Pancreatic polypeptide cells of rat pancreas after chronic ethanol feeding.

Male Wistar rats, (2 months old) were randomly divided into two groups according to the diet offered (C-control and E-ethanol treated rats). Final body weight was significantly increased but pancreatic weight as a percentage of body weight was decreased in ethanol treated rats. Volume density, number of pancreatic poly peptide (PP)-cells per islet and per micron 2 of islet were significantly increased. PP-cells were abundant and occupied the whole periphery of islets in the splenic part of the pancreas. Those cells showed strong immunopositivity. At the ultrastructural level PP granules had predominantly less electron density. The mean diameter of PP granules was significantly increased and the number of granules of larger diameter was greater in the E group of rats, than in the controls.

Diapedesis of thrombocytes from capillary into the intercellular space of interscapular brown adipose tissue and their increase by Ca-Sandoz.

Diapedetic capacity of the rat thrombocytes to leave capillaries of the interscapular brown adipose tissue (IBAT) and infiltrate the interstitium has been observed by conventional electron-microscopy. Thrombocytes that reach IBAT interstitium are morphologically completely different from lumenary ones. The interstitial thrombocyte has a prominet head region (1.51 x 2.12 microns) and very long phylopodium (3.43 microns). Experimental conditions which induced drastic changes in morphology of interstitial thrombocytes were: sucrose overfeeding (10% over 2 days); a 24 hour starving after sucrose overfeeding and Ca-Sandoz drinking (480 mg/L Ca2+ during 2 days). The thrombocytes in the IBAT interstitium can be classified as activated according to: a) pseudopode extension; b) swollen open canalicullar system (OCS); c) endocytosis via coated pits and vesicles; and d) structural changes in alpha granules excreted to the interstitium through OCS. In the IBAT interstitium of 24-hour starved rats after sucrose overfeeding, a thrombocytic layer was observed. It was suggested that thrombocyte adrenalin, stored in dense bodies, was selectively included in the IBAT supply without mediation of the central nervous system.

Gastrin producing G-cells after chronic ethanol and low protein nutrition.

Male Wistar rats, (2 months old), randomly divided according to the diet offered to four groups (C-control; A- alcoholized, PD-protein-deprived, A-PD- alcoholized protein-deprived). In group A and A-PD rats, the number of gastrin producing G-cells was significantly lower. The volume density of G-cells was significantly decreased in alcoholic rats. Fasting serum gastrin level (FSGL) significantly raised due to combined effect of alcohol consumption and protein malnutrition. In group A rats, the profile area of G-cells and their nuclei increased. In PD rats, the profile area of G cells also increased. There were no differences in nucleus/cell ratio due to alcohol ingestion alone, but it decreased significantly in PD and A-PD rats. Pale and lucent types of granules were predominantly seen in G-cells of animals of group A and A-PD. Mean diameter of granules increased in A, PD and A-PD rats. Other endocrine cells (ECL, D, EC) also decreased in number in A rats. Somatostatin producing D-cells decreased significantly in A-PD rats, both in fundic and pyloric mucosa.

Intra-amniotic Chlamydia trachomatis infection.

Chlamydia trachomatis is one of the most prevalent genital pathogens in pregnant women. Ascending, transcervical infection may reach fetal membranes creating chorioamnionitis or amniotic fluid infection. The aim of this study was to examine amniotic fluids obtained during cesarean section for the presence of chlamydial IgM- and IgG-specific antibodies, and for the presence of C. trachomatis antigen. Five of 52 (9.6%) amniotic fluid samples were seropositive. Two of 52 (3.8%) amniotic fluid samples had C. trachomatis antigen in the epithelial cells of the amnion. In conclusion, our data indicate that there is a high rate of transmission of C. trachomatis from mother to infant and that the pathogen can be identified in the amniotic fluid.

Simplified sample preparation for fluoride determination in biological material.

A simple and rapid preparation method for the determination of fluoride in biological materials (blood and food) of various origins, is described. A homogenized sample was placed in a plastic diffusion cell and calcium phosphate added, it was then dried at 55 degrees C and treated with 70% HClO4 and 40% AgClO4. After digestion for 24 h at 55 degrees C, the fluorides released were fixed on the upper part of a diffusion cell containing a thin layer of NaOH. The analyses of the diffused fluoride were carried out with an ion-selective electrode. The proposed microdiffusion method, without mineralization, enables quantitative separation of the fluoride from the biological samples.

The effect of dose on maternal-foetal transfer of fluoride in rabbits.

Placental transfer of fluoride was investigated by fluoride determination in the bones and teeth of newborn rabbits whose mothers had been treated with fluoride during pregnancy. The mothers were given doses of 0, 0.10, 0.52 and 1.05 mmol fluoride per kg body weight as sodium fluoride, from the 16th day after conception to the end of pregnancy. All the doses produced a significant increase of fluoride level in the bones and teeth of newborn rabbits, indicating that the placenta was no barrier for the passage of fluoride.

Direct determination of lead in blood by differential pulse stripping voltammetry.

A simple and rapid method for determination of lead in blood by differential pulse stripping voltammetry (DPS) without preliminary mineralization of the sample, is described. Lead determination was performed in native samples of blood in the electrolyte of Britton-Robinson buffer (pH 5-5.5). The proposed method is shown to be reproducible and in good agreement with the reference technique AAS used on mineralized blood samples. Our results indicate that differential pulse stripping voltammetry is a reliable, rapid and highly sensitive method for blood lead analysis.

Levels of lactate dehydrogenase and creatine kinase in plasma of fluoride-treated rabbits.

The effect of different concentrations of fluoride (as sodium fluoride) on the enzymes lactate dehydrogenase (LDH, EC 1.1.1.27) and creatine kinase (CK, EC 2.7.3.2) in rabbit plasma was investigated in vivo and in vitro. Rabbits were dosed orally, for 30 days, with fluoride at 2, 10 or 20 mg per kg body weight as sodium fluoride. LDH and CK levels were determined at 10-day intervals during the treatment. Fluoride, depending on the applied dose, inhibited, had no effect or stimulated the investigated enzymes in vivo. On the other hand, fluoride had no effect on the activity of LDH and CK in vitro.

Protein concentration in immunological preparations and pyrogenic reaction of rabbits.

The drop of arterial pressure was found to be caused by proteins injected intravenously into test animals. The drop was related to the protein concentration and the volume of the dose. Low pressure was caused neither by the histamine nor by the protein characterized by its property of liberating histamine. Antihistamines had no influence on quantitative or qualitative depressory reaction, caused by the presence of dissolved proteins. Concentrated protein solutions, when applied intravenously, did not contribute to identifying the pyrogenic reaction, but had a damaging effect on the organism and homeostasis of animals used in the experiments.

[Relation between dental plaque fluoride content and caries].

The purpose of this study was to investigate the effect of plaque fluoride content on dental caries in children aged 12 years, from the area with minimal (Valjevo) and optimal and/or higher fluoride concentrations (Vranjska Banja) in drinking water. The caries prevalence was determined by the standardized epidemiological method. The fluoride concentration in dental plaques was measured with the Ion-selective electrode. The fluoride content in the plaque of children from Valjevo (7.8 ppm) was significantly smaller than in children from Vranjska Banja (36.5 ppm). In 12-year old individuals with the lowest fluoride concentrations in the plaque the greatest values of the caries prevalence were found. The increase in plaque fluoride content provoked the decrease in caries prevalence. The lowest values of caries index were found in children with 21 to 30 ppF in dental plaque.

[Meteorotropism in children with cerebral palsy].

INTRODUCTION: Members of the Rehabilitation team and families of children with cerebral palsy are daily engaged in various activities with these children. Weather changes (the atmospheric pressure especially) on particular days, cause difficult performance of anticipated therapeutic exercises, occupational therapy, speech therapy, education, daily living etc. The aim of this study was to underline the possible connection between performing daily activities of children with cerebral palsy and weather changes. MATERIAL AND METHODS: There were 16 children with cerebral palsy, 2 to 14 years of age, 8 females and 8 males. Parents of these children got a questionnaire where they had to write down, on daily bases, one of the following: A--correct and easy to perform exercises, B--difficult to perform exercises, C--impossible to perform exercises--the child indisposed, for a period of three months. For the same period of time the atmospheric pressure changes and the temperature were recorded by a professional. RESULTS: Our findings are related to the relationship between the efficiency of physical treatment in children with cerebral palsy and meteorological phenomena. The weather changes did not influence about 75% of the patients. In the other group of patients the unsuccessful exercise was associated with periods of extensive cyclonic activity: 15 to 17 of March 1991 (the decrease of atmosperic pressure from 1010 to 999 mbara), 15 to 17 of April 1991 (the decrease of atmospheric pressure from 1000 to 987 mbara), 5 to 7 of May 1991 (the decrease of atmospheric pressure from 1000 to 987 mbara). DISCUSSION: Before and during cyclonic activity increases the concentration of the positive air iones which interferes with neuromuscular excitation and causes serotonin hyperproduction in the central nervous system, thus causing irritation and the lack of motivation for imposed demanods. CONCLUSION: About 23% of children with cerebral palsy show meteorotropism in terms of weather changes influencing the efficiency of exercises. The cyclonic activity has unfavourable influence on exercises, while the stable atmospheric pressure or anticyclonic activity has a favourable one in children with cerebral palsy who show meteorotropism. So, favourable and unfavourable days for therapeutic exercises can be clearly differed.
