A possible role played by cAMP in the stimulating action of ACTH and hydrocortisone on lactose E. coli K-12 operon was studied. It was shown that ACTH caused no effect in the E. coli WZ-78/F'lac (cya855) and E. coli CA8001 (L1) strains with destroyed positive cAMP control system of the lactose operon function, at the same time producing a stimulating effect on the lactose operon in the strains of wild type, i.e. E coli 200PS/F'lac and E. coli 3000. Hydrocortisone stimulated the lactose operon function both in E. coli 3000 and in the mutant E. coli CA8001 (L1). It was supposed that the accelerating effect of ACTH on the lactose operon was mediated through cAMP; as to hydrocortisone--it stimulated the lactose operon function independently of cAMP.
A comparative study of the effect of cortisone on the beta-galactosidase synthesis in E. coli K-12 strains with an induced (E. coli 200 PS/Iac), constitutive (E. coli ML-308), and superrepressed (E. coli 200is) type of the enzyme synthesis and in the cells of rat liver demonstrated that the hormone proper had no derepressive effect. An increase of the beta-galactosidase synthesis occurred only in the presence of specific substrate inductors. It is supposed that the principal link in the action mechanism of cortisone on the laclose operon of E. coli and the enzyme production in the cells of the rat liver is preliminary derepression of the genome areas by means of the substrate inductors.
The influence of cAMP, ACTH and glucose on the stimulation of chloramphenicol acetyl transferase synthesis in the cells of E. coli CSH-2/R222 and E. coli WZ-78/R222. (cya855) was examined. Glucose proved to decrease the enzyme synthesis in E. coli CSH-2/R222, causing catabolite repression; 5 mM of cAMP and 1000 mug/ml ACTH overcame the latter. The enzyme synthesis in E. coli WZ-78/R222 was insensitive to the catabolite repression; as to ACTH - it failed to cause stimulation ofchloramphenicol acetyl transferase synthesis in E. coli WZ-78/R222.
A study was made of the effect of cyclic cyclic 3',5'-adenosinmonophosphate (c-AMP) on the production of chloramphenicol-acetyltranspherase (CAT) enzyme by whole bacterial cells in the strains of E. coli CSH-2/R222 and WZ-78/R222 (cya855). It was shown that CAT synthesis in the E. coli WZ-78/R222 strain was two times less in comparison with the E. coli CSH-2/R222 strain. C-AMP only insignificantly increased the production of CAT by the E. coli CSH-2/R222 strain, but considerably influenced the production of this enzyme by the WZ-78/R222 strain.
In this work the results of obtaining HBcAg-producing attenuated Salmonella strains, serovars S. enteritidis and S. typhimurium, and their comparative study is presented. As revealed in this study, attenuated S. enteritidis strain E-23 and S. typhimurium strain T-10, producing HBcAg, induce cell-mediated and humoral immune response to HBcAg after injected into anovals. After injection S. typhimurium strain T-10 induces a much higher titer of specific antibodies than S. enteritidis strain E-23. The level of specific antibodies induced by recombinant HBcAg seems to correlate with the capacity of salmonellae for survival inside macroorganisms.
The behavior of S. enteritidis recombinant strain E-23/pKDNA(3.1) with HBc antigen expressed by the eukaryotic promoter (DNA vaccine) in the body of mice after their infection per rectum and in the culture of macrophages was studied. The expression of HBc was shown to lead to the loss of the capacity of salmonellae for persistence in parenteral lymphoid tissue and for inducing the formation of anti-HBc antibodies. The study of the interaction of salmonellae with the macrophage culture revealed that the synthesis of HBc antigen inhibited the intracellular multiplication of salmonellae.
