Enzymes of the purine interconversion system in chronic lymphatic leukemia: decreased purine nucleoside phosphorylase and adenosine deaminase activity.

Activities of adenosine deaminase (ADA), adenosine kinase (AK), adenine phosphoribosyltransferase (APRT), hypoxanthine guanine phosphoribosyltransferase (HGPRT), and purine nucleoside phosphorylase (PNP), all enzymes of the purine interconversion system, were determined in lymphocytes of 25 patients with chronic lymphatic leukemia (CLL) and in 23 controls. A statistically significant decrease of PNP activities and a reduction of ADA activities at borderline levels were found in the patients, whereas for the other enzymes assayed no deviation from normal values was observed.

Purine base transport in normal and mutant erythrocytes.

The uptake of adenine and hypoxanthine in HGPRT-deficient and normal human erythrocytes was measured using a rapid filtering centrifugation technique. The transport of hypoxanthine as well as of adenine is impaired in the mutant cells. The transport of hypoxanthine into HGPRT-deficient erythrocytes differs from that into normal cells with respect to a higher accumulation capacity, to lower initial velocities and to the kinetic properties of the translocator. In addition, a higher accumulation capacity and lower initial velocities of adenine uptake could be demonstrated in mutant cells. A linkage of the purine translocator with purine phosphoribosyltransferases associated with the erythrocyte membrane is discussed.

Effect of benzoyl peroxide on skin surface lipids.

Investigations have been performed on the effect of benzoyl peroxide on skin surface lipids by direct lipid extraction with petrol ether and quantitative determination. Though there was no change after 1 week treatment, after 4 weeks a significant reduction of the casual level as well as well as the replacement sum could be observed.

[Angiotensin converting enzyem (ACE) - a blood chemistry parameter in the diagnosis of sarcoidosis].

Serum angiotensin converting enzyme (ACE) activity was studied in 50 patients with sarcoidosis (39 active, 11 inactive sarcoidosis), as well as in 50 control patients (34 chronic lung diseases, 9 Hodgkin-disease, 7 rheumatoid arthritis). There is a significant difference (p less than 0.001) of ACE-activity between sarcoidosis patients and controls, and between active (without steroid treatment) and inactive sarcoidosis. Steroid treatment apparently lowers ACE-activity in sarcoidosis, however, without evidence for clinical improvement. Increased ACE-activity was also found in a patient with primary biliary cirrhosis.

Effect of cyproterone acetate on skin surface lipids.

Skin surface lipids in female patients with acne were examined before and after treatment with the oral contraceptive SH 209 AB (2 mg cyproterone acetate and 0.05 mg ethinyl estradiol) by direct extraction with petrol ether and photometrical determination. A significant reduction in the skin surface lipids was seen, together with a remarkable alleviation of the disease.

[Comparative evaluation of methods for predicting fetal lung maturity based on serial amniotic fluid tests (author's transl)].

12 pregnant females in the last trimester were subjected to serial amniocentesis. Although there was medical indication for amniocentesis the pregnancies were normal. All samples were evaluated with four different methods. We used two physical (Wilhelmy balance and foam test) and two chemical (L/S ratio densitometrically and planimetrically measured) methods. The Curves of individual pregnancies were compared with the reference values of 109 samples of amniotic liquid of uncomplicated pregnancies. Reference values for the various techniques reported in the literature are generally based on aggregate data collected from a number of pregnant females, which are then consolidated to obtain mean values for each gestational week. We found a continuous rise of all parameters, both in the reference value and in eight individual pregnancies. Besides we found a strikingly extraordinary run of the curves in four individual pregnancies. Here we recognized a rapid ascent and descent of surfactant concentration within short periods, which were not identical with the reference values. All four methods showed results with a statistical significant conformity, only the foam test showed a little lower significance.

[Extracorporeal uric clearance for the demonstration of in vivo bound uric acid].

Although numerous in vitro experiments demonstrated a variable degree of urate bound to human serum albumin and other macromolecules of human plasma, only few data are available on in vivo bound urate. Extracorporal clearances of uric acid, urea nitrogen, creatinine and phosphorus were performed at the beginning, the midtime and the end of 10 hours of hemodialysis. A continuous decrease of uric acid clearance was observed in ratio with the capacity of the artificial kidney used. In contrast to uric acid the clearances of urea nitrogen, creatinine and phosphorus remained constant during the time of dialysis. From the fall of uric acid clearance, the authors conclude a partial binding of uric acid in human plasma.

Lipid composition of the rat kidney.

The lipid fractions free fatty acids, triglycerides, cholesterol, cholesterol esters and phospholipids of rat kidney cortex, outer and inner medulla were analyzed by thin-layer chromatography. The concentrations of 23 individual fatty acids in the lipid fractions of each zone were determined by gas chromatography; 14 of them were evaluated by an analysis of variance. For most of the lipid fractions a corticomedullary concentration gradient was found, the highest concentrations occurring in the cortex. The lipid classes primarily determined the pattern of individual fatty acid concentrations. A significant concentration difference between the three zones only existed for 4 of 14 fatty acids. Long chain fatty acids were found in a higher concentration in the phospholipids of cortex and outer medulla (about 95%) in comparison to the inner medulla (65%).

Influence of surface hapten density of sheep red blood cells on the hemolytic or blocking activity of IgG antibodies.

The hapten density on the surface of sheep red cells, coated with benzylpenicilloic acid (BPO) was determined. This was achieved by comparison of the inhibitory capacity of the test cells with that of a standard cell preparation of known antigen density. IgG-induced hemolysis was much more affected by the recution of haptenic sites/cell than IgM-induced hemolysis. It was concluded that more than 18,000 protein islets on the sheep red cell surface bearing at least four haptenic sites were necessary for IgG-induced lysis. Target cells with 190,000 BPO groups/cell could be protected by anti-BPO-IgG against the lytic effect of simultaneously added IgM. Using target cells with 240,000 haptenic groups/cell or more a synergistic hemolytic action of IgG-and IgM-anti-BPO antibodies could be observed.
