Relationship between mitochondrial ATPase and membrane lipids from developing human placenta.

Mitochondrial lipids and ATPase activity have been studied in developing human placenta. The enzyme activity in the placenta increases steadily with gestational age until the 18th to 21st week, and decreases in the second half of pregnancy. Both Mg2+ and Ca2+ ions have been found to activate the enzyme system. The solubilization of membrane lipids by deoxycholate inhibits the enzyme activity. Total lipid and total phospholipid contents of the mitochondrial membranes also increase during the early development of the placenta with peaks in the 14th to 17th week, after which the levels decrease. However, the percentage composition of individual phospholipids remains almost the same throughout pregnancy. The profile of the ATPase activity exhibits a correlation with that of membrane lipids in human placenta during intrauterine development of the tissue.

Comparative studies on acid phosphatase isozymes of developing human placenta and fetal liver.

Acid phosphatases from human placenta and fetal liver during ontogenic development have been studied by electrophoresis on polyacrylamide gels. Three isozymes have been detected in liver as well as in early and term placenta, and one more in placenta at mid-gestation. These isozymes behaved differentially towards heat and a number of inhibitors at different weeks of gestation. Quantitative determination of each isozyme also showed stage-specific variation.

Long-term contraception with Depo-Provera: a clinical evaluation.

Depo-Provera has been used as a long-acting injectable contraceptive. The results of the study carried out for more than 2 years indicate that the depot formulation of medroxyprogesterone acetate (DMPA, Depo-Provera) at a dose of 150 mg at 90-day intervals appears to be acceptable to some women of this part of India, though many of them experience menstrual problems. However, irregular and/or excessive menstrual bleeding diminishes with long-term use of the contraceptive and a high percentage of users become amenorrheic. No major adverse reactions were observed and the minor side effects are comparable to those reported for the oral contraceptives. There was a tendency for weight-gain by the patients and the endometrium became atrophic or "resting" with the prolongation of the therapy.

Studies on some unusual lipids of the preputial glands of rats and mice and their hormonal influence.

Major neutral lipid fractions of the preputial glands of mice have been identified by analytical and preparative thin layer chromatography (TLC) and gas liquid chromatography (GLC) as wax esters, neutral plasmalogen, alkyl diglyceride, triglyceride and cholesterol. An unusual lipid fraction between wax esters and neutral plasmalogens was identified as alkyl acetate, which has been reported earlier to function as sex attractant. The lipid pattern of the preputial glands of rats in both sexes was similarly studied. Three unknown bands of lipids were identified by TLC between wax ester and alk-1-enyl diglycerides, which on further analysis by GLC and infrared spectroscopy gave preliminary indication of aliphatic diester waxes. Administration of androgen stimulated the biosynthesis of these substances.

Effect of ovariectomy and replacement therapy on the tissue lipid pattern in rats.

Ovariectomy increases the percentage of total lipids in liver, kidney and uterus of intact cyclic rats. Estrogen and progesterone, when administered individually to ovariectomized rats, caused a decrease in the total lipid content of all tissues. Th effect of progesterone in estrogen-primed rats is not significant. Triglyceride and cholesterol content increases after ovariectomy; treatment with estrogen in ovariectomized rats led to a decrease in the concentration of these lipids. Progesterone has no significant effect on these lipids but showed an antagonistic action when given in estrogen-primed ovariectomized rats. The proportions of ethanolamine, choline and inositol phospholipids decreased after spaying and increased when estrogen was given to spayed rats. Progesterone alone had effect only on the uterus whereas progesterone administered to estrogen-primed rats showed an antagonistic effect in all tissues.

Biochemical studies following ovulation induction with clomiphene citrate and human menopausal gonadotropin in infertile Indian women.

Biochemical studies were done in 65 anovulatory women before and following induction of ovulation with ethinyl estradiol/clomiphene citrate/human chorionic gonadotropin and also clomiphene citrate/human menopausal gonadotropin/human chorionic gonadotropin to determine the metabolic adverse effects, if any, of such therapy. Significant increases occurred in lipids and lipoprotein levels, while minor changes were seen in liver function enzymes and fasting blood sugar. Sialic acid decreased significantly after therapy in both the regimens, and both induction therapies seem to be free of adverse metabolic effects of hormones. On comparison with normal ovulatory women, this study suggests that some of the biochemical parameters can serve as reliable indicators of ensuing ovulation.

Alterations in blood sugar, protein, and lipid levels in Indian women by norethisterone enanthate.

Fasting levels of sugar, protein, and lipids, namely, cholesterol, triglyceride, and phospholipid, were determined in the blood of 150 women taking an injectable contraceptive, norethisterone enanthate, 200 mg every 60 days for 24 months. Triglyceride and cholesterol showed a statistically significant decrease from 10 and 16 months of use; however, both values returned to control level after use for 24 months. Serum phospholipids and total protein did not show significant changes, while the blood sugar level increased significantly from 12 months onwards and remained elevated (P less than .001) up to the end of the trial period.
