Retention mechanism of L-glutamide-derived noncrystalline stationary phases in reversed-phase high-performance liquid chromatography and application for separation of nucleic acid constituents.

Double alkylated L-glutamide-derived noncrystalline stationary phases Sil-DSG and Sil-DBG have been prepared by coupling N',N''-dioctadecyl-N-[4-carboxybutanoyl]-L-glutamide (DSG) and N',N'',-dibutyl-N-[4-carboxybutanoyl]-L-glutamide (DBG) with aminopropylated silica (Sil-APS). TEM observations of DSG and DBG showed that lipid DSG can aggregate in organic solvents (methanol, chloroform, toluene, etc.) and self-assembled nano fibers are observed while such fibrous aggregations are not observed for DBG. The resulting chromatographic data have been provided information about its selective interaction with guest molecules (PAHs) in RP-HPLC. We have observed that the carbonyl groups in Sil-DSG exist in ordered state by forming a condensing thin layer over silica surface while DBG cannot form such an ordered state due to its lower order of short alkyl chain. The ordered carbonyl groups present in Sil-DSG promotes multiple carbonyl pi-benzene pi interactions with guest PAHs isomers which enhance the selectivity for these compounds. The contribution of pi-pi interactions was also supported by the substantial effects on the selectivity of benzene and nitrobenzenes. The effect of pi-electron containing solvent on the retention behavior of the PAHs was also studied. The selectivity for nucleic acid constituents, i.e. nucleosides and its bases were also evaluated by Sil-DSG and the selectivity for these compounds on Sil-DSG was compared with the selectivity of conventional polymeric ODS phase. It has been found that Sil-DSG provided higher selectivity for nucleic acid constituents than polymeric ODS and that HPLC packing materials can be efficiently employed for routine analysis of these compounds. The effect of methanol content on the separation behavior of nucleosides was also studied.

Conjugated linoleic acid and chromium lower body weight and visceral fat mass in high-fat-diet-fed mice.

More than half of the U.S. population has a body mass index of 25 kg/m2 or more, which classifies them as overweight or obese. Obesity is often associated with comorbidities such as diabetes, cardiovascular diseases, and cancer. CLA and chromium have emerged as major dietary supplements that reduce body weight and fat mass, and increase basal metabolic rate in animal models. However, studies show that CLA induces insulin resistance in mice and in humans, whereas Cr improves insulin sensitivity. Hence, we designed the present study to examine the combined effect of CLA and Cr on body composition and insulin sensitivity in a Balb/c mice (n = 10/group) model of high-fat-diet-induced obesity. CLA alone lowered body weight, total body fat mass, and visceral fat mass, the last of which decreased further with the combination of CLA and Cr. This effect was accompanied by decreased serum leptin levels in CLA-fed and CLA + Cr-fed mice, and by higher energy expenditure (EE) and oxygen consumption (OC) in CLA + Cr-fed mice. Serum levels of glucose, insulin, the pro-inflammatory cytokines, tumor necrosis factor-alpha (TNF-alpha), and interleukin-6 (IL-6), as well as insulin resistance index (IRI), decreased with CLA, whereas CLA and Cr in combination had significant effects on insulin and IL-6 concentrations and IRI. In summary, CLA + Cr decreased body weight and fat mass in high-fat-diet-fed mice, which may be associated with decreased leptin levels and higher EE and OC.

National drug policy of Bangladesh: some pitfalls in implementation.

Drug development, manufacturing and marketing have become one of the most important sectors throughout the world for various reasons. Realizing its importance, the World Health Organization (WHO) has stressed the need of a formulated drug policy in every country of the world. Bangladesh responded very early to this. Experiences over the decades have shown that the said policy could not fulfill in toto the declared objectives. Our aim is to describe some of the lacunae for which total implementation of drug policy is still struggling. For this purpose, we meticulously analysed our Drugs Control Ordinance 1982 (with an amendment brought in 1994) and upcoming National Drug Policy. We mainly focussed on three factors pointed out by WHO, which are crucial of a drug policy, which include price limitation, quality and rational use. We found great variations in price which need to be controlled. Regarding quality, safety and efficacy, we have to be more cautious. Rational use of drugs is also an area to be focussed on. Every physician should prescribe proper drug with due indication and in rational way. This pitfall of implementation might happen in many countries of the world and it needs a global discussion so that the future policy makers can mitigate the demand of the nation.

Molecular shape selectivity through multiple carbonyl-pi interactions with noncrystalline solid phase for RP-HPLC.

A new approach for the synthesis of double-alkylated L-glutamide-derived stationary phases to use in RP-HPLC is described. TEM observation of lipid distearylglutamide (DSG) showed the formation of fibrous aggregates in methanol or in chloroform through intermolecular hydrogen bonding among the amide moieties while dibutylglutamide (DBG) cannot aggregate in aqueous or organic media due to its lower order of short alkyl chain. DSG and DBG were covalently bonded to silica via amino-propyl linkages. Lipid membrane analogues (e.g., DSG) attached to the silica surface have been found in noncrystalline and solid states and can form supramolecular assemblies with specific properties based on their highly ordered structures in aqueous and organic media. 13C CP/MAS NMR and suspension (in methanol)-state 1H NMR, elemental analysis, and DSC measurements were used to characterize Sil-DSG and were compared with the three other octadecyl phases, i.e., monomeric C18, polymeric C18, and silica grafted poly(octadecyl acrylate) Sil-ODA25. The chromatographic behavior of the new RP material was investigated using detailed retention studies of planar and nonplanar polyaromatic hydrocarbons (PAHs) and nonpolar aromatic positional isomers. Aspects of shape selectivity were also evaluated with Standard Reference Materials 869a, Column Selectivity Test Mixture for Liquid Chromatography. Detailed chromatographic study revealed that Sil-DSG showed extremely enhanced molecular shape selectivity compared with the other phases investigated. The higher molecular shape selectivity obtained by Sil-DSG can be explained by a carbonyl pi (present in lipid-grafted stationary phases)-benzene pi (present in guest PAHs) interaction mechanism, and these interactions are more effective for ordered carbonyl groups.

Dioctadecyl L-glutamide-derived lipid-grafted silica as a novel organic stationary phase for RP-HPLC.

Dialkyl L-glutamide-derived lipids have been found to form supramolecular assemblies and to show specific properties based on their highly ordered structures in aqueous and organic media. To use these unique properties to create molecular recognition systems, dioctadecyl L-glutamide-derived lipid-grafted porous silica particles (Sil-DSG) were newly prepared and used as a stationary phase in reversed phase liquid chromatography. Compared with conventional ODS (octadecylated silica), the Sil-DSG column showed remarkably higher selectivity for polycyclic aromatic hydrocarbons (PAHs). Especially, Sil-DSG recognized the molecular linearity and planarity of PAHs. Suspension state 1H NMR and 13C CP/MAS-NMR spectroscopies showed that the alkyl chains on the Sil-DSG are in gauche form and their mobility is strongly restricted at room temperature. This paper discusses higher selectivity of Sil-DSG with a carbonyl pi-benzene pi interactions.

Peptic ulcer perforation: management of high-risk cases by percutaneous abdominal drainage.

Sixty-six patients were selected as high-risk cases of duodenal ulcer perforation. After resuscitation with intravenous fluids and nasogastric suction, a widebore percutaneous intra-abdominal drain was put in under local anaesthesia. There were three (4.5%) deaths; 58 (87.8%) patients improved satisfactorily. High-risk peptic ulcer perforation patients can be managed by putting in an intra-abdominal drain supported by conservative treatment.

Adolescent self reported reproductive morbidity and health care seeking behaviour.

BACKGROUND: Addressing reproductive health issues of women is now on the global social agenda in the new millennium. Maternal mortality has long been the only indicator of women's health even though reproductive morbidity occurs far more frequently and seriously affects women's lives. In this paper, an attempt was made to assess the magnitude of self reported gynaecological morbidity unrelated to childbearing among the adolescents irrespective of their marital status. The paper also explored the determinants of health care seeking behaviour of the adolescents for their reproductive ailments. METHODS: Both quantitative and qualitative data were collected for this study. Cross sectional study was conducted both in rural and urban areas using a multistage cluster sampling technique. A nationally representative data on 2883 adolescents irrespective of their marital status were analysed. RESULTS: Analysis revealed that a large proportion of the adolescents (64.5%) reportedly has been suffering from gynaecological morbidity. The most frequent form of morbidity was menstrual disorders (63.9%) followed by lower abdominal pain (58.6%), burning sensation during urination (46.1%), genital itching (15.5%), vaginal discharge (3.4%) etc. Multivariate logistic regression analysis revealed that older adolescents aged 15-19 years, family income, type of family, type of residence and hygienic practice during menstruation appeared to be influencing factors for adolescents reproductive morbidity. The results also revealed that about one fifth (18.0%) sought health care for their gynaecological ailments indicating that adolescents were unaware about their reproductive morbidity (p<0.05). For assessing the factors influencing their health care seeking for reproductive morbidity, multivariate logistic regression analysis found significant positive association with adolescents aged 15-19 years, having autonomy in treatment, working status, adolescents of joint or extended family (p<0.05). DISCUSSION: Adolescent reproductive health in Bangladesh indicates high incidence of maternal morbidity. The incidence of these health problems varied by socio-economic and demographic characteristics of the adolescents. Recommendations to address these problems include encouragement of female education, introduction of family life education in school curricula, creating community awareness for seeking health care and empowerment of women in household decision making process.
