Polymorphism of HLA-DRB1 gene shows no strong association with ulcerative colitis in Chinese patients.

The genetic factors predisposing to ulcerative colitis (UC) have remained totally unclear to date. This study aimed to investigate the role of HLA-DRB1 genetic polymorphism in the susceptibility to develop UC in Chinese patients. HLA-DRB1 genotyping was carried out in 72 unrelated patients with UC and 314 healthy controls by using polymerase chain reaction-sequence-specific primers (PCR-SSP). All of the patients and healthy controls are Han people in China. The frequency of DRB1*07 allele was increased in UC patients compared with healthy controls (19.4% vs. 9.2%, P = 0.0229, OR = 2.372, 95%CI: 1.181-4.766), but the significance disappeared when given Bonferroni correction (P(C) = 0.2977). Furthermore, compared with healthy controls, although HLA-DRB1*07, DRB1*16/DRB1*09 and DRB1*07/DRB1*12 genotypes were increased in frequency in the patients with extensive colitis, and the patients without extraintestinal manifestations (EIMs) carried an increased frequency of HLA-DRB1*07 and DRB1*07/DRB1*12 genotypes, these differences did not reach statistical significance after Bonferroni correction. HLA-DRB1 alleles showed no strong association with UC, and no HLA-DRB1 alleles or genotypes were strongly associated with clinical subgroups of UC in Chinese patients.

[Clinical significance of the expression of lung resistance protein in non-small cell lung carcinomas].

OBJECTIVE: To investigate the expression of the lung resistance-related protein (LRP) in non-small cell lung carcinomas (NSCLC) and evaluate its clinical significance. METHODS: Using immunohistochemistry, LRP was examined in 69 NSCLC specimens obtained by either transbronchial biopsy via fibrobronchoscopy or transcutaneous needle biopsy or surgical resection. Of the 69 patients from whom tumor specimens were obtained, 52 were male and 17 were female. RESULTS: LRP expression was detected in 39/69 (57%) tumor specimens. There was no correlation between LRP expression and NSCLC histological classification (P > 0.05). No significant differences were found in gender and among age groups. The positive rates of LRP were 67% (18/27), 55% (17/31), 52% (23/44), 64% (16/25), 83% (5/6), 57% (36/63) and 50% (3/6) in adenocarcinoma, squamous, staging T1-2, T3-4, N3, M0 and M1 respectively. It was shown that LRP expression was not associated with primary tumor size and metastasis status (N,M). NSCLC patients with positive LRP expression responded more poorly to chemotherapy than those with negative LRP expression (P < 0.05). CONCLUSIONS: The frequencies of multidrug resistance caused by LRP are similar between adenocarcinoma and squamous. Expression of LRP is related to multidrug resistance of NSCLC, which in turn related to the efficacy and prognosis of chemotherapy. The examination of LRP expression may be valuable for choice of chemotherapy regimen.

[Analysis of the clinical salience for sudden hearing loss in acoustic neuroma].

OBJECTIVE: To avoid misdiagnosis and misapplied therapy of acoustic neuroma. METHODS: 92 cases of acoustic neuroma treated between 1983 and 1997 were retrospectively reviewed. Among them, 20 patients (21 ears, 21.7%) presented with sudden hearing loss as the starting symptoms. The diagnosis was based on audiological and radiologic examinations. RESULTS: The clinical and audiological analysis demonstrated that 57.1% of the ears showed hearing loss over 71 dBHL, and all ears had deteriorated ABRs. Acoustic reflex was nonreactive in all 10 ears tested. Positive findings in CT scan were 88.89%. CT pneumoencephalography or MRI provided useful information of diagnosis in those with negative findings of place CT scan. CONCLUSION: ABR should be used as a routine test for patients with sudden deafness. When ABR was abnormal, CT scan around the internal auditory meatus was needed.

[Studies on respiratory phase switching effects of Bötzinger complex].

The effects of electrical stimulation of Bötzinger complex (Bot.C) on respiratory rhythm were investigated in 40 urethane anesthetized adult rabbits. The results were as follows. (1) A short train stimulation delivered in the early inspiratory phase produced a transient inhibition of phrenic discharge. The stimulus, when delivered in the mid or late inspiratory phase, could cause a premature termination of the inspiratory phase ("inspiratory off-switch") and a switching to the expiratory phase, which was accompanied with the reduced duration of the consecutive expiratory phase. There was a negative linear correlation between the threshold intensity of inspiratory off-switching and delivery time of stimulation. (2) A short train stimulation delivery in the expiratory phase elicited a transient phrenic discharge. The discharge in the late expiratory phase was followed by a premature onset inspiration. This effect was also dependent on the strength and delivery time of the stimulus. The results suggest that the Bot.C is involved in the central control of respiratory phase-switching.

[Selectivity tuning in multi-binary eluents for reversed-phase liquid chromatography (RPLC)].

In this article, the retention equation and the relationship between retention parameters and the parameters of molecular structure deduced from statistical thermodynamics in RPLC have been used to explain the difference of selectivity towards a particular species of compounds polycyclic aromatic hydrocarbons (PAHs). Methanol/water, acetonitrile/water and isopropanol/acetonitrile have been provided in advance, then the retention behaviors of sixteen PAHs under three binary solvent systems have been investigated. It is found that each pair of binary solvents of methanol/water, acetonitrile/water and isopropanol/acetonitrile has its own unique selectivity. The best selectivity obtained for acenaphthene and fluorene is methanol/water system for fluoranthene and pyrene is acetonitrile/water, and for benzo[g,h,i]perylene and dibenzo[a,h]anthracene is isopropanol/acetonitrile. So a three-stepwise gradient elution of multi-binary mobile phase can be chosen for separation of 16 PAHs.

The effect of thyrotropin receptor antibodies on the proliferation of FRTL-5 cells and the expression of protooncogene c-fos mRNA.

OBJECTIVE: Hyperthyroidism and a diffuse goiter are the main symptoms of Graves' disease (GD) associated with autoantibodies to thyroid-stimulating hormone (TSH) receptor (TRAb). The present study was conducted to evaluate effects of autoantibodies in patients with GD (TRAb-IgG) on induction of the proliferation and c-fos mRNA expression in FRTL-5 cells (Fisher rat thyroid cell line). METHODS: Highly purified IgG fractions were isolated from 11 patients with GD, TRAb-IgG and 15 normal individuals (normal controls) with Protein A Sepharose CL-4B affinity column chromatograph. FRTL-5 cells, which had been grown to subconfluency and deprived of TSH for a few days. Then, these cells were used for measuring cAMP content, 3H-thymidine incorporation in cells and the expression of c-fos mRNA respectively. RESULTS: After stimulation of TRAb-IgG, the cAMP production and 3H-thymidine incorporation in FRTL-5 cells were much higher than those from normal controls (P < 0.05 respectively). Using 32P labelled v-fos probe by the Northern Blot method, the expression of c-fos mRNA could be induced by IgGs from patients with GD. CONCLUSIONS: These data suggest that the stimulation of TRAb-IgG followed by cAMP production and 3H-thymidine incorporation is related to the induction of c-fos mRNA and, thus, to the growth of FRTL-5 cells.

[Effects of glycine and strychnine microinjected into unilateral and bilateral Bötzinger complex on phrenic nerve discharges in rabbits].

The effects of microinjection of glycine and strychnine into unilateral and bilateral Bötzinger complex (Böt. C) on phrenic nerve discharges were observed in 34 urethane anaesthetized, vagotomized, paralyzed and artificially ventilated rabbits. The results are as follows: (1) microinjection of glycine into unilateral Böt. C induced an increase of respiratory rate and expiratory phrenic nerve discharges; (2) microinjection of glycine into bilateral Böt. C induced non-rhythmic tonic phrenic nerve discharges with an amplitude 40%-70% of control; strychnine could competitively antagonize the effects of glycine; and (3) microinjection of strychnine into bilateral Böt. C induced a decrease of respiratory rate and the amplitude of phrenic nerve discharges, in addition to prolongation of expiratory duration while inspiratory duration remained unchanged. The above results suggest that the Böt. C plays a crucial role in initiating and sustaining expiration.

[Population kinetic program for Michaelis-Menten elimination].

The microcomputer program, PKP-ME (population kinetic program for Michaelis-Menten elimination), is presented to estimating population pharmacokinetic parameters of drug obeying Michaelis-Menten kinetics and predicting individual pharmacokinetic parameters. A data set consisting of 69 steady-state phenytoin concentration-dosage pairs from 44 out-patients was analyzed with the present program. Five population kinetic parameters and their standard deviations were given by PKPME. The capability predicting individual phenytoin dosage was assessed by the retrospective analysis of data of 44 patients and the prospective analysis of 15 patients. A good correlation existed between predicting dose and actual dose. Population correlation coefficient was equal to 0.99 at P = 0.05.
