Expression of the cell-binding domain of human fibronectin in E. coli. Identification of sequences promoting full to minimal adhesive function.

Two cDNA subfragments containing the cell-attachment site of human fibronectin (FN) were expressed as beta-galactosidase fusion proteins in E. coli. The products were purified to homogeneity by monoclonal antibody affinity chromatography and assayed for activity in a standard cell-adhesion assay. A fusion protein containing an 80 kDa fragment of human FN appeared functionally equivalent to intact FN purified from human plasma, whereas a truncated fusion protein of 33 kDa still containing a previously postulated cell-attachment site was approx. 50-fold less active. Our study establishes a system for analyzing adhesive protein function by DNA manipulation, rules out any major role for eukaryotic post-translational modifications in FN adhesive function, and localizes additional functional activity to a 1.3 kb region.

[Endocrinological background of pituitary desensitization during pulsatile LH-RH therapy for ovulation induction in amenorrheic women].

In order to elucidate the endocrinological background of patients in whom ovulation induction by pulsatile LH-RH administration resulted in failure, 16 women with hypothalamic amenorrhea were treated with 10 to 20 micrograms of LH-RH injected subcutaneously every 2 hrs. Six women did not ovulate, and 5 of them showed pituitary desensitization since the basal gonadotropin concentration gradually decreased and the response to LH-RH test (100 micrograms i.v.) became blunted by the treatment. No significant differences between the ovulated and desensitized groups were seen in basal LH, FSH, E2, PRL levels, LH/FSH ratio and response to LH-RH test performed prior to the treatment. In addition, the plasma LH-RH profile after subcutaneous injection of 10 micrograms of LH-RH was highly pulsatile in both groups. However, in the desensitized group, all were obese, showed impaired GH response to both insulin tolerance and GH-RH tests, and had episodic LH secretion with higher frequency compared to the ovulated group. These results suggest that the desensitized women had occult pituitary dysfunction and hypersecretion of endogenous LH-RH which stimulated the pituitary close to the level of desensitization. The mechanism of hypersecretion of endogenous LH-RH is discussed.

[Subcutaneous long-term pulsatile administration of LH-RH to patients with hypothalamic amenorrhea].

The effects of pulsatile LH-RH administration on ovulation induction and luteal phase activity were studied in 8 women with hypothalamic amenorrhea (2, with primary amenorrhea and 6 with secondary). Ten to 20 micrograms of LH-RH was subcutaneously injected by autoinfusion pump every 2 hours for more than 56 days. Six out of 8 women responded to the treatment and showed a total of 18 ovulatory cycles. The mean length of the follicular and luteal phases was 25.9 +/- 6.37 days and 11.9 +/- 0.43 days, respectively. The serum progesterone level in mid luteal phase was 12.0 +/- 2.35 ng/ml and was in the normal range for naturally cycling women. Two patients with secondary amenorrhea conceived in the 2nd and 3rd ovulatory cycle, respectively. In two patients who showed no response to LH-RH pulsatile administration for 56 days of treatment, serum LH and FSH levels were gradually lowered and the response to 100 micrograms of LH-RH iv injection was remarkably attenuated, suggesting that the pituitary gland was desensitized by the treatment.

Heme metabolism and turnover of cytochrome P-450 in tumor-bearing mouse livers.

Hepatic heme metabolism and in vitro translation of poly(A) + RNA from livers were studied to elucidate the mechanism of the reduction of microsomal cytochrome P-450 in tumor-bearing mouse livers. Hepatic delta-aminolevulinic acid synthase activity in male C57BL/6N mice (0.203 nmol/mg protein/h) at 8 days after the transplantation of Ehrlich ascites tumor cells was the same level as that of normal livers (0.206 nmol/mg protein/h). On the other hand, hepatic heme oxygenase activity of tumor-bearing mice (0.482 nmol/mg protein/10 min) had increased 8 days following i. p. transplantation of tumor cells when compared with that of normal mouse livers (0.296 nmol/mg protein/10 min). SDS-gel electrophoresis of in vitro translation products of poly(A) + RNA extracted from membrane-bound polysomes of the livers from tumor-bearing mice showed no significant differences from that of normal controls in the region of cytochrome P-450, that is, the molecular weight region of Mr = 47,000-60,000, although microsomal protein content in this region estimated by Coomassie-blue staining was reduced. These results suggest that the decrease of heme biosynthesis or the translation of cytochrome P-450 mRNA played a very small part in the reduction of microsomal cytochrome P-450 in tumor-bearing mice.

Recognition of the ribosomal RNA structures by purified nucleolar RNA methyltransferase.

Previously we have isolated the specific RNA methyltransferase from the nucleoli of Ehrlich ascites tumor cells. The purified enzyme was found to be specific for methylation of C5 position of cytosine residue in ribosomal RNA in vitro (Obara, 1982b). In the present study, we have investigated the recognition mechanisms of RNA structure by this enzyme from the points of view of both primary and secondary structures. Analysis of in vitro methylation product by ribonuclease T1 digestion indicated the methylation-site(s) was limited to a certain number of nonanucleotide. The next experiments with either Sl nuclease or actinomycin D and ethidium bromide suggested that the enzyme modified only cytidine residue in or located close to the double stranded part of RNA. On the other hand, the characterization of analogues of cytidine residue in the RNA at molecular level showed that the methylation of rRNA was inhibited by either cytidine, CDP or CTP, but little inhibition was observed in the presence of cytosine, 5-methylcytidine and CMP.

Isolation of immunochemically distinct form of cytochrome P-450 from microsomes of tulip bulbs.

A highly purified cytochrome P-450 was obtained from the microsomes of tulip bulbs (Tulipa gesneriana L.). The molecular weight (Mr = 52,500) and amino acid composition of this plant cytochrome P-450 are similar to those reported for rat livers. On the contrary, Ouchterlony double diffusion analyses indicated that cytochrome P-450 isolated from tulip bulbs shares no common antigenic determinants with those of 9 other plants, in spite of the presence of comparable contents of cytochrome P-450 and/or trans-cinnamate 4-monooxygenase with tulip bulbs.

[Antigenicity of the human trophoblast].

In this a rabbit immunological serum against human trophoblast was obtained after triple intramuscular superficial villi extract injection from chorion of 6--7 week pregnancy with Freund's adjuvant. In immunofluorescent indirect reaction to this serum made on chorion and placenta sections some most intense specific fluorescence was obtained in cytoplasm of primordial cyto- and synytiotrophoblast. Some favourable influence of antigenic differences between ovum and mother especially implantation has been suggested.
