Regulation of tRNA methyltransferase activities by spermidine and putrescine. Inhibition of polyamine synthesis and tRNA methylation by alpha-methylornithine or 1,3-diaminopropan-2-ol in Dictyostelium.

Inhibitors of polyamine synthesis (alpha-methylornithine and 1,3-diaminopropan-2-ol) were used to study the relationship between polyamine synthesis and specific methylations of tRNA in Dictyostelium discoideum during vegetative growth. Polyamine concentrations were found to be 10 mM for putrescine, 1.6 mM for spermidine and 7 mM for 1,3-diaminopropane throughout the growth stage. On treatment of growing amoebae with alpha-methylornithine or with 1,3-diaminopropan-2-ol (each at 5 mM), the syntheses of putrescine, spermidine and 1,3-diaminopropane were arrested within 4h. After polyamine synthesis had ceased, the incorporation of methyl groups into tRNA was considerably decreased under conditions that had no effect on the incorporation of uridine into tRNA, or on net syntheses of protein and of DNA. The following nucleosides in tRNA were concerned: 1 methyladenosine, 5-methylcytidine, 7-methylguanosine, 2-methylguanosine, N2N2-dimethylguanosine and 5-methyluridine (ribosylthymine). The corresponding tRNA methyltransferases, determined in Mg2+-free enzyme extracts, proved to be inactive unless polyamines were added. Putrescine and/or spermidine at concentrations of 10 mM or 1-2 mM respectively stimulate the transmethylation reaction in vitro to a maximal rate and to an optimal extent at exactly the same concentrations as found in vegetative cells. In contrast, 1,3-diaminopropane, which is formed from spermidine, does not affect the methylation of tRNA in vitro at physiological concentrations. Putrescine and/or spermidine stabilize the tRNA methyltransferases in crude extracts in the presence but not in the absence of the substrate tRNA. The results support the view that S-adenosylmethionine-dependent transmethylation reactions can be regulated by alterations of polyamine concentrations in vivo.

Measurements of polyamines and their acetylated derivatives in cell extracts and physiological fluids by use of an amino acid analyzer.

A fast and sensitive method for the determination of free polyamines and their acetylated derivatives is presented. The separation is carried out on a Durrum DC-6A cation-exchange resin with an automated amino acid analyzer. The determination is based on a step wise elution with a sodium chloride-sodium citrate buffer system. Detection is done by fluorescence of the o-phthaldialdehyde-polyamine conjugates. The sensitivity is in the picomole range. No prior purification step is needed. The method has been applied to cell extracts and urine samples.

Synthesis of transfer ribonucleic acids with uridine or 2'-O-methylribothymidine at position 54 in developing Dictyostelium discoideum.

In amoebae of Dictyostelium discoideum the ribothymidine (rT) content of tRNA is 0.9 mol%, but decreases progressively during development into spores. To elucidate which nucleosides replace rT at position 54 in developmental tRNA we have characterized 'vegetative' and 'developmental' tRNAs from the slime mould. Specific tRNAs were separated by two-dimensional gel electrophoresis. During early developmental stages, all tRNA species that could be separated by this method were newly synthesized. A new tRNA with uridine in place of rT and having an electrophoretic mobility similar to 'vegetative' tRNAAsn was detected during the early preaggregation stage. This 'development' tRNA was also extracted from purified polysomes. When development proceeds from preaggregation to postaggregation, tRNAs accumulate with 2'-O-methylribothymidine in place of rT. We suggest that these developmental tRNAs are important for the synthesis of specific developmental proteins.

Teratological study of the herbicide 4-chloro-2-methylphenoxyacetic acid in rabbits.

4-Chloro-2-methylphenoxyacetic acid (MCPA) is an aryloxyacetic acid derivative categorised as a plant hormone herbicide. The aim of the study was to evaluate the effect of MCPA on pregnant females and the prenatal development of rabbits. The substance tested was administered orally to pregnant New Zealand White rabbits from day 6 to day 27 of gestation at doses of 5, 10 and 25 mg kg(-1) day(-1). The animals were killed on day 28 of gestation and live fetuses were examined for gross, skeletal and visceral anomalies. Administration of MPCA did not induce any signs of maternal toxicity. There was a significant decrease of fetal and placental weight compared with controls at the highest dose of MPCA. No adverse effect of the substance tested was seen on uterine content variables, e.g. corpora lutea, pre-implantation and post-implantation loss, early, late resorptions, live and dead fetuses and sex ratio. Rabbit fetuses treated with the middle and highest doses of MPCA had a significantly elevated incidence of skull and pelvic bone delays. In conclusion, prenatal administration of MCPA did not exhibit a teratogenic effect on rabbit fetus development.

The product of human cytomegalovirus UL73 is a new polymorphic structural glycoprotein (gpUL73).

OBJECTIVES: This work focuses on human cytomegalovirus (HCMV) UL73, which encodes for a putative transmembrane glycoprotein that is highly conserved among herpesviruses. STUDY DESIGN: pUL73 expression was analyzed both in transiently transfected and in HCMV-infected cells using a pUL73-specific antiserum by immunoblot and immunofluorescence. Sequencing analysis from several clinical isolates and laboratory-adapted strains was also performed. RESULTS: pUL73 expressed in transiently transfected cells consists in a polypeptide of the expected size (15-18 kd) with cytoplasmic localization. In infected cells, pUL73 is expressed with true-late kinetics and localizes both in perinuclear granular formations and on the cell surface. A broad band (39-53 kd), sensitive to O-glycosidase digestion was detected in purified virus. In addition, sequence analysis showed that the N-terminal portion of pUL73 from clinical isolates is highly polymorphic. CONCLUSIONS: UL73 encodes for a new structural glycoprotein (gpUL73) expressed on the cell surface of infected cells and highly polymorphic among clinical isolates.
