Catalepsy induced by carbachol microinjected into the pontine reticular formation of rats.

Rats were implanted stereotaxically with permanent guide tubes aimed at the pontine reticular formation. Carbachol was microinjected in the awake, freely moving rats. Catalepsy was evaluated with the horizontal bar test. Intense and long-lasting, dose-dependent catalepsy was observed following microinjections of 2-8 micrograms of carbachol. Pretreatment with intraperitoneally administered scopolamine significantly reduced the cataleptogenic effect of carbachol. These results show that the pontine reticular formation is part of the neuroanatomical substrate of catalepsy induced by cholinergic drugs.

Molecular analysis of the Drosophila nuclear lamin gene.

A complete nucleotide sequence of a 4.2-kb genomic fragment containing the Drosophila lamin gene and flanking sequences is presented. Primer extension experiments and sequence analysis revealed that transcription starts from a single promoter. The lamin maternal 2.8-kb transcript and the 3.0-kb zygotic transcript are generated from two alternative polyadenylation sites. The gene contains four exons. The first intron is 7 bp upstream of the first AUG site. The two other introns are located within the alpha-helical rod domain of the protein: one in coil 1B in the 42-amino-acid domain that is absent in vertebrate cytoplasmic intermediate filament proteins and the other in coil 2 at a position different from intron positions within the vertebrate intermediate filament genes. Together with the sequence homology analysis, the data suggest either that the lamin gene was the ancestral gene of intermediate filament genes or that the lamin gene diverged from other intermediate filament genes early in evolution.

[Migraine in childhood: lipid metabolism and its implications].

Hyperlipemia has been recently described in children with migraine, being suggested that this alteration is in the base of the disease. In 35 children with intense migraine we have measured the plasmatic levels of cholesterol, triglycerides and lipoproteins. In all patients the triglycerides and LDL levels were normal; the cholesterol levels were elevated in 4 children; in 8 it was observed an elevation of HDL fraction and in 11 VLDL was the elevated fraction. Facing this results, we think that hyperlipemia and hypercholesteremia are not in etiologic relation with the migraine in children.

Carnitine concentrations in dialysed and undialysed patients with chronic renal insufficiency.

Free carnitine, acylcarnitine and total carnitine serum concentrations have been measured in chronic renal insufficiency patients under conservative treatment, in patients under regular haemodialysis treatment and in healthy controls. In the undialysed patients the levels of free carnitine, acylcarnitine and total carnitine were all clearly higher than those of the control group. The free carnitine and total carnitine levels of undialysed subjects were also higher than in regularly haemodialysed patients, showing that dialysis produces plasma carnitine losses that are not compensated for by endogenous synthesis of carnitine (this finding supports published reports of tissue carnitine deficiency in patients undergoing regular haemodialysis). The acylcarnitine levels of dialysed and undialysed patients were not significantly different, however; both were very much higher than that of control group. The hypercarnitinaemia of the patients under conservative treatment suggests that the impairment of renal function causes a reduction in the elimination of carnitine via the kidney.

[Comparative study between CPK, LDH and their isoenzymes in the detection of carriers of Duchenne's type muscular dystrophy].

Results of a comparative study of serum activity levels of creatine kinase, lactodehydrogenase and their isoenzyme in mothers of 13 children with Duchenne progressive muscular distrophy are presented. CPK sensibility was 100%, while that of LDH1/LDH2 and LDH5 were 23% and 38,4% respectively. It is concluded that CPK has a great liability index, and that it is superior to LDT and their isoenzyme for detection of carriers of Duchenne progressive muscular distrophy.

[Allopurinol in progressive muscular dystrophy (author's transl)].

Authors have treated with allopurinol ten children with progressive muscular dystrophy, whose ages were between 15 months to eleven years. An initial and monthly clinical evaluation of muscular strength, Gowers's sign and pseudohypertrophy was performed. Monthly determinations of CPK, LDH, aldolase, GOT and serum urate; every two months electromyography was also made. In three patients a complete recovery of strength, Gowers's sign was found and at the same time pseudohypertrophy diminished; the age average was three years and four months. In four patients a partial recovery was found; the age average was nine years and four months. In three patients there was no answer to treatment, they were contracted and had deformities, being age average ten years. No variations on monthly controls of CPK, LDH, aldolase and GOT were observed. On these facts their opinion is that allopurinol is an effective drug in the treatment of progressive muscular dystrophy and that its' effectivity is better when treatment is initiated early and, of course, before patients present deformities and contractures.

Diagnosis of rupture of the fetal membranes by glucose and fructose measurements.

A new method is presented for differentiating amniotic fluid from cervical mucus, based on differences in the respective carbohydrate concentrations present in these fluids. Glucose and fructose are present in high concentrations in cervical mucus during pregnancy, average values being 240 and 30.4 mg/100 ml, respectively. Their concentrations in amniotic fluid, however, are low, averaging 39 and 3.3 mg/ml, respectively. Examination of 215 women between 36 and 42 weeks' gestation, using either glucose or fructose values, revealed a high degree of discrimination between amniotic fluid and cervical mucus. The best results, however, were obtained by employing the calculated linear sum 3.32 log glucose plus log fructose for each sample. Inaccuracy rates were low in comparison to those of other diagnostic methods, such as pH values, ferning, and Nile blue sulfate staining. A further advantage of the method is that it remains fairly accurate even when the time interval between rupture of the membranes and the investigation exceeds 24 hours.

New studies on placental transfer of fosfomycin.

Passage of fosfomycin across the placental barrier and the dynamics of maternal and fetal levels were determined in a group of ten mothers in active labor. To obtain a series of intrauterine fetal samples the Saling technique was used. There exists a clear relation between the levels reached in the mother and in the fetus and in function of time, as well as between the amount of antibiotic in the fetal blood 90 min after injection, and the weight of the placenta.

Enteropathogenic E. coli gastroenterocolitis in neonates treated with fosfomycin.

The effect of calcium salt of fosfomycin in the treatment of 43 neonates suffering from acute gastroenterocolitis produced by enteropathogenic E. coli is evaluated. The minimal inhibitory concentration of these E. coli was, generally, lower than 128 mug/ml. Dosages of 150-200 mg/kg body weight/day were administered orally every 8 h. This treatment lasted for 4 days only. Clinical evolution was favorable in 38 (88%) babies and bacteriological evolution in 30 (70%). In eight cases a different flora to the initial was selected during the treatment with fosfomycin. None of the cases treated showed any toxic alteration attributed to the antibiotic.

Fosfomycin in treatment of respiratory bacterial infections.

A study was made of 40 patients suffering from clinically and bacteriologically demonstrated bacterial respiratory infections which were all treated with fosfomycin as the only antibiotic. Studies were made of the sensitivity of the isolated organisms and control of the eradicated and selected organisms. Analytical controls were performed on all patients before, during and after treatment. The average dose employed was 60 mg/kg/day, divided into four doses and administered parenterally. The duration of the treatment fluctuated between 6 and 11 days in 87% of the cases. A clinical and bacteriological evaluation was made of the results. The clinical response was satisfactory in 75% of the cases, partial in 7.5% and poor in 17.5%. The bacteriological evolution was satisfactory in 62.5%, partial in 12.5% and poor in 20%. It was not possible to control the evolution in 15% of the patients. The concentrations of the antibiotic were determined in the sputum of 6 patients and fluctuated between 0.6 and 12.9 mug/ml. In no case could any significant disorders be attributed to the antibiotic.

Influence of enteral oxygen administration on the slow electrical activity of the intestine and stomach.

Thirty-eight starved cats, anesthetized with pentobarbital (Nembutal) sodium, received enteral administration of oxygen and other gases; the effect on intestinal motility as expressed by electrical activity was measured. Oxygen caused a notable increase in amplitude and frequency of slow electrical waves, while carbon dioxide and nitrogen caused no visible alterations. Phentolamine hydrochloride and propranolol hydrochloride together with atropine sulfate decreased the amplitude and frequency of oscillations to near zero; subsequent administration of enteral oxygen caused a notable increase in electrical activity, while enteral carbon dioxide produced no alterations. On the basis of our previous observations that enteral oxygen enhances oxygen and blood delivery to the gut wall, we suggest that the rise in stomach and intestinal electrical activity during enteral oxygen administration in conditions of surgical stress may be associated with an increase in oxygen and blood delivery to the gut wall.

Fosfomycin in treatment of chronic urinary tract infection.

A study was made of fosfomycin in the treatment of chronic urinary infection in 51 patients. 72% of the 67 bacteria isolated and tested in the laboratory with discs containing 50 mug of fosfomycin were found to be sensitive to the antibiotic. The majority of E. coli, Klebsiella, P. mirabilis and Ps. aeruginosa strains were sensitive to the antibiotic. Generally, 60 mg of fosfomycin/kgbody weight was administered intramuscularly 6-hourly for 7-15 days. A clinical and bacteriological cure was obtained in 39 patients (76%), and after 4-6 weeks in 31 patients (61%). Blood and urine fosfomycin levels were determined in seven patients, three with normal renal function, three with moderate renal insufficiency, and one with severe renal insufficiency. The antibiotic levels and elimination time maintained a clear relation with the degree of renal insufficiency.

[Analysis of a Yersinia enterocolitica isolated from human diarrheic feces in Argentina].

Some serotypes of Yersinia enterocolitica might cause diarrheas and/or invasive infections. The aim of this work was to analyze virulence factors and associated markers in a strain of Y. enterocolitica isolated from human diarrheic feces. The strain analyzed was included in the biotype 1A. The virulence markers determination as well as the search of the genes vir F and ail, were negatives. However, it was demonstrated enterotoxin production at 20 degrees C, and at 37 degrees C in osmolarity conditions and pH similar to the human intestine. The enterotoxin presented reactivity for the infant mouse test, although it could not be proven the presence of yst gene by PCR. The results obtained by us, coincident with those of other investigators, indicated that certain clinical isolates of Y. enterocolitica of the biotype 1A ("avirulent"), could be the etiological agent of the illness through other mechanisms of virulence, that would differ from those previously characterized in species of enteropathogenic Yersinia.
