, laboratory and histological findings between two groups of patients: Group A was composed of rheumatic patients operated upon for valvular substitution. Group B included patients who were taken to cardiac surgery for non rheumatic reasons.

[Psychometric characteristics of the Spanish version of the Wender++-Utah Scale of retrospective evaluation of ADHD].

INTRODUCTION: The diagnosis of attention-deficit hyperactivity disorder among adolescents and adults, when they were not diagnosed during childhood, requires retrospective evaluation of the disorder. Parents are usually the best source of information about childhood conduct, but sometimes it is impossible to get them. The Wender-Utah Rating scale was developed as an aid in the retrospective evaluation of symptoms of attention-deficit, hyperactivity e impulsivity. The objective of this study was to determine the consistency and the validity of this questionnaire in Spanish. METHOD: We evaluated medical students of the Universidad Autónoma de Puebla in Mexico with the Wender-Utah Questionnaire-Spanish. Some of them were evaluated again fifteen days later. We gave them the parents Rating Scale. We calculated the internal and the test-retest consistency and also the correlation between this Questionnaire and the Parents Rating Scale. RESULTS: We evaluated 946 students, 400 had the test-retest procedure and 532 returned the Parents Rating Scale. The internal consistency was superior to .80. The correlation symptoms of attention deficit, hyperactivity and impulsivity. The test-retest consistency was at least .80, for the evaluation of with the Parents Rating Scale were moderate. CONCLUSIONS: This Spanish translation of the Wender-Utah Questionnaire has adequate consistency and reproducibility so it can be used as a screening tool, also the validity of the different versions suggested can be evaluated.
