We report the application of a sandwich technique in flow injection systems which afford low consumption of expensive reagents and two reagent recirculation systems. The potential applicability of the technique thus developed was assessed by determining glucose in serum samples by the enzymatic glucosidase/peroxidase method. It was possible to perform up to 450 determinations with the same amount of reagent used to perform 50 determinations by batch procedures. The sampling rate was 80 determinations per hour with a 0.9% relative standard deviation.
Legionella pneumophila has been shown to survive and multiply in a variety of intracellular environments, including protozoa and human mononuclear phagocytes. However, the mechanism by which this organism acquires iron in the intracellular environment has not been studied. Since L. pneumophila does not produce siderophores, alternative methods of iron acquisition were investigated. Virulent strains of L. pneumophila were able to grow in media containing as little as 3 microM iron, whereas avirulent cells required a minimum of 13 microM iron for growth. Neither virulent nor avirulent cells were able to utilize 55Fe bound to transferrin. When incubated in the presence of 55Fe in the form of ferric chloride, both virulent and avirulent cells accumulated equal amounts of iron. The uptake of iron was energy dependent as indicated by inhibition of 55Fe uptake at 4 degrees C and preincubation of the cells with KCN. Treatment of virulent cells with pronase or trypsin had no effect on iron uptake. In contrast, pronase or trypsin treatment of avirulent cells resulted in increased uptake of iron. Iron reductase activity in both virulent and avirulent cells was demonstrated, with the highest specific activity associated with the periplasmic fraction. Maximum reductase activity of virulent cells occurred with NADH as the reductant. In contrast, avirulent cells showed a twofold increase in enzyme activity when NADPH was used as the reductant. These results suggest that an iron reductase is important in iron acquisition by L. pneumophila.
Several big agricultural fermentation plants for producing of "biogas" are in operation in the GDR. Problems of distances to other objects, utilisation of methane and storage of liquid manure (slurry) are discussed. Some results of experiments about the immissions in the environment are demonstrated. They have point out, that the emissions of microorganisms and viruses cannot be demonstrated in a distance of more than about 200 meters. It is the most important hygienic acknowledge, that the distance of organoleptic observation is always greater than the epidemiologic or sanitary one. Therefore the production of energy by anaerobic fermentation of liquid manure and sewage sludge seems to be an useful way from hygienic point of view.
Various biological materials were tested for their growth-promoting activity of several bifidobacterial species in a synthetic medium containing ample sources of inorganic salts, vitamins, nitrogen, and carbon. It was found that only Bifidobacterium adolescentis and B. longum (ATCC 15708) grew optimally or near optimally in the synthetic medium. All the other bifidobacteria tested grew optimally only in the synthetic medium supplemented with a growth promoter. The best growth promoters for all bacteria were bovine casein digest and yeast extract rather than human milk whey. Other growth promoters, including human and bovine milk wheys, hog gastric mucin, and bovine serum albumin digest were effective with some bacterial species but not with others. Bifidobacteria also grew well when the bovine casein digest (20 mg/ml) was used as the nitrogen source. Only the yeast extract was able to improve growth under these circumstances. The nature of these growth factors has not yet been determined.
Ferrous iron uptake studies in Bifidobacterium bifidum var. pennsylvanicus were carried out in a well-defined salt solution termed "modified Hanks solution" at both high iron concentrations (LAFIUS conditions) and low concentrations (HAFIUS conditions). Various divalent metals, Mn2+, Zn2+, Ni2+ and Cu2+, inhibited iron uptake under HAFIUS conditions in a non-competitive manner, and in a pseudo-competitive manner under LAFIUS conditions. Cr2+ had no effect. Co2+ inhibited iron uptake competitively under HAFIUS conditions. Metabolic affectors that inhibited iron uptake both under HAFIUS and LAFIUS conditions were: tetraphenylphosphonium chloride, diethylstilbesterol, vanadate, carbonylcyanide-m-chlorophenyl-hydrazone, and a mixture of valinomycin and nigericin. Substances that stimulated iron uptake were KCl, valinomycin, and nigericin. Iron uptake under LAFIUS conditions in piperazine-buffered modified Hanks solution was higher than that in the acetate-buffered solution, and acetate inhibited iron uptake in the piperazine buffer. HAFIUS showed no difference. It is concluded that iron uptake in bifidobacteria is driven by an ATPase-dependent proton-motive force and that both the pH gradient and membrane potential are involved in this process. Mn2+, Zn2+, Ni2+, and Cu2+ may be transported via LAFIUS, but not HAFIUS. HAFIUS may transport only Co2+ in addition to Fe2+.
