Application of a computer program for the mapping of a gene locus to the disputed PGM3 localization on human chromosome 6.

The program GENEMAP, which performs the computations of a model for the mapping of a gene locus (Baur & Spitzer 1978), is applied to the localization of the phosphoglucomutase 3 with family data as published by Lamm et al. (1972), Rittner & Kalbheim (1975), Albert et al. (1977) and Kömpf et al. (1977). On the basis of Lamm's recombination fractions the combined odds were 13 : 1 in favor of an orientation of PGM3 to the HLA--B-side.

[The clinical significance of plasma glycoside concentrations in patients with cardiac pacemakers (author's transl)].

308 digitalized out-patients with artificial cardiac pacemakers were explored for signs of glycoside toxicity with simultaneous determination of digoxin plasma levels 12 hours after the last dose. The incidence of different side effects commonly attributed to overdigitalization did not allow prediction of toxic plasma levels. 55% of all glycoside levels were within the therapeutic range, 34% were below 0.7 ng/ml and only 11% above 2.0 ng/ml. With the most commonly prescribed maintenance doses of the glycosides used (digoxin 0.5 mg, beta-acetyldigoxin 0.4 mg, beta-methyldigoxin 0.2 mg, lanatosid C 1.0 mg) therapeutic plasma levels were reached regularly in 60-65% of the patients. A significant correlation existed between plasma glycoside concentrations and renal function as well as age, but glycoside concentrations could not be correlated with the age of the patients. There were no indications for interactions of the different glycosides prescribed with diuretics or oral antidiabetics.

[The action of stabilised pilocarpine drops on the human eye (author's transl)].

The use of certain additional substances enables preparation of pilocarpin solutions which remainstable for at least 4 years, while the usual pilocarpin solutions were subject to a relatively early decomposition. A double-blind study with 30 patients demonstrates that there is no difference between the pharmacologic effect at the pure and the modified pilocarpin solution, which justifies the use of solutions with added substances to stablize the effective component.

[Program for the computation of plausibilities of paternity by means of serological findings. II. Demonstration of examples (author's transl)].

By demonstrating three actual cases of disputed paternity the validity of the computer program described in the first part of this paper is shown, namely: the information with respect to possible unrecognized exclusions pointing to chances of extended family studies; the standard deviation and distribution curve giving an actual survey of statistical parameters of a given case of disputed paternity.

[Biological availability of diagoxin in patients with or without gastric resection (Billroth II) (author's transl)].

Biological availability of digoxin tablets was measured during maintenance therapy in ten hospitalized patients who had had a Billroth II gastric resection at least two years previously. Twelve patients on digoxin maintenance for heart failure but without gastro-intestinal disease served as controls. The mean value of daily digoxin urinary excretion over ten days in the resection group was 38.09 +/- 0.70% of the administered dose. The serum-digitalis level 12 and 24 hours after the last dose of glycoside (0.5 mg) was 1.30 +/- 0.04 ng/ml and did not significantly differ from that of the control group, nor did digoxin elimination in urine and the digoxin/creatinin excretion ratio. It is concluded that two-third gastric resection with exclusion of antigrade duodenal passage does not influence biological availability of digoxin given in tablets.

[The relation between genotypical and phenotypical plausibilities of paternity (author's transl)].

Applying the formula of Okajima [2,3] to the genotypical possibilities of a given phenotypical child-mother-putativ father constellation one can compute for each of these possibilities a genotypical probability of paternity. It is possible to determine the discrete distribution function, the mean value, and the variance of these genotypical probabilities of paternity and it is shown that the phenotypical probability of paternity is equal to the mean value of the genotypical probabilities of paternity.

Further data on the development of red blood cell antigens Lua, Lub, and Cob.

One hundred and fifty-five cord cells were tested for the red blood cell antigens Lua, Lub and Cob in order to collect data on the early postnatal expression of these markers. Additionally, dosage studies were carried out in 8 - 10-month-old heterozygous children. Antigens Lua and Lub revealed to be significantly less expressed in children of both ages compared with those of their mothers, whereas no such differences could be demonstrated in the expression of the antigen Cob.

Polymorphism of the human phosphoglycolate phosphatase in Northrhine-Westphalia (F.R.G.) and its application to paternity testing.

Blood samples from 563 unrelated German and from 110 Turkish individuals living in the Düsseldorf area were studied for phosphoglycolate phosphatase polymorphism. The distribution of the observed phenotypes in the population genetic study did not diverge from the expected values according to Hardy-Weinberg law. In our series, the gene frequencies were calculated as follows: (a) Germans: PGP1 = 0.851, PGP2 = 0.118, PGP3 = 0.031, (b) Turks: PGP1 = 0.973, PGP2 = 0.018, PGP3 = 0.009. The assumed autosomal codominant mode of inheritance was confirmed by the examination of 109 mother-child pairs and by analysis of 70 cases of disputed paternity. The plausibility to exclude German non-fathers from paternity is 12.78%.

Rates for tic disorders and obsessive compulsive symptomatology in families of children and adolescents with Gilles de la Tourette syndrome.

The aim of this study was to assess rates for tic disorders and obsessive compulsive psychopathology in families of children and adolescents with Gilles de la Tourette syndrome (TS). Diagnoses were based on the DSM III-R criteria. Obsessive compulsive psychopathology, that did not fulfill the criteria for obsessive compulsive disorder (OCD) was additionally assessed and termed obsessive compulsive symptoms (OCS). The authors hypothesized that comorbid OCD or OCS in TS patients predicts a higher familial loading with obsessive compulsive symptomatology. The study cohort included 87 patients with TS who were evaluated clinically and with the use of a structured psychiatric interview. All available parents (152/174; 87%), several sibs (49/93; 53%) and some second degree relatives (27/659; 4.1%) were also interviewed. For other first and second degree relatives the family history method was used. Familial rates for TS were clearly elevated. Rates for chronic tic disorders (CT) were considerably lower than in previous studies. Additionally, tic disorders not otherwise specified (TDNOS) were diagnosed in a substantial number of first degree (15/267; 5.6%) and second degree relatives (36/659; 5.5%). OCD in parents (4/174; 2.3%) did not occur in an above baseline rate. However, both OCD (14/87; 16.1%) and OCS (15/87; 17.2%) were frequently associated with TS in index patients. Interestingly, 10 of 16 fathers with OCS also had a tic disorder. Obsessive compulsive psychopathology clustered in families. It is concluded that genetic studies in TS could profit from adhering to a conservative diagnostic approach to both tic disorders and OCD. The familial clustering of OCS/OCD in conjunction with the elevated paternal rate for the co-occurrence of tic disorders and OCS might indicate heterogeneity of TS.

Linkage analysis in nuclear families. 1: Optimality criteria for affected sib-pair tests.

The affected sib-pair method can be applied to investigate linkage between a marker locus and a disease. Several statistics have been proposed to test if the observed pattern of marker alleles shared identically by descent (ibd) is compatible with the null hypothesis of no linkage. Here, we consider different optimality criteria for sib-pair linkage tests. While for recessive inherited diseases the mean test is found to be uniformly (in theta) most powerful, it can also be shown that, irrespective of the mode of inheritance, the mean test is the locally optimal test.
