Anti-inflammatory steroids without pituitary-adrenal suppression.

When two new steroids, methyl prednisolonate and methyl 20-dihydroprednisolonate, were applied locally their anti-inflammatory activities were nearly equivalent to those of the parent compound prednisolone in the cotton pellet granuloma bioassay. However, when these two derivatives were administered systemically, their anti-inflammatory activities were weaker than those of the parent compound. Furthermore, unlike the parent compound, these new anti-inflammatory steroids did not suppress pituitary-adrenal function or cause liver glycogen depletion in rats.

Local antiinflammatory activity of acid ester derivatives of prednisolone.

The antiinflammatory activity of two steroid acid esters, methyl prednisolonate and methyl 20-dihydroprednisolonate, synthesized by modifying the 17 beta ketol side chain of prednisolone has been investigated in the cotton pellet granuloma bioassay in rats. The inhibitory effect of the ester derivatives on granuloma formation was equivalent to that of the parent compound. At dose of 5 mg/cotton pellet, methyl prednisolonate exhibited a highly significant thymolytic effect comparable to that of prednisolone, while methyl 20-dihydroprednisolonate had a weaker thymolytic effect. However, the local antiinflammatory activity of the two esters was not accompanied by a decrease in adrenal weight observed with the parent compound.

Inhibition of p-aminophenol metabolism: a possible mechanism of enhancement of aniline hydroxylation.

The p-hydroxylation of aniline has been traditionally determined by measurement of p-aminophenol (PAP) formation. Comparison of isotopic and colorimetric procedures indicate that the actual amount of aniline metabolized exceeds the among of PAP recovered. Data suggest that enhancement of p-hydroxylation of aniline by acetone, malaoxon and paraoxon may result from inhibition of further metabolism of PAP by the microsomal cytochrome b5-dependent desaturase system. Involvement of the desaturase system is supported by observations that: (a) metabolism of PAP was reduced by starvation and stimulated when starvation was followed by feeding a high carbohydrate diet; (b) enrichment of hepatic microsomes with detergent purified cytochrome b5 decreased the amount of aniline apparently metabolized, as measured by the amount of PAP recovered, and (c) a high correlation occurred between effects of acetone, malaoxon and paraoxon on reoxidation of cytochrome b5 and capacity of these three compounds to enchance aniline metabolism.

Hazardous wastes, hazardous materials and environmental health inequity.

This paper reviews issues associated with the equity of locating hazardous waste sites and hazardous materials. Reports and case studies indicate that hazardous waste sites and the locations of hazardous materials are disproportionately situated near minority communities, especially African-American communities. This inequitable placement of hazardous waste sites is of concern, since exposure to toxic waste can adversely affect human health. Proximity to these sites may place these minority communities at higher risk of developing cancers and respiratory, cardiovascular, and neurological diseases, and of incurring increased levels of individual and family stress. The health of persons in minority communities near hazardous waste sites is further compromised by their lack of access to adequate health care. The potential health risks borne by racial and ethnic minorities and by low income communities as a consequence of exposure to toxic waste constitutes environmental inequity. In order to decrease the burden of these risks, we recommend developing environmental policies that address environmental inequity; conducting detailed demographic and health studies that assess the impact of exposure to toxic waste on minority populations; and devising educational programs to sensitize professional service providers and prevent exposure by community residents. This paper identifies research needs and opportunities.
