[Detection of the measles virus genome in the peripheral blood lymphocytes of patients with chronic and acute glomerulonephritis].

The presence of the measles (rubeola) virus genome was searched for in the lymphocytes from the peripheral blood of patients suffering from the acute (16 persons) or chronic (164 persons) glomerulonephritis. Dot hybridization technique with the plasmid borne probes to the measles viral genes NP, P and H have been used for the search. The measles viral genome has been detected in 58% of lymphocytes from the patients with the chronic glomerulonephritis and in 50% of lymphocytes from the patients suffering from the acute form of the disease. The genome was not found in the material from the control group including donors and traumatology ward patients. 25 samples of lymphocytes from the patients with the chronic glomerulonephritis contained the RNA that was not hybridizable with the viral genes probes by dot hybridization technique, thus containing no genes homologous to parotitis viral genes. The average titer of anti-measles antibodies in the serum from patients with chronic glomerulonephritis the lymphocytes of which contained the measles viral genome was 1:304, while it was 1:154 for patients with the negative probes. The average anti-measles antibodies titers are the same (1:166 and 1:142) for analogical groups of patients with acute form of disease.

[Immunoenzyme analysis of adenovirus hexons. Indication and characteristics of rabbit and mouse antibodies against hexons].

An enzyme-linked immunosorbent assay (ELISA) was developed for analysis of rabbit and mouse IgG antibodies specific to adenoviral hexon. The anti-hexon antibodies were detected by capture with purified hexon coated onto polystyrene microtiter plates and visualizing them by respective anti-IgG horseradish peroxidase conjugates. In the sera from hyperimmunized rabbits and mice as well as in the mouse ascite fluids the ELISA procedure revealed primarily type-specific (epsilon) and genus-specific (alpha) antigenic determinants in hexon but not those of intermediate specificities.

[Immunoenzyme analysis of adenovirus hexon. Determination of antibodies from hyperimmunized chickens].

We have elaborated three systems of enzyme-linked immunosorbent assay (ELISA) for detection of chicken IgG antibodies specific for hexon antigens of three immunologically distinct adenovirus groups: those of mammalian adenoviruses (Mastadenovira), typical avian adenoviruses (Aviadenovira) and of egg-drop syndrome-76 (EDS-76) virus. In each system the antibodies against respective hexons were specifically detected. In mammalian adenovirus hexons the ELISA detects primarily the type-specific (epsilon) and genus-specific (alpha) antigenic determinants. The time course of anti-hexon antibodies content was followed during immunization. The level of anti-hexon antibodies in egg yolk reflects adequately their content in blood serum. The technique is suitable for serological diagnosis of chicken adenoviral infections as well as for characterization of egg-yolk antibodies obtained by preparative hyperimmunization of hens.

[Study of the antiviral activity of a poly I : poly-C complex with poly-L-lysine in monkeys].

Antiviral activity of poly-I-poly-C complex with poly-L-lysine was studied on macaco rhesus. The complex bifilamentous polyribonucleotide induced active production of serum interferon and provided pronounced protection of the monkeys infected intracutaneously with the variolovaccine virus (10 LD50 for the monkeys in intracutaneous infection). The effectiveness of the protective effect depended on the scheme and route of the drug administration. The highest prophylactic and therapeutic effect was provided by local administration of the complex in a dose of I mg per I kg of the body weight, the incubation period being increased 2--3 times and the period of the skin affections being decreased approximately 2 times. The results of the studies on the effect of poly-I-poly-C complex with poly-L-lysine were evident of definite prophylactic activity of the drug against experimental vernal encephalitis in the monkeys. The animals not treated with the inductor died on the 16th or 17th day after infection because of the paralysis of the trunc and extremities muscles. The clinical evidences of the disease in the animals treated with the drug were not uniform: from complete health to death.

[The persistence of the distemper virus in continuous human cells].

Two cultures chronically infected with distemper virus (HEP-2 and L-41) were obtained. The cultures produced a small-plaque cell-associated virus and a virus-specific antigen which was demonstrated by the fluorescence antibody technique in 40%-60% of the cells. The chronically infected cells produced interferon as judged by their resistance to superinfections with heterologous viruses. The virus-carrier state was characterized by temperature sensitivity.

[Experimental measles infection in rodents].

The sensitivity of newborn hamsters to inoculation with the vaccine L-16 strain of measles virus and the Lec strain isolated from a patient with subacute sclerosing panencephalitis as well as the possibility of persistence of these viruses in the animals were studied. Intracerebral inoculation of the L-15 strain was shown to produce in hamsters acute meningoencephalitis leading to death in 85%-100% of cases. Over 30 days after inoculation, the infectious virus, the virus-specific antigen and virus genome were found in the brain. In the brains of the sick animals, all the structural proteins of measles virus with the exception of hemagglutinin were expressed. After inoculation with the Lec strain, the clinical signs of the disease were less manifest, and mortality was 40%. The infectious virus could be detected in the brain up to 20 days postinoculation, the genome, up to 31 days. All the structural proteins of measles virus were expressed in the brains of the inoculated animals. No persistence of L-16 and Lec strains of measles virus could be demonstrated at langer intervals after inoculation (90-180 days) in the brains of hamsters.

[Avian CELO adenovirus as a possible contaminant in cell cultures of Japanese quail embryos].

Avian adenovirus CELO was found to replicate poorly in Japanese quail embryos (JQE) and cell cultures of them. The infectious process in these systems was latent. The antigen of adenovirus CELO in JQE cell culture was detectable by the fluorescent antibody method (FAM) within the first 24-72 hours after inoculation as fluorescent cytoplasmic granules. Subsequently, fluorescence of nuclei and macrophage cytoplasm was observed. The results indicate that JQE and their cell cultures are not contaminated with avian adenovirus CELO despite regular circulation of this agent among avian populations. The advantages of FAM (rapidity and clearness) for identification of adenovirus as substrates contaminant as compared with other biological methods have been demonstrated.

[Influence of a complex of poly(I). poly(C) and poly-l-lysine on the course of vaccinia in monkeys].

Experiments of 22 Macaca rhesus monkeys were carried out to study the interferon-inducing and antiviral activity of poly(I) - poly(C) and of its complex with poly-l-lysine. The complexed double-stranded polyribonucleotide induced active production of serum interferon and markedly protected the monkeys inoculated intradermally with vaccinia virus (10 monkey ID50 by intradermal inoculation). The effectiveness of the protective effect depended on the schedule and routes of administration of the preparation. The greatest prophylactic and therapeutic effect was achieved by local administration of the complex in a dose of 1 mg/1 kg of body weight. This also prolonged the incubation period by 2-3 times and reduced the duration of persistence of skin lesions approximately by half. By the intravenous route, the best protection was achieved by 2 injections of 2 mg/kg at an interval of 96 hours. Four daily injections of the complex exerted virtually no effect on the course of vaccinia infection. The animals receiving the complexed poly(I) - poly(C) developed virus-neutralizing antibody to the same titres as control animals and were resistant to reinfection with vaccinia virus. A second injection of the complexed poly(I)-poly(C) 96 hours after the primary inoculation induced the same interferon production as the initial administration of the preparation. The monkeys inoculated intravenously with 2 mg/kg poly(I) - poly(C) showed no interferon in their blood serum and were also poorly protected against vaccinia virus infection.

[Effect of a poly I-poly C complex with poly-L-lysine on experimental tick-borne encephalitis].

The influence of the poly(I)-poly(C) complex with poly-l-lysine on the development and course of experimental tick-borne encephalitis was studied in Macaca rhesus monkeys. Prophylactic administration of the substance under study, although not preventing the disease, modified its course and facilitated recovery of the animals. Poly(I)-poly(C) and its complex with poly-l-lysine were shown to have no effect on production of infectious virus or its antigen demonstrable by the immunofluorescence procedure in HEp-2 cells chronically infected with tick-borne encephalitis virus.

[Smallpox vaccination with protection by interferon and an interferon inducer].

The vaccinal process was studied in monkeys vaccinated with smallpox vaccine under the protection of fibroblastic or leukocytic interferons and modified poly(I) . poly(C). Simultaneous vaccination and administration of one of the studied preparations induced intense immunity and at the same time prevented viremia and alleviated local inflammatory reactions caused by the vaccination. The protective effect of the fibroblastic interferon was more marked than that of the leukocytic one in the vaccinal process. When the monkeys received the preparations intramuscularly the leukocytic but not the fibroblastic interferon was detected in the blood.

[Interferon production by various means of administering modified poly I-C].

Modified poly(I) . poly(C) induces appreciable amounts of interferon in Macaca rhesus monkeys not only after intravenous but also after intramuscular, endolumbar, and subconjunctival inoculations. The time course and intensity of interferon production after various routes of the induced inoculation were similar. Maximum interferon production was found at the inoculation site, upon topical application the resistance to reinoculation developed after primary inoculation and persisted for 72 hours, After intravenous inoculation of the doses of the inducer tested the resistance to reinoculation of the modified polyribonucleotide developed after two inoculations made at an interval of 24 hours. A minimal interferon-inducing dose of the modified polyribonucleotide was 10 micrograms/kg for mice and 100 micrograms/kg for Macaca rhesus monkeys.

[Effect of phosphonoacetic acid on experimental vaccinia infection].

The effect of phosphonoacetic acid (PAA) on reproduction of vaccinia virus in chick embryo fibroblast cultures and on the development of vaccinia virus infection in rabbits was studied. In vitro the preparation was tested as a solution, in vivo as a solution and ointment. In rabbits, the preparation was applied epicutaneously and inoculated subcutaneously and intravenously. All these routes were found to be effective. A single application conferred a much lower protective effect than daily administration for 4 days. Poly (I).poly (C) administered subcutaneously was found to be more effective than PAA. The latter in tissue culture exerted a marked inhibiting effect when used in a dose of 100 microgram/ml.

[Interferon induction in monkeys by poly I-C complex with poly-L-lysine].

The spectrum characteristics, the stability to pancreatic RN-ase and hydrolases of the monkey and mouse blood as well as interferon induction in mice and monkeys by poly I: poly C and its complex with poly-l-lysine were studied. The complex was found to be more stable to the treatments tested as well as markedly interferonogenically active in Macaca rhesus monkeys.
