[Clinical studies of 9, 3"-diacetylmidecamycin (author's transl)].

1. We had a chance to administer the "MOM dry syrup' (MOM fine granules) to 22 patients: 6 patients with streptococcal infections and 16 patients with other respiratory tract infections. The clinical efficacy was good in 16 cases and its rate was 80%. 2. The causative organisms were isolated from 9 cases: 6 cases with S. pyogenes, 2 with S. pneumoniae and 1 case with B. pertussis. All of the clinical isolates were eradicated except for the S. pyogenes strains. 3. Any remarked side effects were not observed but only eosinophilia in 2 cases. 4. Because of no bitterness of the MOM dry syrup, all the children take it easily. 5. MOM was effective in all the cases of pertussis and Mycoplasma pneumoniae pneumonia. As for streptococcal and pneumococcal infections, it is necessary to administer MOM under the control of bacterial sensitivity.

An improved short-term culture method for human mammary epithelial cells.

An improved short-term culture method for human mammary epithelial cells was developed and the details are presented. Briefly, after the digestion of biopsy tissues with 0.2% collagenase solution for 16 to 18 hr, 4 consecutive fractions were obtained by differential sedimentation. The last fraction, S-4, contained organoids having alveolar- or duct-like structures and gave rise to mammary epithelial cell growth with minimal contamination of fibroblasts.

Two types of dopaminergic nerve terminals in the rat neostriatum. An ultrastructural study.

In the neostriatum 7.7% of all boutons absorbed alpha-methylnoradrenaline, thus staining their synaptic vesicles profiles granular. These presumably monoaminergic synapses were divided into two types according to ultrastructural features. Type A was composed of a relatively large axon terminal and a dendritic spine with postsynaptic membrane thickening. The axon terminal of type B, that is a bouton en passant, was smaller than the postsynaptic profile, while the synaptic contact was symmetrical. Following nigral coagulation, both types of synapse degenerated. It is concluded that both types of synapse are dopaminergic.

Substance P-like immunoreactivity in plasma of psychotic patients and effects of neuroleptics and electroconvulsive therapy.

Radioimmunoassay of substance P (SP)-like reactivity, using N alpha-tyrosyl-SP, was performed in plasma of psychotic patients. Schizophrenic patients under neuroleptic medication over 1 year, and those patients with organic brain disease showed a significant increase in immunoreactivity. The immunoreactivity in unmedicated schizophrenic patients just before treatment and in depressed patients, both bipolar and major, did not differ significantly from that of controls. The immunoreactivity in unmedicated schizophrenics was followed after medication for a maximum of 5 months. The reactivity decreased in five cases, and increased in three cases. No definite pattern could be discerned between changes in the immunoreactivity, the type of medication administered, and the resultant response to the therapy. Chlorpromazine injection caused no definite acute effect on the plasma level of SP-like immunoreactivity for 120 minutes. Electroconvulsive therapy increased the plasma level of SP-like immunoreactivity for 90 minutes in a male schizophrenic patient.

Culture conditions for a human lymphoma cell line (Raji) and their application to mutation research.

Experimental systems for the detection of somatic cell mutation to hypoxanthine-guanine phosphoribosyl transferase (HGPRT) deficiency in human lymphoma cells were developed. The cell line used was the Raji line established from Burkitt's lymphoma. The cloning efficiency (CE) in soft agar of the cells was enhanced approximately 5-fold by addition of 2 x 10(-4) to 4 x 10(-4)M dithiothreitol (DDT) and 1% human serum, as compared with the CE in control medium. Other thiols such a mercaptoethanol and thioglycerin were also able to increase the CE but not as efficiently as DTT. Under these culture conditions, 10(6) cells per 55-mm dish could be seeded into soft agar medium containing 5 micrograms/ml 6-thioguanine (6-TG) to detect a forward mutation without any metabolic cooperation. The optimum expression time was determined by treating the cells with 125 micrograms/ml ethyl methanesulfonate (EMS) for 20 hr at 37 degrees. A significant increase of mutation frequency was observed approximately 10 days after the 6-TG treatment, by which time the cells had gone through about 10 population doublings. After determining these experimental conditions, a comparison of mutation frequency among three mutagens, EMS, N-methyl-N'-nitro-N-nitrosoguanidine and ICR-170, was carried out. ICr-170 was the most efficient mutagen among these chemicals.

Lactate dehydrogenase isozyme patterns of normal human fibroblasts and their in vitro-transformed counterparts obtained by treatment with CO-60 gamma-rays, SV40 or 4-nitroquinoline 1-oxide.

Lactate dehydrogenase (LDH) isoenzyme patterns of normal human fibroblasts and their in vitro-transformed counterparts were studied. Normal human fibroblasts were derived from embryonic or adult skin or embryonic lung tissues. These cells were transformed into neoplastic cells having unlimited growth potential and abnormal karyotypes by treatment with CO-60 gamma-rays, SV40 or 4-nitroquinoline 1-oxide. No significant difference in LDH isoenzyme patterns was observed in growing or confluent cultures, in the process of in vitro aging of normal human fibroblasts, or in embryonic or adult skin fibroblasts, but there was a considerable difference in isozyme patterns between lung and skin fibroblasts cultured from embryos. All of the transformed fibroblast cell lines derived from lung or skin showed a distinctly increased ratio of B to A subunit (greater than 0.5), whereas normal human fibroblasts did not (less than 0.5). There was no significant difference in total activity of LDH in the various cells used for the present experiments. These findings suggest that the changes of LDH isozyme patterns may be useful as a marker of neoplastic transformation in cultures of normal human fibroblasts.

Ultrastructure of acetylcholinesterase synthesizing neurons in the neostriatum.

Acetylcholinesterase (AChE) ultracytochemistry with thiocholine method was studied in the rat neostriatum. AChE synthesizing neurons were identified by observing reappearance of the enzyme after application of an AChE inhibitor, DFP. Neurons in the neostriatum were classified into 6 types. All such types were demonstrated to synthesize AChE. Two types of efferent neuron (types III and IV) have shown a relatively large amount of AChE and resynthesized the enzyme in the earlier stage after DFP. A tentative model of the neuronal network in the neostriatum was schematically shown.

Uptake and excretion of 67Ga-citrate in malignant tumors and normal cells.

Using in vivo and in vitro experimental models, the uptake and excretion of 67Ga-citrate in tumor cells and normal cells were studied. The time-lapse accumulation of 67Ga in the tumor of rats bearing Yoshida sarcoma reached its peak 24 h after the administration of 67Ga and gradually decreased thereafter. However, the excretion of 67Ga from the tumor was less than that from normal lung. For culture cells in vitro, the uptake of 67Ga increased with lapse of contact time between 67Ga and the cells, but there was no distinct difference between the results for tumor cells and normal skin fibroblasts. The excretion of 67Ga from the cells tended to decrease with prolongation of the contact time, the excretion from tumor cells being only about 10% after a contact time of 24 h. This indicated a significant delay in excretion in comparison with that of normal skin fibroblasts. This delay in the excretion of 67Ga may be an important factor in the tumor accumulation of 67Ga.
