[Uptake of osteotropic agents in the spine in laboratory rats after the administration of biphenyl derivatives].

The effect of the derivative trichlorobiphenyl and of ergocalciferol on calcium uptake into the spinal column was studied in young laboratory rats. The substances were administered in the period of suckling. Before sacrificing the animals the bone seeker 85 Sr was administered. The uptake of the seeker decrease after administration of the derivative trichlorobifenyl. Ingestion of the biphenyl derivative and of ergocalciferol resulted in a mild increase in bone seeker uptake. The increase was comparable to the uptake level in control animals. The effect of the derivative trichlorobiphenyl is accounted for by blockade of cytoplasmic receptors of chondrocytes.

Ultrasonography of primary liver carcinoma (PLC) and targetted cytologic examination.

A primary liver cell carcinoma was disclosed in 11 cases out of a total of 61 patients with a focal hepatic lesion. The authors investigate the diagnostic significance of abdominal ultrasonography (US) as a major diagnostic modality in early detection of PLC. It is concluded that abdominal US is an important contribution to diagnosis of hepatic lesions smaller than 1 cm in diameter. The echographic features of PLC are often unspecific thus requiring a complementary targetted cytologic examination. A short comparison of US computed tomography and liver scintigraphy is given.

[Comparison of the effects of diltiazem and isradipine in patients with chronic stable angina pectoris].

In 21 patients with chronic stable angina pectoris stage II-III according to NYHA, by means of bicycle ergometry antianginous and antiischaemic effects of short-term dilthiazem administration (Blocalcin 60, Lachema) were compared with isradipine (Lomir, Sandoz). Dilthiazem was administered in three doses à 90 mg by mouth (midday, evening and subsequent morning), isradipine in a similar way but in three doses à 5 mg. Both preparations reduced, as compared with placebo, significantly the systolic and diastolic blood pressure at rest and the diastolic pressure during a load, dilthiazem also the systolic pressure during a load. Both drugs raised also significantly the total amount of work performed during an ergometric examination, dilthiazem moreover delayed significantly the onset of stenocardia and significantly reduced the ST depression in lead V5 during the maximal load achieved with all administered drugs. A significant increase of the heart rate at rest and in particular of the heart rate during an equal maximal load after isradipine or after placebo, but in particular after dilthiazem, led in comparison with dilthiazem to a significant rise of Robinson's index during the same maximum load and was obviously the cause of its lower anti-anginous and antiischaemic effectiveness. Tachycardia at rest and after a load following isradipine administration led in four patients to a deteriorated load tolerance, as compared with placebo, in another patient tachycardia caused stenocardia at rest with ST depressions on the electrocardiographic tracing. Based on the assembled results, it seems better and safer to use in the treatment of chronic stable angina pectoris dilthiazem rather than isradipine.

Comparison of protein and lipopolysaccharide patterns of several Coxiella burnetii strains in phase I.

Electrophoresis in polyacrylamide gel gradients in the presence of sodium dodecyl sulphate revealed at least 18 identical protein bands in eight strains of Coxiella burnetii. By staining with Coomassie Brilliant Blue R-250 it was possible to visualize clearly at least 40 proteins. The protein pattern showed the greatest variability in the region from 27 to 18.5 kD. Strains S and Priscilla differed from the other strains also in proteins smaller than 18.5 kD. The lipopolysaccharide pattern obtained by the same technique consisted of from 6 to 12 various bands in the region from 23.5 to 11.8 kD. The protein and lipopolysaccharide patterns of four strains isolated from ticks did not significantly differ from those of strains Henzerling and L-35 isolated from men.

Monoclonal antibodies neutralizing human leukocyte acid- and thermolabile interferon alpha.

Using a high titred human leukocyte IFN alpha preparation which contained both acid- and thermolabile (AL-IFN alpha) and acid- and thermostable IFN alpha species in 9:1 proportion for immunization of BALB/c mice, five hybridomas secreting monoclonal antibodies that reacted with AL-IFN alpha were obtained. In antiviral and antiproliferative tests on HL-60 cells, their products showed high degree of specificity for AL-IFN alpha. The results suggest that both the "normal" leukocyte AL-IFN alpha and the IFN alpha found in sera of autoimmune and other chronic patients might belong to the same subtype of IFN alpha.

Murine monoclonal antibodies against human fibroblast (beta) interferon. Correlation of the neutralization of antiviral and antiproliferative activities.

Eight mouse hybridoma lines secreting monoclonal antibodies (MoAb) to human fibroblast interferon (HuIFN-beta) were established. All MoAbs were capable of neutralizing two different biological activities of HuIFN-beta: the antiviral activity and the antiproliferative activity. Positive correlation was demonstrated between the ability of hybridoma culture supernatants to neutralize the antiviral and antiproliferative effects of human fibroblast IFN. The neutralizing capacity of individual hybridoma culture supernatants depended on the concentration of MoAb in the sample. Only one of IFN-neutralizing MoAbs has shown binding capacity to human fibroblast IFN when used in enzyme immunoassay as solid-phase antigen. This MoAb was purified to homogeneity and its specific neutralizing activity against HuIFN-beta was calculated (3.5 X 10(4) I.U. per mg).

[New possibilities of treatment of pancreatic pseudocyst].

The authors evaluate the therapeutic results in patients with pancreatic pseudocysts where percutaneous puncture under ultrasonic control was used. Five patients were thus cured. In six patients, despite repeated punctures, treatment was not successful and internal drainage was made by surgery. The authors consider treatment of pseudocysts by puncture in suitably selected patients fully indicated because it is an unpretentious and relatively safe method.

[Objective adjustment of hearing aids to hearing disorders].

The aim of a functional prosthesis - a hearing aid - is to improve the comprehensibility of speech. The hearing aid is individually adjusted for the patient. Every deaf patient is subjected first to an extensive otolaryngological and audiological examination programme which comprises the recording of the case-history, assessment of the ENT diagnosis, examinations of threshold and suprathreshold sound audiometry, verbal and sentence audiometry in a sound-proof chamber and in space. To set the most important parameters of hearing aids (frequency characteristics, intensification and dynamic range) directly on the patient's ear, in the ENT department of the health centre of the Uranium Industry in Príbram Rastronics equipment from Denmark is used which makes it possible to achieve optimal adjustment of hearing aids, using several calculations applied on an international scale.

An X-ray, microanalytical and ultrastructural study of cadmium absorption and transport in rat liver.

Accumulation of cadmium in the liver was demonstrated by X-ray microanalysis in every type of experiment, i.e. after injecting Cd into the ligated intestine and after the peroral acute single and combined, subchronic and chronic administration of Cd. Half an hour after its injection, Cd was localized diffusely in the liver; one hour after injection its increased accumulation in the cells caused generalized changes in the endoplasmic reticulum, mitochondria and nuclei. In acute and chronic peroral tests, the hepatocytes of the intermedial and peripheral zone of the lobes were the main storage region. After an acute dose of Cd, the cells in the centrolobular zone were hydropic, or single-cell necrosis developed; after the longer effect of combined doses the latter was manifested as centrolobular focal necrosis. Cd was not demonstrated in the lesions. Chronic administration did not lead to manifest severe degenerative changes in the liver. Accretions in the mitochondria and on the membranes of the endoplasmic reticulum were identified by means of X-ray analysis with cadmium peaks. Cadmium showed up clearly as L alpha- and L beta-lines at 3.135-3.320 keV. We presume that cadmium is bound in the ribosomes of the endoplasmic reticulum, as well as the mitochondria, and is released by the invagination of swelling mitochondria of the peripheral hepatocytes into Disse's spaces.

Multiprogrammable pacemakers: are their advantages fully employed?

Out of 1265 pacemakers implanted between 1981 and 1987 in our center, 50 (4.0 percent) were of SSI M mode. Utilization of the programming capability of these multiprogrammable single-chamber pulse generators was investigated. All devices were programmed to the "energy saving mode" ensuring the longest lifetime possible. Half of them did not have to be reprogrammed for any other reason, the other half had to be programmed for cardiological and/or technical dysfunctions. Full utilization has been made of the programmability of these sophisticated and expensive units and their wide application is justified.

Sterol composition of a delta 5,7-sterol-rich strain of Saccharomyces cerevisiae during batch growth.

Sterol composition was examined during batch growth on complex media containing ethanol, molasses or glucose as the carbon source. The molasses-grown cells exhibited a balanced sterol composition throughout growth, maintaining the proportion of ergosterol to 24:28-dehydroergosterol equal to 1.4. The negative effect of glucose on sterol synthesis manifested itself by decreasing the accumulation of 24:28-dehydroergosterol and total sterols but not of ergosterol. Using ethanol as the sole carbon source, a large amount of 24:28-dehydroergosterol accumulated, partly at the expense of other sterols. The gradual addition of nitrogen source during growth significantly decreased the accumulation of ergosterol, 24:28-dehydroergosterol and of total sterols. A general scheme of regulation of sterol synthesis in baker's yeast is presented.

Monoclonal antibodies neutralizing the antiviral and the antiproliferative activities of human interferon gamma.

Spleen cells from BALB/c mice immunized with human leukocyte interferon gamma (HuIFN-gamma) were fused with mouse NSO myeloma cells. Nine hybridoma lines were found secreting monoclonal antibodies (MoAb) which were able to neutralize the antiviral activity of HuIFN-gamma. All nine MoAb inhibited also the antiproliferative activity of HuIFN-gamma. The ability of tested MoAb to inhibit both the antiviral and antiproliferative activities of HuIFN-gamma supports the suggestion that a common domain on HuIFN-gamma molecule might be responsible for both biological activities. However, ELISA and/or RIA proved unsuitable for detection of these specific MoAb.

Pulmonary gas exchange in chronic obstructive lung disease patients with pulmonary hypertension.

To ascertain if any specific changes are associated with the development of pulmonary hypertension in patients with chronic obstructive lung disease, respiratory gas exchange was measured at rest and during exercise in 52 such patients, of whom 22 already had pulmonary hypertension at rest and/or during exercise. Lung function, gas exchange and haemodynamic indices in the hypertonic group were compared with those in the normotonic group. It was shown that gas exchange was not altered by pulmonary hypertension in any specific fashion. The differences found were related to the higher age of the patients and the higher degree of obstruction present in them, leading in turn to a higher degree of ventilation-perfusion mismatch responsible for the lowered PaO2 ane elevated PaCO2.

The effect of postfusion cell density on establishment of hybridomas.

The effect of postfusion cell density on establishment of hybridomas in primary microculture was studied. Following fusion of BALB/c spleen and NS0 myeloma cells in the ratio of 5:1 we demonstrated that seeding the cells at a concentration of about 20-25 X 10(4) spleen cells per well resulted in maximal number of wells containing only one growing hybridoma clone (40.1-43.7% plated wells). Importance of generation of microculture with one clone for the effectiveness of preparation of mAb-secreting hybridomas is discussed.

Karyotype analysis of hybridomas producing monoclonal antibodies against different antigens.

On the basis of karyotype analysis of the 6 groups of mouse--mouse hybridomas secreting monoclonal antibodies against 5 different antigens we found: (1) The karyotypic variability of clones in all six compared groups of hybridomas was consistent and was influenced neither by the specificity of secreted monoclonal antibodies nor by the character of antigen used for immunization (soluble or particulate). (2) The faster growing hybridomas had a lower mean chromosome number than the slower proliferating clones of the same origin. (3) Karyotypes of subclones isolated from ascites fluid of a mouse inoculated i.p. with clones permanently growing in vitro included a markedly higher mean chromosome number than the karyotypes of originally inoculated hybridoma cells.
