Enhancement of haemopoietic recovery in sublethally gamma-irradiated mice by the joint use of indomethacin and cystamine.

The effects of indomethacin and cystamine, administered prior to gamma-irradiation of mice, were investigated. Synergistic enhancement of haemopoietic recovery was observed under the conditions of a joint use of both drugs. The reason for this synergistic action is probably a joint operation of two mechanisms, i.e., the lowering of the radiosensitivity of haemopoietic stem cells by cystamine and the increase of the proliferation of surviving haemopoietic cells by indomethacin.

Therapeutic effects of the joint administration of magnesium aspartate and adenosine monophosphate in gamma irradiated mice.

The joint administration of magnesium aspartate and adenosine monophosphate, injected on days 1 to 4 post radiation, has been found to exert stimulatory effects on the recovery of hemopoietic functions in sublethally gamma irradiated mice. These therapeutical effects were enhanced in animals protected by peroral administration of cystamine. The treatment scheme used did not modify survival of lethally irradiated mice. The therapeutic effects of magnesium aspartate and adenosine monophosphate in sublethally irradiated mice are explained by the stimulatory action of these drugs on the cell adenylate cyclase system, which influences the erythropoietic functions.

Effect of indomethacin, diclofenac sodium and sodium salicylate on peripheral blood cell counts in sublethally gamma-irradiated mice.

Treatment with indomethacin and diclofenac sodium was found to increase granulocyte counts in the blood of sublethally gamma-irradiated mice. Treatment with sodium salicylate was ineffective in this respect, administration of sodium salicylate together with indomethacin even decreased the indomethacin-induced effects. The results suggest that the hemopoiesis-stimulating effects of non-steroidal anti-inflammatory drugs cannot be correlated with the anti-inflammatory activity but rather with the side effects of these compounds, including the action on gastro-intestinal prostanoid production. This conclusion doubts on the possibility of the usefulness of non-steroidal anti-inflammatory drugs in conditions of the radiation syndrome.

[Formation of the papillary surface of the hands and feet in prenatal development in man].

The development of the papillary terrain on the fingers and toes, palms and soles was studied in 36 fetuses of gestational age ranging from 9 to 26 weeks. Histologic methods on serial sections and studies of the dermal surface of the skin after removal of the epidermis (according to Okrajima) were the main approaches used. The morphogenesis of papillary crests is described at different developmental stages and factors involved in their origin are discussed.

[Long-term study of disorders of heart rhythm in patients with chronic diseases of the respiratory system].

The authors investigated the incidence of arrhythmias by the method of continuous recording of ECG tracings according to Holter in 16 patients with chronic respiratory diseases three times in the course of two years. The incidence of arrhythmias was very frequent, more serious disorders (class 3 or a higher class according to Lown's classification) were very rare. The authors provided evidence of a great variability in the incidence of arrhythmias in the same patients during different periods of investigation. They did not reveal a relationship between the incidence of arrhythmias and the degree of functional affection of the lungs, the severity of hypoxaemia and the drop of the maximum aerobic capacity of the lungs. They did not find a relationship between the incidence of arrhythmias and the pressure in the pulmonary artery, or the size of the right or left ventricle or the thickness of their wall. The authors do not consider the finding of arrhythmias in patients with chronic lung disease an unequivocally serious finding, as reported sometimes in the literature.

Interstrain differences in the responsiveness of mice to glucan with respect to hematological effects and manifestations of late damage.

A study was made of the effect of a single s.c. injection of 4 mg glucan in the mouse strains C57BL/10, C3H and a hybrid population of mice. In mice of the strain C57BL/10 high responsiveness was demonstrated, manifested in a long-term increase in the granulocyte counts in the peripheral blood. These effects were accompanied by a fall in the mean red cell volume and the mean corpuscular hemoglobin, a fall in body weight, and mortality. 10 weeks after administration of glucan histology revealed necrotic foci in the livers of animals of the C57BL/10 strain, and an increased amount of the fibrous connective component in the pulmonary interstitium. The strain C3H exhibited a low responsiveness in respect of the hematological indices, while the hybrids did not show any significant response.

In vivo studies of the relationship between the activation of lipid metabolism, postirradiation bone marrow cell proliferation and radioresistance of mice.

The effect of adaptation to intermittent feeding on the in vivo biosynthesis of fatty acids and total lipids in the epididymal adipose tissue, the liver and the bone marrow was studied in adult male mice (CBA/JPh x C57BL@10 ScSnPh)F1. At the same time the effects of the same experimental stimulus on the rate of regeneration (proliferation) of bone marrow cells after sublethal irradiation of animals and on the overall radioresistance of mice expressed as 30 days survival after whole-body gamma irradiation were determined. Intermittent feeding in mice has been shown to have a significant effect on the biosynthesis of fatty acids and total lipids in all the tissues studied, including bone marrow cells, the intensity of the effect being closely dependent on the duration of the experimental stimulus. Maximum stimulation of lipogenesis during realimentation was observed approximately within 1 week of adaptation, with a reduction of the metabolic responses thereafter. The intensity of bone marrow cell proliferation in mice irradiated in the realimentation phase was inversely proportional to the preirradiation degree of biosynthesis of fatty acids and total lipids: in a period of lower lipogenetic capacity of cells in the tissue studied (around the weeks 2-5 of adaptation) an increase in the regeneration potential of bone marrow cells was observed together with increased radioresistance of the mice. During the 1-week of adaptation the opposite proved to be the case. Attention is drawn to the possible participation of prostaglandins and lipid peroxides in the responses observed.

Effect of indomethacin on the proliferative and differentiation activity in the haemopoietic system of lethally irradiated mice after syngeneic bone marrow transplantation.

The effects of indomethacin treatment on the proliferation and differentiation of haemopoietic stem cells of bone marrow grafts in lethally irradiated mice were investigated. Indomethacin was given subcutaneously, on days 3 to 5 after irradiation, in 6 doses of 0.05 mg per mouse. On day 6 after irradiation, an increased self-renewal of the settled colony-forming cells and an increased amount of differentiated cells were observed in the spleens of the mice treated. Simultaneously, a decreased maturation of the erythroid cells was demonstrated. The effects observed may be explained by the indomethacin-induced inhibition of prostaglandin biosynthesis.

Pyrophosphate scintigraphy and other non-invasive methods in the detection of cardiac involvement in some systemic connective tissue diseases.

Thirteen patients with systemic lupus erythematosus, 8 patients with polymyositis, and 6 patients with spondylitis ankylopoetica (Bechterew's disease) underwent clinical cardiologic examination and scintigraphy of the myocardium (99mTc-pyrophosphate), ECG, echocardiography, polygraphy, and their blood pressure was taken. The aim of the study was to ascertain how such a combination of non-invasive examinations can help in recognizing a cardiac involvement. In systemic lupus erythematosus cases one or more positive findings were revealed in 9 patients (69%), in 4 patients all examinations were negative (31%). Four patients (50%) with polymyositis had positive findings. In patients with spondylitis ankylopoetica positive findings occurred in 2 cases (33%). The study has shown that a combination of non-invasive cardiologic methods increases the probability of detecting cardiac involvement in systemic connective tissue diseases.

Separation of human lymphoid cells by affinity chromatography and cell surface labelling by hydroxyethyl methacrylate particles using monoclonal antibodies.

The hydroxyalkyl methacrylate gel Separon, Sepharose 6MB and polystyrene dishes with bound antibodies were used for affinity chromatography of cells isolated from human tonsils and peripheral blood. Enrichment of T cells by filtration through anti-human immunoglobulin Separon or Sepharose 6MB columns was comparable to that achieved by "panning" on dishes or by Nylon wool column fractionation. High enrichment of B cells was obtained when cells pretreated with monoclonal anti-T cell antibodies were fractionated on columns or dishes where anti-mouse antibody was bound. Hydroxyethyl methacrylate copolymer microspheres were conjugated with monoclonal antibodies directed against surface markers of human lymphocytes or with anti-mouse immunoglobulin antibodies and used for surface labelling of cells. The results obtained by immunofluorescence were in good correlation with labelling by hydroxyethyl methacrylate immunobeads.

Lactosamine type asialooligosaccharide recognition in NK cytotoxicity.

Inhibition of pig NK cell activity by asialooligosaccharides (aOS) isolated from human serum glycoproteins was investigated. Train-tennary aOS (aOSIII) of ceruloplasmin was found to be the most potent inhibitor up to the concentration 0.1 micrograms/ml, which is in agreement with its highly specific binding to NK-activity-enriched pig lymphocytes (with a morphology similar to human large granular lymphocytes (LGL]. Only lectins with the specificity to Gal(beta 1----4)GlcNAc or Gal(beta 1----3)GalNAc structures exhibited inhibition of NK cytotoxicity. F(ab)2 fragments of rabbit antibodies against pig spleen membrane lectin cross-reacting with the pig liver membrane lectin completely inhibited NK activity when preincubated with the effectors or present in the incubation mixture during the assay. These data suggest that lectin receptors on cells of pig NK-activity-enriched fraction specific for aOSIII and antigenically related to membrane lectins isolated from pig spleen and liver, are involved in the NK recognition of several xenogeneic targets.

Therapeutic efficiency of olaquindox in gamma-irradiated mice.

The antibacterial agent olaquindox (2-[N-(2-Hydroxy-ethyl)carbamoyl]-3-methyl-quinoxalin-1,4-dioxide) given in tap water at daily doses of 0.2 to 0.5 mg exerts beneficial effects on the body weight decrease and survival of gamma-irradiated mice. Beneficial effects are achieved especially by the postradiation administration of olaquindox. It is presumed that the mechanisms of the therapeutic effects of olaquindox are connected with its action on the composition of intestinal flora of mice.
