Comparative absorption kinetics of imidazole and salicylic acid in volunteers after administration of ITF 182 tablets and suppositories.

Comparative absorption kinetics in volunteers of a new antiinflammatory drug (Selezen), in the form of 750 mg tablets and suppositories, were studied. The two components of the drug, imidazole and salicylic acid were found in plasma. Pharmacokinetic parameters were calculated according to a first order absorption. Salicylic acid showed a maximum concentration 59.2 +/- 5 min and 75.4 +/- 7.6 min after the administration of the tablet and suppository respectively; and imidazole after 86.3 +/- 10.8 min and 75.2 +/- 5.4 min, respectively.

Zetidoline, a specific dopamine antagonist devoid of antiserotonergic activity in rodent brain.

In the present study the interaction of zetidoline, a new antipsychotic, with the serotonergic system of the rodent brain was investigated in comparison with known neuroleptics. The pharmacological tests were L-5-hydroxytryptophan syndrome in mice and tryptamine-induced clonic seizures in rats. Biochemical tests were the in vitro binding of 3H-zetidoline to rat brain membranes and the serotonin metabolism and turnover in vivo in the rat cerebral cortex and brainstem. The results indicate that zetidoline, unlike haloperidol and chlorpromazine, at doses higher than those active on the dopaminergic system, shows no or very weak antiserotonergic activity in behavioral tests.

A cytogenetic study on workers exposed to low concentrations of benzene.

A cytogenetic study was performed on 22 healthy workers engaged in benzene production and exposed to low concentrations of benzene, ranging from 0.2 to 12.4 p.p.m. (threshold limit value 10 p.p.m.). Workers were divided into two groups according to the different levels of exposure, inferrable also from the concentration of benzene in the alveolar air and the levels of urinary phenols. Each exposed subject was paired with a suitable control, living in the same area and of similar smoking habits and age. No statistically significant increase of sister chromatid exchange (SCE) frequency was observed in the exposed groups. In controls, SCE frequency was positively correlated with age and smoking habits. Among structural chromosomal aberrations, only the chromosome-type ones were significantly higher in exposed than in control subjects, and their increase was still significant when gaps were discarded.
