Extraction and isolation of triazine herbicides from water and vegetables by a double trap tandem system.

The ability of a two-trap tandem system, one containing a nonspecific adsorbing material, such as graphitized carbon black (Carbopack B), and the other one filled with a sulfonic acid type silica-based cation exchanger (SCX), in extracting and isolating basic compounds from real matrices was evaluated by applying this device to the determination of triazine residues in water and vegetables. After percolation through the Carbopack column (extraction cartridge) of water samples or suitably prepared vegetable extracts, the two traps were connected in series, a methylene chloride-methanol mixture was allowed to flow along them, and triazines displaced from the extraction cartridge were selectively reabsorbed via salt formation on the strong acid exchanger column (isolation cartridge). After the column was washed, the analytes were removed from the isolation cartridge by 0.7 mL of aqueous methanol containing 70 mmol/L KCl. After the internal standard was added, the final solution was directly injected into the "high-performance" liquid chromatographic apparatus, which was operated isocratically in the reverse-phase mode with UV detection at 220 nm. The analytical recoveries of eight triazines from the two matrices considered ranged between 95% and 100%. The limits of sensitivity of this method for triazines were set at 10 ng/g of vegetable material and 10 ng/L of water by sampling 100 mL of it.

Glomus tumours of the hand.

Based on 8 cases of glomus tumour of the hand, observed over the last 3 years, the authors review the clinical features and pathological anatomy of this rare condition. They propose nuclear magnetic resonance as a preoperative instrumental study as an alternative to angiography because it is non-invasive, free from risk and is clinically very effective.

Recovery of post-arrest cardiac performance: effect of fructose-1,6-diphosphate in the isolated rabbit heart.

The effect of fructose-1,6-diphosphate (FDP) on the recovery of post-arrest cardiac performance was assessed in isolated rabbit heart preparations subjected to cold ischemic cardioplegic arrest (CICA). Twenty-eight hearts were perfused with Krebs-Henseleit (KHBS) solution followed by 40 minutes of CICA. After CICA normothermic reperfusion with KHBS was restarted. The experimental preparations were divided in four groups of seven hearts each: one group served as control and the other three were perfused with KHBS supplemented with FDP (500 mg/l) given before CICA, after CICA or at both study points. Left ventricular end-diastolic pressure, left ventricular systolic pressure, dP/dt and perfusion pressure were measured at different pre- and post-arrest phases. Time analysis of recovery was also performed. The results demonstrated that FDP supplied prior to and after CICA: prevents the ventricular wall rigidity induced by the ischemic arrest, improves cardiac contractile force both in the basal condition and after cardioplegia, reduces the perfusion pressure, reduces the time to recovery of cardiac contractility. Our findings in the isolated rabbit heart are consistent with published data suggesting FDP may limit the impairment of cardiac dynamics induced by ischemia and improve the recovery after cardiac arrest.

Management of cervical cancer and surgical-pathological staging (SPS). Report of our clinical case series.

FIGO staging is imprecise in a relevant number of cases of cervical cancer, especially in advanced stages, when the prognosis and the choice of the therapy are most delicate. The Authors examine their case series about the index of correction of FIGO staging after Surgical Pathological Staging (SPS). Surgical Pathological Staging was applied systematically in 788 cases and revealed errors in FIGO staging in 16% of cases at stage I; 77% at stage II; and 96% at stage III. SPS allows a more precise knowledge of neoplastic diffusion and consequently to the elimination of many false advanced stages and to adequate the treatment. Furthermore 5 year survival rate confirms the role of SPS and Surgical therapy alone or combined with Radiotherapy and Chemotherapy in every stages of diffusion of cervical cancer.

True hermaphroditism: diagnosis and surgical treatment.

We documented a new case of 46,XY true hermaphroditism in an 18-year-old patient, reared as a female at birth, with ambiguous genitalia and primary amenorrhea. At laparotomy, bilateral dysgenetic ovotestis containing gonadoblastoma were found. Karyotypes of peripheral lymphocytes and cells from tissue of both gonads were all 46,XY. Plastic surgery was conducted to transform ambiguous genitalia to the full female phenotype. Clitoreduction by glans resection and fastening of the corpora cavernosa in the labia major with enlargement of the vaginal orifice were performed. The results was a small eretcile organ with preserved sensitivity and a female capable of normal sexual activity.

Rapid determination of simazine and atrazine in water at the parts per trillion (10(12) level. Extraction by a miniaturized carbopack B trap followed by high-performance liquid chromatography.

A rapid and simple method for the determination of trace amounts of simazine and atrazine in water is described. A 250-ml volume of water is pre-concentrated by passage at a flow-rate of about 30 ml/min through a small trap containing 50 mg of graphitized carbon black (Carbopack B). After washing with 150 microliters of methanol, the two herbicides are desorbed with 700 microliters of dichloromethane-methanol (60:40, v/v). After removal, of the solvent, the extracted sample is fractionated and analysed by reversed-phase high-performance liquid chromatography with UV detection at 220 nm. A single assay can be completed within 40 min from the receipt of the water sample. Recoveries of simazine and atrazine added to 250 ml of water at the level of 50 ng/l were 97.2 and 95.8% and the limits of detection were 0.07 and 0.15 ng. respectively. At the 50 ng/l level in water, the coefficients of variation for simazine and atrazine were 3.7 and 4.0% (n = 7), respectively.

Liver cytosolic protein-bound folates in phenobarbitone-treated rats.

The effect of acute phenobarbitone treatment on the distribution of endogenous bound folates in three specific cytosolic folate-binding proteins (FBP-C), has been studied in rat liver. The bound folate amount shows no difference between treated and control rats. As it is well known that FBP's preferentially bind longer-chain polyglutamates, the content of which is markedly lower in phenobarbitone-treated rat liver, it might be suggested that shorter chain folates also bind to FBP's in these animals.

Protein bound folates in liver of castrated rats.

The effect of castration and testosterone treatment on the distribution of [3H] radioactive and endogenous bound folates in hepatic cytosolic folate binding proteins (FBP-C) has been studied in rats. The distribution of [3H] radioactive bound folates in these FBP's shows no significant difference in the three experimental group animals. On the contrary a significant decrease in the amount of endogenous bound folates is observed in castrated if compared with control rats, particularly marked for FBP-CI and FBP-CII bound folates. The testosterone treatment of castrated rats partially restores bound folate levels. The decrease of bound folates in castrated rats might be ascribable to a lower availability of longer-chain forms, almost the ones that bind to FBP's; however lower binding protein content and/or lower affinity for ligands cannot be excluded.

Folylpolyglutamate synthetase activity in regenerating rat liver.

Regenerating liver of hepatectomized rat showed marked changes in folylpolyglutamate pattern. The activity of folylpolyglutamate synthetase, investigated at various times after the operation, showed a slight increase at 12 h, reached its highest value at 36 h and, for all the time studied, remained at the upper level as compared with controls. These results may contribute to explain the higher incorporation in regenerating liver of 3H folic acid into folylpolyglutamates at a higher number of glutamate residues and is consistent with the hypothesis that the addition of glutamate represents a control step in folate coenzyme synthesis and consequently of one-carbon metabolism.

Liver folylpolyglutamate synthetase activity in folic acid-deficient rats.

To investigate the influence of folate deficiency on the addition process of glutamyl unit to folylpolyglutamates, liver folylpolyglutamate synthetase was studied. The enzyme activity appears slightly decreased in deficient liver as compared with control. This decline might be more evident in the hepatic cell owing to the lesser availability of reduced forms, preferred substrates of the enzyme.

Cervical pathology in young patients.

The authors have studied 152 patients aged between 15 and 20 year old who underwent colposcopy and colpocytology. They have compared the incidence of white colposcopic lesions with the results of the cytology. According to literature the results have pointed out that the very young patients are "at risk-patients" for serious cervical lesions and that depistage of gynecologic tumors must be done in all the other women.

Endometrial hormonal receptors as marker of benign breast disease (B.B.D.).

The problem of hormonal treatment of benign disease and mastodynia is still dividing Clinicians and Researchers. The Authors, in order to select the patients suitable to be treated, propose the assay of Estradiol and Progesterone Receptors in the most accessible hormone-dependent tissue: the Endometrium. Author's data point out a quite different behaviour of Estradiol and Progesterone cytoplasmic receptors in the Endometrium of women with B.B.D. and controls. The knowledge of this information in the single patients will release from subjectivity when deciding about B.B.D. therapy or when trying to correct unsatisfactory treatment.

The time course of [3H]PteGlu uptake and its incorporation into pteroylpolyglutamates in liver of phenobarbitone treated rats.

The uptake of an injected dose of [3H]PteGlu and its incorporation into folate forms in the liver of control and phenobarbitone-treated rats were investigated at a number of time intervals from 1 to 24 h. At 1 h the hepatic uptake of the label was quite similar in both groups of animals and radioactivity was incorporated only in pteroylmonoglutamates. At 6 h label was present also in pteroylpolyglutamate forms and radioactivity values were similar in the two groups. At longer time periods the radioactivity was lower in treated animals than in control ones. The whole data show that phenobarbitone on the one hand does not modify the transport of the vitamin; on the other, it does interfere with "pteroylpolyglutamate synthetase activity".
